Using devitalized moss for active biomonitoring of water pollution

This paper presents the results of an experiment carried out for the first time in situ to select a treatment to devitalize mosses for use in active biomonitoring of water pollution. Three devitalizing treatments for the aquatic moss Fontinalis antipyretica were tested (i.e. oven-drying at 100 °C, oven-drying with a 50-80-100 °C temperature ramp, and boiling in water), and the effects of these on loss of material during exposure of the transplants and on the accumulation of different heavy metals and metalloids were determined. The suitability of using devitalized samples of the terrestrial moss Sphagnum denticulatum to biomonitor aquatic environments was also tested. The structure of mosses was altered in different ways by the devitalizing treatments. Devitalization by boiling water led to significantly less loss of material (p < 0.01) than the oven-drying treatments. However, devitalization by oven-drying with a temperature ramp yielded more stable results in relation to both loss of material and accumulation of elements. With the aim of standardizing the moss bag technique, the use of F. antipyretica devitalized by oven-drying with a temperature ramp is recommended, rather than other devitalization treatments or use of S. denticulatum.

Influences of ozone (O3) on light-saturated rates of photosynthesis in crop leaves have been well documented. To increase our understanding of O3 effects on individual- or stand level productivity, a mechanistic understanding of factors determining canopy photosynthesis is necessary. We used a canopy model to scale photosynthesis from leaf to canopy, and analyzed the importance of canopy structural and leaf ecophysiological characteristics in determining canopy photosynthesis in soybean stands exposed to 9 concentrations of [O3] (37–116 ppb; 9-h mean). Light intensity and N content peaked in upper canopy layers, and sharply decreased through the lower canopy. Plant leaf area decreased with increasing [O3] allowing for greater light intensity to reach lower canopy levels. At the leaf level, light-saturated photosynthesis decreased and dark respiration increased with increasing [O3]. These data were used to calculate daily net canopy photosynthesis (Pc). Pc decreased with increasing [O3] with an average decrease of 10% for an increase in [O3] of 10 ppb, and which was similar to changes in above-ground dry mass production of the stands. Absolute daily net photosynthesis of lower layers was very low and thus the decrease in photosynthesis in the lower canopy caused by elevated [O3] had only minor significance for total canopy photosynthesis. Sensitivity analyses revealed that the decrease in Pc was associated with changes in leaf ecophysiology but not with decrease in leaf area. The soybean stands were very crowded, the leaves were highly mutually shaded, and sufficient light for positive carbon balance did not penetrate to lower canopy leaves, even under elevated [O3].

In this study, the interactions between microplastics, chosen among the most widely used in industry such as polypropylene (PP) and high-density polyethylene (HDPE), and a model freshwater microalgae, Chlamydomas reinhardtii, were investigated. It was shown that the presence of high concentrations of microplastics with size >400 μm did not directly impact the growth of microalgae in the first days of contact and that the expression of three genes involved in the stress response was not modified after 78 days. In parallel, a similar colonization was observed for the two polymers. However, after 20 days of contact, in the case of PP only, hetero-aggregates constituted of microalgae, microplastics and exopolysaccharides were formed. An estimation of the hetero-aggregates composition was approximately 50% of PP fragments and 50% of microalgae, which led to a final density close to 1.2. Such hetero-aggregates appear as an important pathway for the vertical transport of PP microplastics from the water surface to sediment. Moreover, after more than 70 days of contact with microplastics, the microalgae genes involved in the sugar biosynthesis pathways were strongly over-expressed compared to control conditions. The levels of over-expression were higher in the case of HDPE than in PP condition. This work presents the first evidence that depending on their chemical nature, microplastics will follow different fates in the environment.

Plastic pollution is a growing global concern. In the present study, we investigated plastic pollution in 21 species of sea fish and 6 species of freshwater fish from China. All of the species were found to ingest micro- or mesoplastics. The average abundance of microplastics varied from 1.1 to 7.2 items by individual and 0.2–17.2 items by gram. The average abundance of mesoplastics varied from 0.2 to 3.0 items by individual and 0.1–3.9 items by gram. Microplastics were abundant in 26 species, accounting for 55.9–92.3% of the total number of plastics items in each species. Thamnaconus septentrionalis contained the highest abundance of microplastics (7.2 items/individual). The average abundance of plastics in sea benthopelagic fishes was significantly higher than in freshwater benthopelagic fishes by items/individual. The plastics were dominanted by fiber in shape, transparent in color and cellophane in composition. The proportion of plastics in the stomach to the intestines showed great variation in different species, ranging from 0.5 to 1.9 by items/individual. The stomach of Harpodon nehereus and intestines of Pampus cinereus contained the highest number of plastics, (3.3) and (2.7), respectively, by items/individual. Our results suggested that plastic pollution was widespread in the investigated fish species and showed higher abundance in comparison with worldwide studies. The ingestion of plastics in fish was closely related to the habitat and gastrointestinal tract structure. We highly recommend that the entire gastrointestinal tract and digestion process be used in future investigations of plastic pollution in fish.

The antibiotic resistance genes (ARGs) from urban waste may spread to the environment with the discharge of leachate. Fifteen types of ARGs, including tetracycline, sulfonamides, AmpC β-lactamase and the class 1 integron gene were detected in the samples from the largest leachate treatment plant (LTP) in Guangzhou and its effluent receiving bodies (soil and surface water). The results showed that ARGs in leachates were in high levels and varied with seasons. The abundance of ARGs in the influent from high to low was in the turn of summer, winter, spring. About 2 to 4 orders of magnitude of ARGs were eliminated by the whole leachate treatment process. The predominant ARGs in the receiving soil were intI1, tetB, sul2, tetA and tetX, while those in the receiving surface water were sul2, intI1 and sul1, and the concentrations of ARGs in the receiving bodies were higher than those in the other natural bodies by 1 to 2 orders of magnitude. In addition, the results of bivariate correlation analysis showed that the abundances of ARGs (tetC, tetW, sul1, sul2, intI1 and FOX) were in significant correlation with the concentrations of heavy metals (Cu, Zn, Ni and Cr) (p < 0.05). LTPs are more likely to be sources of ARGs than wastewater treatment plant (WWTP) and need to be focused on.

In recent years measurable concentrations of non-steroidal anti-inflammatory drugs (NSAIDs) have been shown in the aquatic environment as a result of increasing human consumption. Effects of five frequently used non-steroidal anti-inflammatory drugs (diclofenac, diflunisal, ibuprofen, mefenamic acid and piroxicam in 0.1 mg ml−1 concentration) in batch cultures of cyanobacteria (Synechococcus elongatus, Microcystis aeruginosa, Cylindrospermopsis raciborskii), and eukaryotic algae (Desmodesmus communis, Haematococcus pluvialis, Cryptomonas ovata) were studied. Furthermore, the effects of the same concentrations of NSAIDs were investigated in natural algal assemblages in microcosms. According to the changes of chlorophyll-a content, unicellular cyanobacteria seemed to be more tolerant to NSAIDs than eukaryotic algae in laboratory experiments. Growth of eukaryotic algae was reduced by all drugs, the cryptomonad C. ovata was the most sensitive to NSAIDs, while the flagellated green alga H. pluvialis was more sensitive than the non-motile green alga D. communis. NSAID treatments had weaker impact in the natural assemblages dominated by cyanobacteria than in the ones dominated by eukaryotic algae, confirming the results of laboratory experiments. Diversity and number of functional groups did not change notably in cyanobacteria dominated assemblages, while they decreased significantly in eukaryotic algae dominated ones compared to controls. The results highlight that cyanobacteria (especially unicellular ones) are less sensitive to the studied, mostly hardly degradable NSAIDs, which suggest that their accumulation in water bodies may contribute to the expansion of cyanobacterial mass productions in appropriate environmental circumstances by pushing back eukaryotic algae. Thus, these contaminants require special attention during wastewater treatment and monitoring of surface waters.

Antimicrobial resistance is a growing and significant threat to global public health, requiring better understanding of the sources and mechanisms involved in its emergence and spread. We investigated the abundance of antibiotic resistance genes (ARGs) before and after treatment in five wastewater treatment plants (WWTPs) located in different areas of the Monastir Governorate (Tunisia). Three of these WWTPs (Frina, Sahline and Zaouiet) use a conventional activated sludge process as secondary treatment, whereas the WWTP located in Beni Hassen applies an ultraviolet disinfection step after the activated sludge process and the WWTP located in Moknine treats wastewater using naturally aerated lagoons as a secondary treatment process. The abundance of six ARGs (blaCTX-M, blaTEM, qnrA, qnrS, sul I and ermB) and the class 1 integron-integrase gene (intI1) were determined by quantitative PCR. All ARGs and the intI1 gene were detected in the wastewater samples, except the blaCTX-M gene, which was not detected in both influent and effluent samples from Sahline and Beni Hassen WWTPs, and the qnrS gene, which was not detected neither in the WWTP influent in Moknine nor in the WWTP effluent in Beni Hassen. Although the relative concentration of ARGs was generally found to be similar between samples collected before and after the wastewater treatment, the abundance of blaCTX-M, blaTEM, and qnrS genes was higher in the effluent of the Frina WWTP which, unlike other WWTPs, not only receives domestic or industrial sewage but also untreated hospital waste. To the best of our knowledge, this study quantified for the first time the abundance of ARGs in different Tunisian WWTPs, and the results agree with previous studies suggesting that conventional wastewater treatment does not efficiently reduce ARGs. Therefore, these findings could be useful to improve the design or operation of WWTPs.

While the potential of plants to uptake polycyclic aromatic hydrocarbons (PAHs) is widely acknowledged, empirical evidence of the effects of this process on local atmospheric PAH concentrations and human health is tenuous. We measured gaseous PAH concentrations using passive samplers in urban tree-covered areas and adjacent open, treeless areas in a near-road environment in Finland to gain information on the ability of urban vegetation to improve air quality. The ability of urban, mostly deciduous, vegetation to affect PAHs was season dependent: during summer, concentrations were significantly higher in tree-covered areas, while in the fall, concentrations in open areas exceeded those in tree-covered areas. During winter, concentrations in tree-covered areas were either lower or did not differ from those in open areas. Results of this study imply that the commonly believed notion that trees unequivocally improve air quality does not apply to PAHs studied here.

Accumulations of heavy metals in urban soils are highly spatial heterogeneity and affected by multiple factors including soil properties, land use and pattern, population and climatic conditions. We studied accumulation risks of Cd, Cu, Pb and Zn in unban soils of Beijing and their influencing based on the regression tree analysis and a GIS-based overlay model. Result shows that Zinc causes the most extensive soil pollution and Cu result in the most acute soil pollution. The soil's organic carbon content and CEC and population growth are the most significant factors affecting heavy metal accumulation. Other influence factors in land use pattern, urban landscape, and wind speed also contributed, but less pronounced. The soils in areas with higher degree of urbanization and surrounded by intense vehicular traffics have higher accumulation risk of Cd, Cu, Pb, and Zn.

Short-chain chlorinated paraffins (SCCPs) are ubiquitous in the environment and might cause adverse environmental and human health effects. Little is known about the relative toxicity of different SCCP compounds especially during development. The objective of this study was to characterize and compare effects of seven SCCP groups at environmentally relevant levels, using a zebrafish (Danio rerio) model. Observations on malformation, survival rates at 96 h post fertilization (hpf), and hatching rates at 72 hpf indicated that the C10- groups (C10H18Cl4, 1,2,5,6,9,10-C10H16Cl6 and C10H15Cl7) were more toxic than the C12- groups (C12H22Cl4, C12H19Cl7 and 1,1,1,3,10,12,12,12-C12H18Cl8) and Cereclor 63L. The C10- groups were also more potent than C12- groups and Cereclor 63L in decreasing thyroid hormone levels. Among the three compounds within the C10- group, the compounds with less chlorine content had stronger effects on sub-lethal malformations but less effects on triiodothyronine (T3) and tetraiodothyronine (T4). Only C10H18Cl4 significantly decreased the mRNA expression of tyr, ttr, dio2 and dio3 at a dose-dependent manner suggesting that the specific mode of actions differ with different congeners. The mechanisms of disruption of thyroid status by different SCCPs could be different. C10H18Cl4 might inhibit T3 production through the inhibition effect on dio2. These results indicate that SCCP exposure could alter gene expression in the hypothalamic-pituitary-thyroid (HPT) axis and thyroid hormone levels. The mechanisms of disruption of thyroid status by different SCCPs could be different. Our results on the relative developmental toxicities of SCCPs will be useful to reach a better understanding of SCCP toxicity supporting environmental risk evaluation and regulation and used as a guidance for environmental monitoring of SCCPs in the future.
