In the last decades, the very fast improvement of the analytical instrumentation has led to the possibility of quickly and easily getting a lot of data; in turn, the need of advanced statistical methods suitable to extract the full information furnished by instruments has increased. Such kind of data treatments is particularly important in any case of continuous monitoring of one or more parameters, so the microclimate monitoring is a typical example for this application. Microclimate control is essential in the conservation of Cultural Heritage (CH), but decisions on optimal conservation parameters cannot base only on existing norms that do not take into account the environment’s history. Often CH has survived for many centuries in conditions that must be considered risky but also a stable state (equilibrium) resulting from a long adaptation process during which a more or less heavy damage occurred to the materials. Any successive change of microclimate parameters has interrupted this equilibrium conditions and has induced further damage to material until a new equilibrium is reached; dimension and frequency of changes are proportional to the expected damage. This thermodynamic consideration provides the background for a CH conservation project based on microclimate control and highlights the importance of environmental monitoring for the identification of equilibrium parameters to be maintained. In 2010, we monitored the microclimate of an important historical building in Rome, the Mamertino Carcer, before its opening to visitors. One year later, we repeated the monitoring in the presence of visitors, and here, we present a careful choice of multivariate data treatments adopted for an enough, simple and immediate evaluation of the microclimatic changes; this allows an easier understanding also for persons with not too deep scientific background, such as Superintendents and, in turn, really useful information to provide suggestions for a conservation project. Results evidenced the expected loss of isolation of the site that occurred by opening to visitors; this led to wider excursions of both temperature and relative humidity and, in turn, to a worsening of the conservative conditions. Surely, a monitoring of particulate matter, correlated to air fluxes and, in turn, to microclimate, is of fundamental importance for the conservation of frescoes and will be object of one of our future diagnostic interventions in the site.

Many anthropogenic pollutants in coastal marine environments can induce immune impairments in wild fish and reduce their survival fitness. There is a pressing need to establish sensitive and high throughput in vivo tools to systematically evaluate the immunosuppressive effects of contaminants in marine teleosts. This study reviewed a battery of in vivo immune function detection technologies established for different biological hierarchies at molecular (immune function pathways and genes by next generation sequencing (NGS)), cellular (leukocytes profiles by flow cytometry), tissues/organ system (whole adult histo-array), and organism (host resistance assays (HRAs)) levels, to assess the immune competence of marine medaka Oryzias melastigma. This approach enables a holistic assessment of fish immune competence under different chemical exposure or environmental scenarios. The data obtained will also be useful to unravel the underlying immunotoxic mechanisms. Intriguingly, NGS analysis of hepatic immune gene expression profiles (male > female) are in support of the bacterial HRA findings, in which infection-induced mortality was consistently higher in females than in males. As such, reproductive stages and gender-specific responses must be taken into consideration when assessing the risk of immunotoxicants in the aquatic environment. The distinct phenotypic sexual dimorphism and short generation time (3 months) of marine medaka offer additional advantages for sex-related immunotoxicological investigation.

Brassinosteroids are well known to mitigate biotic stresses; however, their role to induce tolerance against Verticillium dahliae is unknown. The current study employed V. dahliae (Vd) toxin as pathogen-free model system to induce stress on cotton callus growth, and its amelioration was investigated using 24-epibrassinolide (EBR). Results revealed that EBR has ameliorative effects against Vd toxin with greater seen effect when callus was treated with EBR prior to its exposure to Vd toxin (pre-EBR treatment) than EBR applied along with Vd toxin simultaneously (co-EBR treatment). Pre-EBR-treated calli remained green, while 65 and 90% callus browning was observed in co-EBR- and Vd toxin-alone-treated callus, respectively. Likewise, the fresh weight of the pre-EBR-treated callus was 52% higher than Vd toxin-alone treatment, whereas this increase was only 23% in co-EBR-treated callus. Meanwhile, EBR treatment of the cotton callus has also increased the contents of chlorophylls a and b, carotenoids, total phenols, flavonoids, soluble sugars, and proteins and increased the activity of enzymes involved in secondary metabolism like polyphenol oxidase (PPO), phenylalanine ammonialyase (PAL), cinnamyl alchol dehydrogenase (CAD), and shikimate dehydrogenase (SKDH) over Vd toxin-alone treatment with higher increments being observed in pre-EBR-treated callus. Furthermore, EBR treatment mimicked the DNA damage and improved the structure of mitochondria, granum, stroma thylakoids, and the attachment of ribosomes with the endoplasmic reticulum. This EBR-mediated mitigation was primarily associated with substantially increased contents of photosynthetic pigments and regulation of secondary metabolism.

High costs of conventional protein feed sources including soybean meal (SBM) generated the need for finding other alternatives. Thus, the present study was designed to evaluate the impact of graded replacements of SBM by sunflower seed meal (SFM) with or without enzyme supplementation on growth performance, digestive enzymes, carcass traits, and blood profile of broiler chickens. A total of 240 unsexed 1-week-old broiler chicks (Hubbard) were randomly divided into eight treatment groups of 30 chicks each in five replicates each of six chicks in a factorial design (4 × 2) arrangement, including four levels of SFM (0, 25, 50, and 75% replacing SBM) and two levels of enzyme (0- or 0.1-g/kg diet) supplementation. Performance traits including feed conversion ratio, body weight, and weight gain were significantly (P < 0.01) improved with increasing SFM up to 50% substitution for SBM or with enzyme supplementation in broiler diet during the experiment. However, feed intake of broiler chicks was decreased with enzyme supplementation (P < 0.05). The activities of digestive enzymes (protease and amylase) were significantly (P < 0.05) influenced and enhanced by SFM and enzyme inclusion in diets, respectively. The activities of protease and amylase were improved with SFM diet supplemented with 0.1 g/kg enzyme in comparison with those with the un-supplemented diet. The evaluated carcass traits were not statistically (P > 0.05) influenced by feeding SFM meal or enzyme addition. Biochemical blood parameters were significantly (P < 0.01) affected by SFM, enzyme, or their interaction in broiler diets, except for globulin that was not affected by dietary enzyme. It is concluded that increasing SFM level in the diet up to 50% replacing SBM with the supplementation of enzyme improved the growth performance and enhanced positively carcass traits as well as the activity of digestive enzymes in broiler chickens.

Dendrimer-functionalized multi-walled carbon nanotubes (MWCNT) for heavy metal ion removal from wastewaters were developed. Triazole dendrimers (TD) were built directly onto the carbon nanotube surface by successive click chemistry reactions affording the zero- and first-generation dendrimer-functionalized MWCNT (MWCNT-TD1 and MWCNT-TD2). The Moedritzer-Irani reaction carried out on the amino groups present on the MWCNT-TD2 sample gave the corresponding α-aminophosphonate nanosystem MWCNT-TD2P. Both MWCNT-TD2 and MWCNT-TD2P nanosystems have been characterized by physical, chemical, and morphological analyses. Their chelating abilities towards the toxic metal ions Pb²⁺, Hg²⁺, and Ni²⁺ and the harmless Ca²⁺ ion have been experimentally evaluated in the two different sets of experiments and at the salt concentrations of 1 mg/mL or 1 μg/mL by inductively coupled plasma mass spectrometry (ICP-MS). The results of these studies pointed out the interesting chelating behavior for the phosphonated nanosystem towards the Hg²⁺ ion. The complexation mode of the best chelating system MWCNT-TD2P with mercury was investigated through density functional theory (DFT) calculations, suggesting a chelation mechanism involving the two oxygen atoms of the phosphate group. The synthesized dendrimers, supported on the multi-walled carbon nanotubes, have shown the potential to be used for the selective toxic metal ion removal and recovery.

The objective of this study was to evaluate the effects of alkaloid extracts of Prosopis juliflora (Sw.) D.C. pods obtained by two extraction methods as compared with sodium monensin on the gas production kinetic, mitigation of methane, and rumen fermentation products using wheat bran or Tifton 85 hay as substrates, by the semi-automatic in vitro gas production technique. A completely randomized design was adopted, and two natural additives were tested made from mesquite pod (alkaloid extract I and alkaloid extract II) at three levels (3.9, 7.9, and 12 μg), sodium monensin 5 μM (positive control), and no inclusion of additives (negative control). The volume of gases produced by the degradation of the fibrous fraction of wheat bran was influenced by the concentration of the extract I added to the medium, and the amounts of 7.9 and 12 μg were equal to monensin at the lowest value. The degradation rate of the fibrous carbohydrates with additive extract I at 12 μg was lower in relation to monensin. When Tifton 85 hay was utilized, alkaloid extract I provided a shorter colonization time as compared with monensin at the added amounts of 7.9 and 12 μg and higher production of gases from the fibrous fraction but without interfering with the total volume of gases produced during 96 h of fermentation of carbohydrates. In the periods of 12 and 24 h of incubation, utilizing alkaloid extract I, the mean values of methane production with wheat bran and Tifton 85 hay were lower than monensin (p < 0.05) when the respective amounts of 7.9 and 12 μg were added. Alkaloid extract I has similar potential to sodium in reducing production of total gases, methane, and the acetate/propionate ratio.

Behavioral ecotoxicology has become very important in the short time since a change in behavior is very often the first response to environmental altered conditions. We investigated the influence of trace metal intake on the spatial orientation performances of the carabid beetle Parallelomorphus laevigatus, fundamental ability for its survival. The aim of this study was to consider the solar orientation as behavioral biomarker for exposure to trace metal contamination. Therefore, we tested the ability of solar orientation of specimens of this species, fed with shrimps contaminated with three different concentrations of Cu, Zn, or Hg. We carried out the orientation tests after 1, 3, 7, and 10 days of contaminated feeding. Subsequently, we fed these beetles with not contaminated shrimps and again tested them after 1, 3, 7, and 10 days. For all three metals considered and, regardless of the degree of contamination of the food, we have found a progressive and significant counterclockwise displacement of the angle of orientation and a corresponding progressive reduction in the precision in the directional choices by the animals. We also noticed a clear growing recovery in the normal orientation by these insects after returning to their feeding with uncontaminated food. In conclusion, we can consider the orientation in space of P. laevigatus as a behavioral biomarker for exposure to trace metal contamination. We believe that the intake of trace metals may induce the insects to make mistakes in their spatial orientation, due to an acceleration of their biological clock. Such a clock malfunction is not definitive, since the return to a normal diet restores P. laevigatus the ability to re-make the correct directional choices. Ultimately, our results confirm the usefulness of behavioral ecotoxicology investigations; moreover, they stimulate the opportunity to deepen the understanding of functioning of the biological clock in the animals.

Phosphorus (P) fractionation and sorption behavior as well as the abundance and community composition of phosphorus-solubilizing bacteria (PSB) in sediments, including inorganic phosphate-solubilizing bacteria (IPB) and organic phosphate-mineralizing bacteria (OPB), were investigated in 27 sampling sites of five sections in the Three Gorges Reservoir (TGR) in December 2012. The calcium-bound phosphorus (CaCO₃∼P) accounted for the largest part for P fractions in the sediment of TGR, which was paralleled with IPB outnumbering OPB. Furthermore, some PSB isolates, such as Micromonospora sp., Aminobacter sp., and Arthrobacter sp., were shared by the IPB and OPB. Thus, some IPB species functioned as OPB and vise versa, which could be further reflected by a negative and significant relationship between PSB (IPB + OPB) number and content of CaCO₃∼P together with acid-soluble organic phosphorus (ASOP). Spatially, the highest ASOP content in the section Mudong and the highest CaCO₃∼P plus iron-bound phosphorus (Fe(OOH)∼P) as well as equilibrium phosphorus concentration (EPC₀) in the sections of Yunyang and Zigui due to sediment sequestration by the dam, as well as the positive relationship between EPC₀ and the ratios of different phosphorus species and phosphorus sorption maximum (Q ₘₐₓ), jointly indicated pathway diversification and potential risk of phosphorus release mediated by PSB in TGR.

Presented are results of a study on accumulation and distribution of ²¹⁰Po and ²¹⁰Pb in the fruitbodies of parasol mushroom (Macrolepiota procera) and risk to human consumer due to exposure from highly radiotoxic decay particles emitted by both radionuclides. Mushrooms were collected from 16 forested places in central and northern regions of Poland. Activity concentrations of ²¹⁰Po and ²¹⁰Pb were determined after radiochemical separation of nuclides and subsequent measurement using validated method and alpha spectrometer. Results showed on spatially heterogeneous distribution of the ²¹⁰Po and ²¹⁰Po activity concentrations in M. procera and two interpolation maps were prepared. Activity concentrations of nuclides in dried caps of M. procera were in the range from 3.38 ± 0.41 to 16.70 ± 0.33 Bq∙²¹⁰Po ∙kg⁻¹ and from 5.11 ± 0.21 to 13.42 ± 0.30 Bq∙²¹⁰Pb ∙kg⁻¹. Consumption of M. procera foraged in central and northern Poland should not contribute significantly to the annual effective radiation doses from ²¹⁰Po and ²¹⁰Pb due to amount of both nuclides accumulated by fungus in caps.

Integrated aerobic granules were first cultivated in two sequencing batch reactors (SBRs) (A1 and A2). Then, A1’s influent organic loading rate (OLR) was changed from alternating to constant (cycling time was still 6 h), while A2’s cycling time varied from 6 to 4 h (influent OLR strategy remained alternating). After 30-day operation since the manipulative alternations, granule breakage happened in two reactors at different operational stages, along with the decrease of granule intensity. Granule diameter in A1 declined from the original 0.84 to 0.32 cm during the whole operation, while granules in A2 dwindled to 0.31 cm on day 22 with similar size to A1. Both the amount of total extracellular polymeric substances (EPSs) and the protein were declining throughout the operation, and the large molecular weight of protein was considered closely related to the stability of aerobic granules. The relative AI-2 level decreased at the same time, and influent OLR strategy might had more evident impact on quorum sensing (QS) ability of sludge compared with starvation period. Combined with microbial results, the decline of total EPS amount in two reactors could be concluded as follows: During the reactor operation, some functional bacteria gradually lost their dominance and were eliminated from the reactors, which finally caused granule disintegration. In summary, the results further confirmed that alternating OLR and proper starvation period were two major factors in effective cultivation and stability of aerobic granules from the perspective of QS.
