In-utero exposure to HT-2 toxin affects meiotic progression and early oogenesis in foetal oocytes by increasing oxidative stress 全文

HT-2 toxin (HT-2), a mycotoxin produced by Fusarium species, is detected in a variety of cereal grain-based human food and animal feed. Apart from its well-established immunotoxicity and haematotoxicity, it also causes reproductive disorders. In the present study, we revealed the adverse effects of HT-2 on early oogenesis at the foetal stage. Pregnant mice were orally administered with HT-2 for 3 days at mid-gestation. Oocytes from female foetuses exposed to HT-2 displayed defects in meiotic prophase, including unrepaired DNA damage, elevated recombination levels, and reduced expression of meiotic-related genes. Subsequently, increased oxidative stress was observed in the foetal ovaries exposed to HT-2, along with the elevated levels of reactive oxygen species, malondialdehyde, catalase, and superoxide dismutase 1/2, thereby resulting in impaired mitochondrial membrane potential and cell apoptosis. Furthermore, pre-treatment with urolithin A, a natural compound with antioxidant activities, partially reversed the delayed meiotic process by alleviating oxidative stress. Since early oogenesis is essential to determine female fertility in adult life, this study indicated that brief maternal exposure to HT-2 toxin may compromise the fertility of a developing female foetus.

PM₂.₅ has a major impact on the gastrointestinal system, but the specific mechanism behind this action is not fully understood. Current studies have focused on the relationship between PM₂.₅ and intestinal flora disorder, while ignoring the important influence of diet on gut microbes. In this study, SD rats were fed either a normal, high-fat, or high-carbohydrate diet for two months and exposed to PM₂.₅ (7 mg/kg b.w.) by intratracheal instillation. The results showed that the body and kidney weights of the rats in the high-fat diet group were significantly increased relative to those with a normal diet, and changes in the intestinal microbes and metabolites induced by PM₂.₅ were observed. Rats in the high-carbohydrate diet group had a significant response, and the diversity and richness indices of the flora were reduced (p < 0.05); additionally, intestinal Biffidobacterium and Lactobacillus were enriched, while many endogenous metabolites were found. Some amino acids derivatives and long-chain fatty acids were increased (p < 0.05). Both diet structure and PM₂.₅ exposure can affect the composition of gut microbiota, and intestinal metabolites may be associated with cell membrane damage when a high-carbohydrate diet interacts with PM₂.₅. This study considers multiple dietary factors to further supplement the evidence of intestinal damage via PM₂.₅.

Polystyrene nanoparticles (PS NPs), originated from breakdown of large plastic wastes, have already caused much concern for their environmental risks on health. This current study was aimed to reveal the toxicological mechanism of PS NPs on developing zebrafish and macrophage cells. To fulfill this purpose, 42 nm PS NPs were exposed to the early development stage of zebrafish for 5 days, the decreased heart rate and locomotor activity of zebrafish larvae were observed. The fluorescent PS NPs were used to precisely assess the accumulation of PS NPs in zebrafish larvae, and the results indicated that PS NPs not only accumulated in digestive system, but also infiltrated into the liver. More importantly, the transcriptomic analysis revealed that a total of 356 genes were differentially expressed and the KEGG class map showed significant differences in the MAPK pathway upon PS NPs treatment. Meanwhile, the induction of oxidative stress and inflammation were also observed in zebrafish larvae. Furthermore, PS NPs also induced oxidative damage and inflammatory response in RAW 264.7 cells, which activated p38 MAPK signal pathway and finally induced cell apoptosis. Our study provides a new understanding of MAPK signaling pathway involved in toxicity mechanism.

Agricultural plastic greenhouse (PG) production can extend the growing season of crops to satisfy domestic consumption in countries such as China. Workers in PGs have potential higher phthalate exposure risks than the general population as phthalate accumulation has been observed in greenhouse soil, air, and crops. To date, biomonitoring tests of phthalates for the working population have not been carried out. To address this shortage, we conducted a pilot study in Shaanxi Province, China, among 35 healthy PG workers by follow-up recording their seasonal dietary habits and work activities and urine sample collection and measurement between 2018 and 2019. The objectives were to uncover the association between phthalate metabolites and the population characteristics, seasonal and diurnal variations and causes, and to estimate exposure risks and contributions of exposure pathways from PG production systems. A total of 13 phthalate metabolite concentrations (Σ₁₃ phthalate metabolites) ranged from 102 to 781 (5th-95th) ng/mL (median: 300 ng/mL). Mono-n-butyl phthalate (MNBP) made up 51.3% of Σ₁₃ phthalate metabolites, followed by the sum of four di-2-ethylhexyl phthalate (DEHP) metabolites (24.2%), mono-2-isobutyl phthalate (MIBP) (13.4%), and mono-ethyl phthalate (MEP) (9.8%). The concentrations of MNBP and MIBP in summer were significantly higher than the levels in winter (p < 0.0001). A total of 62.3% of the PG worker population was shown to have exposure risks, and the proportion was as high as 79.4% in summer. Phthalate exposure of the workers from PG production systems constituted over 20% of the total creatinine-based daily intake, and consuming vegetables and fruit planted in PGs and inhalation in PGs were the two largest exposure pathways. Our findings demonstrate that it is important to protect workers in PGs from phthalate exposure risks, and phasing out the use of plastic materials containing phthalates in PGs is imperative, to guarantee food safety in PGs.

Almaden, Calero, and Guadalupe reservoirs (San Jose, CA, USA) are small (<13 million m³) surface water reservoirs polluted by the former New Almaden Mining District, North America’s most productive historical mercury (Hg) mine. Stevens Creek Reservoir (Cupertino, CA, USA) also has elevated fish Hg concentrations, but no historical mining source. We report a 15-year dataset to evaluate the effectiveness of line diffuser hypolimnetic oxygenation systems (HOSs) in reducing methylmercury (MeHg) concentrations in reservoir water and fish after four consecutive years of operation. HOSs were installed in each reservoir to increase dissolved oxygen concentrations in bottom water, thereby suppressing the activity of anaerobic bacteria (e.g., sulfate-reducing bacteria) known to produce MeHg. Before HOS operation, MeHg concentrations increased in bottom waters of all four reservoirs during periods of thermal stratification and profundal hypoxia. MeHg concentrations decreased significantly in bottom waters during HOS operation, with mean reductions of 63%–85% below pre-oxygenation concentrations. However, MeHg concentrations were unchanged or increased in surface waters. This could be the result of enhanced mixing between surface and bottom waters as a result of line diffuser oxygenation, or continued Hg methylation occurring in the oxic water column and littoral sediments. Despite little change in whole water column MeHg concentrations, we observed modest but significant declining trends in fish tissue Hg in Guadalupe and Stevens Creek reservoirs. Results suggest that oxygenation, rather than directly lowering MeHg in water, may have mixed nutrients into surface waters, thereby enhancing primary productivity and indirectly affecting Hg bioaccumulation by diluting concentrations in phytoplankton.

Studies of algal bloom early warning systems have rarely paid attention to the dynamics of excessive proliferation of phytoplankton (EPP), which occurs prior to algal blooms, or to the sensitivity of a lake to EPP based on multiple environmental factors. In this study, we investigated EPP dynamics in large lakes and identified major factors that influenced the lake's vulnerability to EPP, to improve algal bloom early warning systems. High temporal moderate resolution imaging spectroradiometer (MODIS) images and multi-source daily site monitoring data of large lakes in the middle–lower Yangtze River basin were analyzed. Then, the floating algal index (FAI) and resource use efficiency (RUE) by phytoplankton were used to investigate the EPP dynamics and lake's vulnerability to EPP, respectively. Moreover, generalized linear models were used to assess the relative importance of environmental factors on RUE. The results indicate that the lakes freely connected (FC) to the Yangtze River (Dongting Lake and Poyang Lake) had lower FAIs but higher RUEs than the non-connected lakes (NC; Chaohu Lake and Taihu Lake). The key factors affecting RUE-FC were standard deviation of water level within 30 days(WL30), particulate matter <10 μm(PM₁₀), and relative humidity(Hum), which explained 15.91% of the variations in RUE. The key factors affecting RUE-NC were ozone(O₃), basin normalized difference vegetation index standard deviation(BNDVISD), and dissolved oxygen(DO), which explained 35.28% of the variations in RUE. These results emphasize the importance of air quality in influencing or reflecting EPP risks in large lakes. In addition, basin vegetation and hydrological rhythms can influence NH₄⁺ through non-point source loading. Algal bloom early warning systems can be improved by routine monitoring and forecasting of potential environmental factors such as air quality and basin vegetation.

This study focused on the seasonal variation and source identification of heavy metals (HMs) while considering effects of municipal wastewater (MWW) in peri-urban farms of Hue city, central Vietnam. Moreover, associated non-carcinogenic and carcinogenic health risks from consuming vegetables containing HMs were also assessed considering the hazard quotient and cancer risk, respectively. Therefore, concentrations of Fe, Mn, Zn, Cu, Cr, Cd, Pb, and As were determined in irrigation water, soil, and lettuce samples collected during dry and wet seasons from one upstream site where irrigation water has no impact on MWW as well as from two downstream sites in farms on the outskirt of the city. Although irrigation water and soil in the same farms were not polluted as strongly, lettuce samples were polluted with Cd, Zn, and Pb. Furthermore, levels of soil Cu and As and HMs (except for Cu) in lettuce in the wet season were significantly higher (p < 0.05) than those in the dry season, indicating the impact of MWW with seasonal change. The health risk assessment via lettuce consumption demonstrated an unacceptable carcinogenic risk owing to Cd and a cumulative non-carcinogenic risk owing to selected HMs in the lettuce, while all other risks were negligible. Correlation and principal component analyses were performed to identify HM sources, indicating that Cu, Zn, Cd, Pb, Cr, and As in irrigation water and soil could have anthropogenic sources (e.g., untreated MWW, fertilizer use); meanwhile, irrigation-water and soil Fe, Mn, As, and Cr could originate from non-anthropogenic sources (e.g., parent materials weathering). This study revealed that rapid urbanization together with high precipitation leading to urban floods in Hue city was a significant factor spreading HMs in agricultural farms, suggesting the importance of wastewater treatment system, which can reduce the HM load in the city to protect the local food production.

Characteristics and sources of ambient particle elements in urban Beijing were studied by hourly observations in two size fractions (PM₁₀ and PM₂.₅) during November and December 2017 using an online multi-element analyzer. The reconstructed oxide concentrations of 24 elements (from Al to Pb) comprise an appreciable fraction of PM₁₀ and PM₂.₅, accounting for 37% and 17%, respectively on average. We demonstrate the benefit of using high-time-resolution chemical speciation data in achieving robust source apportionment of the total elemental PM₁₀ (PM₁₀ₑₗ) and elemental PM₂.₅ (PM₂.₅ₑₗ) mass using positive matrix factorization (PMF). Biomass burning, coal combustion, secondary sulfate, industry, non-exhaust traffic and dust were identified in both size fractions (with varying relative concentrations), which accounted on average for 4%, 12%, 5%, 2%, 14%, and 63%, respectively to the total PM₁₀ₑₗ, and 14%, 35%, 21%, 6%, 12% and 12%, respectively to the total PM₂.₅ₑₗ. Biomass burning and coal combustion exhibited higher concentrations during haze episodes of the heating season. In contrast, secondary sulfate and industry contributed more to haze episodes during the non-heating season. The fractional contribution of dust was mostly high during clean days, while the fractional non-exhaust traffic emission contribution was similar throughout the measurement period. The non-exhaust traffic emissions contributed locally, while the remaining sources were dominated by neighboring areas. Furthermore, trajectory analysis showed that the origin of the industrial sources roughly agreed with the locations of the main point sources. Overall, this work provides detailed information on the characteristics of the elements during different haze events during heating and non-heating seasons.

North-Eastern Brazil saw intensive application of the insecticide pyriproxyfen (PPF) during the microcephaly outbreak caused by the Zika virus (ZIKV). ZIKV requires the neural RNA-binding protein Musashi-1 to replicate. Thyroid hormone (TH) represses MSI1. PPF is a suspected TH disruptor. We hypothesized that co-exposure to the main metabolite of PPF, 4′–OH–PPF, could exacerbate ZIKV effects through increased MSI1 expression. Exposing an in vivo reporter model, Xenopus laevis, to 4′–OH–PPF decreased TH signaling and increased msi1 mRNA and protein, confirming TH-antagonistic properties. Next, we investigated the metabolite's effects on mouse subventricular zone-derived neural stem cells (NSCs). Exposure to 4′–OH–PPF dose-dependently reduced neuroprogenitor proliferation and dysregulated genes implicated in neurogliogenesis. The highest dose induced Msi1 mRNA and protein, increasing cell apoptosis and the ratio of neurons to glial cells. Given these effects of the metabolite alone, we considered if combined infection with ZIKV worsened neurogenic events. Only at the fourth and last day of incubation did co-exposure of 4′–OH–PPF and ZIKV decrease viral replication, but viral RNA copies stayed within the same order of magnitude. Intracellular RNA content of NSCs was decreased in the combined presence of 4′–OH–PPF and ZIKV, suggesting a synergistic block of transcriptional machinery. Seven out of 12 tested key genes in TH signaling and neuroglial commitment were dysregulated by co-exposure, of which four were unaltered when exposed to 4′–OH–PPF alone. We conclude that 4′–OH–PPF is an active TH-antagonist, altering NSC processes known to underlie correct cortical development. A combination of the TH-disrupting metabolite and ZIKV could aggravate the microcephaly phenotype.

Riverine nitrogen loading to the continental shelf sea is important for terrestrial–marine linkage and global nitrogen cycling and leads to serious marine environmental problems. The budget and cycle of riverine nitrogen over the continental shelf in the East China Sea (ECS) are unknown. Using the tracking technique within a physical–biological coupled model, we quantified the nitrogen budgets of riverine dissolved inorganic nitrogen (DIN) and particulate organic nitrogen (PON) over seasonal to annual scales in the ECS, especially from the Changjiang River, which plays a dominant role in riverine nitrogen input. The horizontal distributions of the Changjiang DIN and PON generally followed the Changjiang diluted water and coastal currents and were affected by stratification in the vertical direction. Their inventory variations were dominated by biological fluxes and modulated by physical ones, and changed most dramatically in the inner shelf among three subregions. Less than half of DIN were converted to PON with most of the rest leaving the ECS through lateral transport pathways, among which the flux through the Tsushima Strait was dominant. With the increasing loading of the Changjiang DIN flux from the 1980s–2010s, lateral transports rather than PON production increased due to limited primary production. Approximately 60 % of the produced PON exported to the sediment and 34 % went to the Tsushima Strait. According to the export production, the DIN from the Changjiang River contributed 12–42 % to the ECS carbon sequestration.
