Metabolic transformation as a diagnostic tool for the selection of candidate promutagens in effect-directed analysis

To implement metabolic activation by S9 rat liver homogenate in the selection of candidate promutagens in effect-directed analysis, we critically assessed the capability of LC-HRMS measurements to detect depletion and formation of metabolites by S9 exposure. The exposure of a reference mixture to S9 led to a depletion by >70% for most compounds. Other processes than metabolism were excluded as significant contribution to compound depletion. Metabolites formed by S9 exposure were identified and S9 metabolism was incorporated in the identification of candidate promutagens in a wastewater treatment plant (WWTP) effluent with mutagenic activity only after metabolic activation by S9. The metabolism by S9 in the WWTP effluent was confirmed. Based on a candidate exclusion of all peaks not depleted, thus not activated by the S9 mix, the number of candidate promutagens was reduced by 40%. Selected remaining candidates were evaluated and identified, but could not be confirmed as promutagens.

Cyhalofop-butyl is a selective herbicide widely employed in paddy field, which can transfer into aquatic environments. However, details of the environmental risk and aquatic toxicity of cyhalofop-butyl have not been fully investigated. In this study, zebrafish (Danio rerio) embryos were exposed to a range of cyhalofop-butyl until 120 hour post-fertilization (hpf) to assess embryonic toxicity of the chemical. Our results demonstrated that cyhalofop-butyl was highly toxic to zebrafish embryos, with concentration-dependent negative effects in embryonic development. In addition, exposure to cyhalofop-butyl resulted in significant increases in reactive oxygen species (ROS) production and cell apoptosis in heart area. The mRNA levels of the genes related to oxidative stress and apoptosis were also altered significantly after cyhalofop-butyl exposure. Moreover, the activity of capspase-9 and caspase-3 were significantly increased. Therefore, we speculated that oxidative stress-induced apoptosis should be responsible for abnormal development during embryogenesis after cyhalofop-butyl exposure.

The aim of this study was to investigate the association between exposure to extremely low frequency magnetic fields (ELF MF), or power frequency fields, and non-specific physical symptoms (NSPS). In a cross-sectional study, personal exposure to ELF MF was measured for 99 adults selected in and around Amsterdam, the Netherlands in 2009–2010. They were scored on 16 NSPS. As a cut-off point for the individual 24-h time weighted average exposure the 80-percentile (0.09 μT) was chosen. As only one man scored “moderately high” on the somatisation scale against nine women, we decided to proceed analyses only with the 48 women. The crude odds ratio (OR) for women was 8.50 (CI 95%: 1.73–46.75), suggesting that for women environmental exposure to ELF MF is associated with an increased score on NSPS. As this is an exploratory cross-sectional study in a relatively small sample, no conclusions regarding causality can be drawn.

We investigated whether there might be excess ovarian cancer mortality among women residing near Spanish industries, according to different categories of industrial groups and toxic substances. An ecologic study was designed to examine ovarian cancer mortality at a municipal level (period 1997–2006). Population exposure to pollution was estimated by means of distance from town to facility. Using Poisson regression models, we assessed the relative risk of dying from ovarian cancer in zones around installations, and analyzed the effect of industrial groups and pollutant substances. Excess ovarian cancer mortality was detected in the vicinity of all sectors combined, and, principally, near refineries, fertilizers plants, glass production, paper production, food/beverage sector, waste treatment plants, pharmaceutical industry and ceramic. Insofar as substances were concerned, statistically significant associations were observed for installations releasing metals and polycyclic aromatic chemicals. These results support that residing near industries could be a risk factor for ovarian cancer mortality.

During the 20th century, the local economy of the Upper Loire Basin (ULB) was essentially based on industrial coal mining extraction. One of the major French coal districts with associated urban/industrial activities and numerous coking/gas plants were developed in the Ondaine-Furan subbasins, two tributaries of the upper Loire main stream. To determine the compositional assemblage, the level and the potential sources of contamination, the historical sedimentary chronicle of the 16 US EPA priority polycyclic aromatic hydrocarbons (PAHs) has been investigated. PAH concentrations were determined using gas chromatography/mass spectrometry (GC/MS) in a dated core, sampled in the Villerest flood-control reservoir located downstream of the Ondaine-Furan corridor (OFC). The most contaminated sediments were deposited prior to 1983 (Σ16PAHs ca. 4429–13,348 ng/g) and during flood events (Σ16PAHs ca. 6380 ng/g – 1996 flood; 5360 ng/g – 2003 flood; 6075 ng/g – 2008 flood), especially in medium and high molecular weight PAHs. Among them, typical pyrogenic PAHs such as FLT, PYR, BbF and BaP were prevalent in most of the core samples. In addition, some PAHs last decade data is available from the Loire Bretagne Water Agency and were analyzed using high-performance liquid chromatography with postcolumn fluorescence derivatization (HPLC/FLD). These results confirm that the most highly contaminated sediments were found downstream of OFC (Σ16PAHs ca. 2264–7460 ng/g). According to the observed molecular distribution, PAHs are originated largely from high-temperature pyrolytic processes. Major sources of pyrogenic PAHs have been emphasized by calculation of specific ratios and by comparison to reported data. Atmospheric deposition of urban and industrial areas, wood combustion and degraded coal tar derived from former factories of coking/gas plants seem to be the major pyrogenic sources. Specifically, particular solid transport conditions that can occur during major flood events lead us to emphasize weathering of former contamination sources, such as more preserved coal tar.

Fluorescence excitation-emission matrix (EEM) spectra coupled with parallel factor analysis (PARAFAC) was used to characterize dissolved organic matter (DOM) derived from macrophyte decomposition, and to study its complexation with Cu (II) and Cd (II). Both the protein-like and the humic-like components showed a marked quenching effect by Cu (II). Negligible quenching effects were found for Cd (II) by components 1, 5 and 6. The stability constants and the fraction of the binding fluorophores for humic-like components and Cu (II) can be influenced by macrophyte decomposition of various weight gradients in aquatic plants. Macrophyte decomposition within the scope of the appropriate aquatic phytomass can maximize the stability constant of DOM-metal complexes. A large amount of organic matter was introduced into the aquatic environment by macrophyte decomposition, suggesting that the potential risk of DOM as a carrier of heavy metal contamination in macrophytic lakes should not be ignored.

Most reef building corals rely on symbiotic microalgae (genus Symbiodinium) to supply a substantial proportion of their energy requirements. Functional diversity of different Symbiodinium genotypes, endorsing the host with physiological advantages, has been widely reported. Yet, the influence of genotypic specificity on the symbiont's susceptibility to contaminants or cumulative stressors is unknown. Cultured Symbiodinium of presumed thermal-tolerant clade D tested especially vulnerable to the widespread herbicide diuron, suggesting important free-living populations may be at risk in areas subjected to terrestrial runoff. Co-exposure experiments where cultured Symbiodinium were exposed to diuron over a thermal stress gradient demonstrated how fast-growing clade C1 better maintained photosynthetic capability than clade D. The mixture toxicity model of Independent Action, considering combined thermal stress and herbicide contamination, revealed response additivity for inhibition of photosynthetic yield in both tested cultures, emphasizing the need to account for cumulative stressor impacts in ecological risk assessment and resource management.

As a highly toxic heavy metal, the pollution and exposure risks of lead are of widespread concern for human health. However, the collection of blood samples for use as an indicator of lead pollution is not always feasible in most cohort or longitudinal studies, especially those involving children health. To evaluate the potential use of urinary lead as an indicator of exposure levels and source apportionment, accompanying with environmental media samples, lead concentrations and isotopic measurements (expressed as 207Pb/206Pb, 208Pb/206Pb and 204Pb/206Pb) were investigated and compared between blood and urine from children living in the vicinities of a typical coking plant and lead-acid battery factory. The results showed urinary lead might not be a preferable proxy for estimating blood lead levels. Fortunately, urinary lead isotopic measurements could be used as an alternative for identifying the sources of children's lead exposure, which coincided well with the blood lead isotope ratio analysis.

Low-methylesterified homogalacturonans have been suggested to play a role in the binding and immobilization of Pb in CW. Using root apices of hybrid aspen, a plant with a high phytoremediation potential, as a model, we demonstrated that the in situ distribution pattern of low-methylesterified homogalacturonan, pectin epitope (JIM5-P), reflects the pattern of Pb occurrence. The region which indicated high JIM5-P level corresponded with “Pb accumulation zone”. Moreover, JIM5-P was especially abundant in cell junctions, CWs lining the intercellular spaces and the corners of intercellular spaces indicating the highest accumulation of Pb. Furthermore, JIM5-P and Pb commonly co-localized.The observations indicate that low-methylesterified homogalacturonan is the CW polymer that determines the capacity of CW for Pb sequestration. Our results suggest a promising directions for CW modification for enhancing the efficiency of plant roots in Pb accumulation, an important aspect in the phytoremediation of soils contaminated with trace metals.

Rapid monitoring and discriminating different anthropogenic pollution is a key scientific issue. To detect the applicability and sensitivity of magnetic measurements for evaluating different industrial pollution in urban environment, characteristics of topsoil from three typical fast developing industrial cities (Jinchang, Baiyin and Jiayuguan in Gansu province, northwestern China) were studied by magnetic and geochemical analyses. The results showed that magnetic susceptibility was enhanced near industrial areas, and PSD-MD magnetite dominated the magnetic properties. Magnetic concentration parameters (χlf, SIRM, and χARM) showed different correlations with heavy metals and PLI in the three cities, indicating significantly different magnetic response to different pollution sources. Principal component analysis showed that ferrimagnetic minerals coexist with heavy metals of Fe, As, Cu, Pb, and Zn in Baiyin and Fe, V, Cu, Mn, Pb, and Cr in Jiayuguan. Fuzzy cluster analysis and regression analysis further indicated that the sensitivity of magnetic monitoring to fuel dust is higher than that to mineral dust near non-ferrous metal smelters, and fossil fuel consumption is an important factor for increasing magnetite content. In all the three cities, the sensitivity of magnetic monitoring to pollutants from steel plants is much higher than that from non-ferrous metal plants. Therefore, magnetic proxies provide a rapid means for detecting heavy metal contamination caused by multi-anthropogenic pollution sources in a large scale area, however, the sensitivity was controlled by pollution sources.
