Occurrence of microplastics in gastrointestinal tracts and gills of fish from Beibu Gulf, South China Sea

Microplastics are widespread across the global oceans, yet the potential risks of the ubiquitous environmental contaminant to marine organisms has been less known. Accumulation of microplastics and associated contaminants in marine fish, may pose adverse impacts to human health via seafood consumption. This study evaluated microplastic contamination in 24 fish species collected from Beibu Gulf, one of the world’s largest fishing grounds in South China Sea. Microplastics were detected in 12 fish species at an abundance of 0.027–1.000 items individual⁻¹ and found in fish stomach, intestines and gills with the count percentage of 57.7%, 34.6% and 7.7%, respectively. Transparent fibers were observed as the predominant microplastics, which might be ingested accidently by fish or transferred through other animals at lower trophic levels. Majority of microplastics were identified as polyester (44%) and nylon (38%), whereas polypropylene (6%), polyethylene (6%), and acrylics (6%) were also found. Relatively, higher microplastic abundances were found in demersal fish compared to the pelagic species. Overall, the abundance of microplastics was documented as relatively low in the commercial fish collected from the open water of Beibu Gulf, South China Sea.

The non-steroidal anti-inflammatory drugs (NSAIDs) ibuprofen and diclofenac are highly prescribed worldwide and their presence in aquatic system may pose a potential risk to aquatic organisms. Here, we systematically assessed their cardiovascular disruptive effects in zebrafish (Danio rerio) at environmentally relevant concentrations between 0.04 and 25.0 μg/L. Ibuprofen significantly increased the cardiac outputs of zebrafish embryos at actual concentrations of 0.91, 4.3 and 21.9 μg/L. It up-regulated the blood cell velocity, total blood flow and down-regulated the blood cell density at concentrations of 4.3 μg/L and higher. In comparison, diclofenac led to inhibition of spontaneous muscle contractions and decreased hatching rate of zebrafish embryos at the highest concentration (24.1 μg/L), while it had negligible effects on the cardiac physiology and hemodynamics. Transcriptional analysis of biomarker genes involved in cardiovascular physiology, such as the significantly up-regulated nppa and nkx2.5 expressions response to ibuprofen but not to diclofenac, is consistent with these observations. In addition, both ibuprofen and diclofenac altered the morphology of intersegmental vessels at high concentrations. Our results revealed unexpected cardiovascular functional alterations of NSAIDs to fish at environmental or slightly higher than surface water concentrations and thus provided novel insights into the understanding of their potential environmental risks.

A combined approach integrating bioenergetics and major biological activities is essential to properly understand the impact of microplastics (MP) on marine organisms. Following experimental exposure of polystyrene microbeads (micro-PS of 6 and 10 μm) at 0.25, 2.5, and 25 μg L⁻¹, which demonstrated a dose-dependent decrease of energy balance in the pearl oyster Pinctada margaritifera, a transcriptomic study was conducted on mantle tissue. Transcriptomic data helped us to decipher the molecular mechanisms involved in P. margaritifera responses to micro-PS and search more broadly for effects on energetically expensive maintenance functions. Genes related to the detoxification process were impacted by long-term micro-PS exposure through a decrease in antioxidant response functioning, most likely leading to oxidative stress and damage, especially at higher micro-PS doses. The immune response was also found to be dose-specific, with a stress-related activity stimulated by the lowest dose present after a 2-month exposure period. This stress response was not observed following exposure to higher doses, reflecting an energy-limited capacity of pearl oysters to cope with prolonged stress and a dramatic shift to adjust to pessimum conditions, mostly limited and hampered by a lowered energetic budget. This preliminary experiment lays the foundation for exploring pathways and gene expression in P. margaritifera, and marine mollusks in general, under MP exposure. We also propose a conceptual framework to properly assess realistic MP effects on organisms and population resilience in future investigations.

After the initial outbreak in China, the diffusion in Italy of SARS-CoV-2 is exhibiting a clear regional trend with more elevated frequency and severity of cases in Northern areas. Among multiple factors possibly involved in such geographical differences, a role has been hypothesized for atmospheric pollution. We provide additional evidence on the possible influence of air quality, particularly in terms of chronicity of exposure on the spread viral infection in Italian regions. Actual data on Covid-19 outbreak in Italian provinces and corresponding long-term air quality evaluations, were obtained from Italian and European agencies, elaborated and tested for possible interactions. Our elaborations reveal that, beside concentrations, the chronicity of exposure may influence the anomalous variability of SARS-CoV-2 in Italy. Data on distribution of atmospheric pollutants (NO₂, O₃, PM₂.₅ and PM₁₀) in Italian regions during the last 4 years, days exceeding regulatory limits, and years of the last decade (2010–2019) in which the limits have been exceeded for at least 35 days, highlight that Northern Italy has been constantly exposed to chronic air pollution. Long-term air-quality data significantly correlated with cases of Covid-19 in up to 71 Italian provinces (updated April 27, 2020) providing further evidence that chronic exposure to atmospheric contamination may represent a favourable context for the spread of the virus. Pro-inflammatory responses and high incidence of respiratory and cardiac affections are well known, while the capability of this coronavirus to bind particulate matters remains to be established. Atmospheric and environmental pollution should be considered as part of an integrated approach for sustainable development, human health protection and prevention of epidemic spreads but in a long-term and chronic perspective, since adoption of mitigation actions during a viral outbreak could be of limited utility.

Organic chemical pollutants associated with microplastic (MP) may represent an alternative exposure route for these chemicals to marine biota. However, the bioavailability of MP-sorbed organic pollutants under conditions where co-exposure occurs from the same compounds dissolved in the water phase has rarely been studied experimentally, especially where pollutant concentrations in the two phases are well characterized. Importantly, higher concentrations of organic pollutants on ingested MP may be less bioavailable to aquatic organisms than the same chemicals present in dissolved form in the surrounding water. In the current study, the sorption kinetics of two model polycyclic aromatic hydrocarbons (PAHs; fluoranthene and phenanthrene) to MP particles in natural seawater at 10 and 20 °C were studied and the bioavailability of MP-sorbed PAHs to marine copepods investigated. Polyethylene (PE) and polystyrene (PS) microbeads with mean diameters ranging from 10 to 200 μm were used to identify the role of MP polymer type and size on sorption mechanisms. Additionally, temperature dependence of sorption was investigated. Results indicated that adsorption dominated at lower temperatures and for smaller MP (10 μm), while absorption was the prevailing process for larger MP (100 μm). Monolayer sorption dominated at lower PAH concentrations, while multilayer sorption dominated at higher concentrations. PE particles representing ingestible (10 μm) and non-ingestible (100 μm) MP for the marine copepod species Acartia tonsa and Calanus finmarchicus were used to investigate the availability and toxicity of MP-sorbed PAHs. Studies were conducted under co-exposure conditions where the PAHs were also present in the dissolved phase (Cfᵣₑₑ), thereby representing more environmentally relevant exposure scenarios. Cfᵣₑₑ reduction through MP sorption was reflected in a corresponding reduction of lethality and bioaccumulation, with no difference observed between ingestible and non-ingestible MP. This indicates that only free dissolved PAHs are significantly bioavailable to copepods under co-exposure conditions with MP-sorbed PAHs.

Cyanobacterial blooms and their secondary metabolites, microcystins (MCs), are not only toxic to aquatic organisms, but also to humans. MCs exert reproductive toxicity in female fish by affecting the oocyte development. However, the mechanism behind MC-LR interference in oocyte development remains largely unknown. In our study, adult female zebrafish were exposed to MC-LR (0, 1, 5, 20 μg/L) for 30 d. After exposure to MC-LR for 30 d, fertilized eggs from the treated females and healthy males were collected and cultured in water without MC-LR. Histomorphological observations showed pathological damage in the ovary after MC-LR exposure, which was mainly characterized by enlarged intercellular spaces, detachment of follicular cells from oocytes, and vacuolation of parenchymal tissues. The 20 μg/L MC-LR treatment caused a remarkable increase in the rate of the zebrafish oocytes germinal vesicle breakdown (GVBD) and a significant decrease in the levels of cyclic adenosine monophosphate (cAMP) and vitellogenin (VTG). In addition, the phosphorylation levels of the extracellular signal-regulated kinases (ERK) were elevated in ovaries from zebrafish exposed to 5 and 20 μg/L MC-LR, and cyclinB phosphorylation levels were also upregulated notably in the 20 μg/L MC-LR group. However, MC-LR exposure did not cause any change in the levels of cAMP-dependent protein kinase (PKA) protein and cdc2 phosphorylation in all the treatments. All the doses of MC-LR reduced the number of eggs, prematurely hatched the fertilized eggs and increased the abnormal rate of offspring generation. In summary, the present study demonstrates that MC-LR promotes oocyte maturation by activating the ERK1/2 and MPF signaling pathways, and cAMP is involved in this process.

Problems with PM₂.₅ pollution in the Pearl River Delta (PRD) have been significantly reduced since the Chinese government released a series of emission control policies including the strengthened controls in the 13ᵗʰ Five-Year Plan. This study assessed the efficacy of emission control measures using the Community Multiscale Air Quality (CMAQ) model to provide data-driven support to government decision making, which is becoming increasingly important. This study aimed to quantitatively evaluate the integrated results of proposed policies for controlling PM₂.₅ concentrations. Accordingly, the regional 2015 emission inventory was modified with recently released government data for the PRD, and scenarios for four dynamical emission-reduction policies (S1–S4) were explored. The results show that all four proposed control measures can help to reduce PM₂.₅ concentrations throughout Hong Kong (HK), Macao, and the PRD economic zone (PRD EZ) by 2020. In all cases, reductions in PM₂.₅ concentrations were larger over PRD EZ than over HK. For HK, the predicted annual concentrations of PM₂.₅ were less than 20 μg/m³ for S1–S3 and less than 15 μg/m³ for S4. For Macao, the predicted annual concentrations of PM₂.₅ were less than 25 μg/m³ for S1 and less than 15 μg/m³ up to S3. Regionally, HK had the lowest PM₂.₅ levels, and the area around Foshan had the highest. Controlling the sources of air pollution (i.e., industry, transport, power production, and other sources) within PRD can get most of the PRD EZ region to below 35 μg/m³. Similar national air quality management efforts could reduce PM₂.₅ levels to less than 25 μg/m³ in the PRD EZ and less than 15 μg/m³ in HK. Control measures in S1 led to significant improvement in Shenzhen and HK, but the S3 option brought the greatest improvement for PRD EZ and Macao. The S4 policy option led to substantial reductions, particularly for HK.

Periphyton, composed of algae, bacteria, protozoa, epiphytes, and detritus, is widely distributed on the surfaces of paddy soils. Little is known about the interactions between the periphyton and arsenic (As) in the paddy soil. In the present study, model paddy ecosystems with and without periphyton were set up to explore the effects of periphyton on As migration and transformation in soil. According to the results, periphyton played dual roles in the mobility of As in soil. Periphyton on the surface of paddy soil could significantly increase the mobility and bioavailability of As in soils in the rice tillering stage because of the increased pH and the decreased Eh. The As uptake by rice also increased in the presence of periphyton. However, a significant fraction of the released As was further entrapped by the periphyton, significantly decreasing As concentration in pore water. As biotransformation genes, including aioA, arrA, arsC, and arsM, were identified in periphyton, with arsM being the most abundant in periphyton and soil. Periphyton significantly decreased the abundance of aioA, but increased the abundance of arsC in soils. Cupriavidus and Afipia, which are involved in As(V) cytoplasmic reduction, significantly increased in the presence of periphyton. Periphyton exerted minor effects on the highly abundant and predominant bacteria but had major effects on the less abundant bacteria in the paddy soil. The results of the present study could facilitate the regulation of As contamination in paddy soil, and enhance our understanding of the role of periphyton in the As biogeochemical cycle.

This study investigated the tissue distribution of phenanthrene (PHE) and the formation of monohydroxy-phenanthrene (OH-PHE) metabolites in Korean rockfish (Sebastes schlegelii). PHE was intragastrically administered to two groups of rockfish. The first group was exposed to PHE at a low dose (10 mg/kg body weight) and the second group was exposed at a high dose (30 mg/kg body weight). The rockfish were analyzed and the levels of PHE were higher in the liver, followed by muscle, and then bile. PHE concentrations in the liver, muscle, and bile were 1.4–26, 0.10–2.01, and not detected (ND)–0.13 μg/g wet weight, respectively. All five monohydroxylated PHE metabolites (1-OH-PHE, 2-OH-PHE, 3-OH-PHE, 4-OH-PHE, and 9-OH-PHE) were detected only in bile. Among these OH-PHE metabolites, 3-OH-PHE was found at the highest concentration from all fish bile samples in both PHE exposure groups, indicating that regioselective OH-PHE formation occurs in rockfish and 3-OH PHE could be a good biomarker of exposure of Korean rockfish to PHE. Suspect screening analysis of the rockfish bile was performed by LC-QTOF/MS, and the formation of two OH-PHE-DNA adducts (thymine–OH–PHE and cytosine–OH–PHE) were identified in the bile sample collected 6 h after rockfish were exposed to the high PHE dose, indicating that OH-PHE metabolites may be toxic to fish. This is the first report on the formation characteristics of OH-PHE metabolites in rockfish and their use as biomarkers of exposure of rockfish to parent PHE.

Nanoplastics are inevitably released into aquatic environments due to their extensive use and the continuous fragmentation of plastics. Therefore, it is imperative to understand the aggregation behaviours that determine the transport and fate of nanoplastics in aquatic environments. In this study, the effects of various metal cations, pH, aging and extracellular polymeric substances (EPS) on the aggregation of polystyrene nanoplastics (nano-PS) in aqueous solutions were systematically evaluated based on aggregation kinetics experiments and Derjaguin–Landau–Verwey–Overbeek (DLVO) theoretical calculation. The concentration, valence and hydration ability of metal cations jointly affected the aggregation of nano-PS. The critical coagulation concentration (CCC) of nano-PS was significantly higher than the ionic strengths in aquatic environments, indicating that the aggregation rate of nano-PS is relatively low in aquatic environments. The results of the aggregation kinetics experiments were consistent with DLVO theory, which showed that the energy barrier of nano-PS was dependent on electrostatic repulsion forces and van der Waals forces, and increased with pH. Nano-PS was artificially aged by UV-H₂O₂, which reduced the hydrophobic nature of the particle surfaces, consequently enhancing the stability of the nanoplastics. EPS (excreted from Chlorella pyrenoidosa) decreased the aggregation rates of nano-PS due to steric effects, which was confirmed by the extend DLVO model. Our results highlight the high stability of nano-PS in aquatic environments, which could help facilitate the evaluation of their environmental impact.
