A quantitative microbial risk assessment model for total coliforms and E. coli in surface runoff following application of biosolids to grassland

In Ireland, the land application of biosolids is the preferred option of disposing of municipal sewage waste. Biosolids provide nutrients in the form of nitrogen, phosphorus, potassium and increases organic matter. It is also an economic way for a country to dispose of its municipal waste. However, biosolids may potentially contain a wide range of pathogens, and following rainfall events, may be transported in surface runoff and pose a potential risk to human health. Thus, a quantitative risk assessment model was developed to estimate potential pathogens in surface water and the environmental fate of the pathogens following dilution, residence time in a stream, die-off rate, drinking water treatment and human exposure. Surface runoff water quality data was provided by project partners. Three types of biosolids, anaerobically digested (AD), lime stabilised (LS), and thermally dried (TD)) were applied on micro plots. Rainfall was simulated at three time intervals (24, 48 and 360 h) following land application. It was assumed that this water entered a nearby stream and was directly abstracted for drinking water. Consumption data for drinking water and body weight was obtained from an Irish study and assigned distributions. Two dose response models for probability of illness were considered for total and faecal coliform exposure incorporating two different exposure scenarios (healthy populations and immuno-compromised populations). The simulated annual risk of illness for healthy populations was below the US EPA and World Health Organisation tolerable level of risk (10⁻⁴ and 10⁻⁶, respectively). However, immuno-compromised populations may still be at risk as levels were greater than the tolerable level of risk for that subpopulation. The sensitivity analysis highlighted the importance of residence time in a stream on the bacterial die-off rate.

This study aimed to investigate the relationships between environmental exposure to metals/metalloids and semen quality, sperm apoptosis and DNA integrity using the metal/metalloids levels in seminal plasma as biomarkers. We determined 18 metals/metalloids in seminal plasma using an inductively coupled plasma-mass spectrometry among 746 men recruited from a reproductive medicine center. Associations of these metals/metalloids with semen quality (n = 746), sperm apoptosis (n = 331) and DNA integrity (n = 404) were evaluated using multivariate linear and logistic regression models. After accounting for multiple comparisons and confounders, seminal plasma arsenic (As) quartiles were negatively associated with progressive and total sperm motility using multivariable linear regression analysis, which were in accordance with the trends for increased odds ratios (ORs) for below-reference semen quality parameters in the logistic models. We also found inverse correlations between cadmium (Cd) quartiles and progressive and total sperm motility, whereas positive correlations between zinc (Zn) quartiles and sperm concentration, between copper (Cu) and As quartiles and the percentage of tail DNA, between As and selenium (Se) quartiles and tail extent and tail distributed moment, and between tin (Sn) categories and the percentage of necrotic spermatozoa (all Ptrend<0.05). These relationships remained after the simultaneous consideration of various elements. Our results indicate that environmental exposure to As, Cd, Cu, Se and Sn may impair male reproductive health, whereas Zn may be beneficial to sperm concentration.

Solomon Islands is rapidly developing its natural resource exploitation sector, but data needed to assess consequent environmental impacts are scarce. We assessed catchments surrounding the Gold Ridge gold mine (Guadalcanal, Solomon Islands) and found that extensive changes in river course, and water and sediment quality have occurred downstream of the gold mine since its development. Sediment run-off from exposed areas associated with the mine pit has increased, elevating turbidity (up to 2450 NTU) and metal and arsenic levels, with levels of the latter being up to 0.141 mg/L in surface waters and 265 mg/kg in sediments. An overfull, inoperative tailings storage facility associated with the currently inactive gold mine with fluctuating arsenic levels (up to 0.087 mg/L in the water; 377 mg/kg in the sediment) presents an ongoing threat to the environment. Arsenic, due to its toxicity, appears to be the greatest threat, with sediment and water guideline levels in rivers exceeded 10-fold and exceeded nearly 20-fold in the tailings dam sediments. Despite elevated metal and arsenic content in the area, no toxic inorganic arsenic was found to have bioaccumulated in locally harvested food. In summary, the natural environment surrounding the Gold Ridge mine has been modified substantially and requires an ongoing monitoring program to ensure the ecosystem services of food and water for the local communities continue to be safe. This study informs not only the local area but also provides a microcosm of the broader global challenges facing the regulation of extractive industries in proximity to subsistence communities.

Cacao from South America is especially used to produce premium quality chocolate. Although the European Food Safety Authority has not established a limit for cadmium (Cd) in chocolate raw material, recent studies demonstrate that Cd concentrations in cacao beans can reach levels higher than the legal limits for dark chocolate (0.8 mg kg−1, effective January 1st, 2019). Despite the fact that the presence of Cd in agricultural soils is related to contamination by fertilizers, other potential sources must be considered in Ecuador. This field study was conducted to investigate Cd content in soils and cacao cultivated on Ecuadorian farms in areas impacted by oil activities. Soils, cacao leaves, and pod husks were collected from 31 farms in the northern Amazon and Pacific coastal regions exposed to oil production and refining and compared to two control areas. Human gastric bioaccessibility was determined in raw cacao beans and cacao liquor samples in order to assess potential health risks involved. Our results show that topsoils (0–20 cm) have higher Cd concentrations than deeper layers, exceeding the Ecuadorian legislation limit in 39% of the sampling sites. Cacao leaves accumulate more Cd than pod husks or beans but, nevertheless, 50% of the sampled beans have Cd contents above 0.8 mg kg−1. Root-to-cacao transfer seems to be the main pathway of Cd uptake, which is not only regulated by physico-chemical soil properties but also agricultural practices. Additionally, natural Cd enrichment by volcanic inputs must not be neglected. Finally, Cd in cacao trees cannot be considered as a tracer of oil activities. Assuming that total Cd content and its bioaccessible fraction (up to 90%) in cacao beans and liquor is directly linked to those in chocolate, the health risk associated with Cd exposure varies from low to moderate.

Radical measures for controlling ambient air pollution sources were employed by the Chinese government during the Asia-Pacific Economic Cooperation (APEC) meeting in 2014, providing a unique case to evaluate the health effect benefits from such measures. To examine the cancer risk reduction from the source control measures during the APEC meeting, we estimated the reduction in population exposure to PM2.5 and PAHs and the reduction in PAHs-associated cancer risk if the control measures were sustained over time. We determined the population exposure to PM2.5 and PM2.5-bound PAHs for the 21.52 million Beijing residents using a Land Use Regression model to determine the spatial distribution of PM2.5 and a Monte Carlo approach to revise indoor/outdoor infiltration factor and time activity patterns. Into the model and approach, we incorporated the spatial variance and indoor/outdoor differences in the PM2.5 and PM2.5-bound PAHs concentrations, based on measurements. We then estimated lung cancer risk using the population attributable fraction (PAF), assuming the control measures were sustained over time. The mean PM2.5 exposure concentration decreased from 37.5 μg/m3 (CI:17.1–74.9 μg/m3) to 24.0 μg/m3 (CI:10.2–47.7 μg/m3), whereas the mean PM2.5-bound equivalent benzo[a]pyrene (BaPeq) exposure concentration decreased from 7.1 ng/m3 (CI:3.3–14.2 ng/m3) to 4.2 ng/m3 (CI:1.8–7.7 ng/m3), resulting in a reduction in the lung cancer PAF from 0.75% to 0.45%, if the measures were sustained over time.

This study examines size-resolved heavy metal data for particles sampled near an urban site affected by non-ferrous metal smelting in China with a focus on how particle sizes impact regional respiratory deposition behavior. Particles with aerodynamic diameters between 0.43 and 9 μm were collected during winter haze episodes from December 2011 to January 2012. The results showed that concentrations of individual trace elements ranged from ∼10⁻²–∼10⁴ ng/m³. Mass size distributions exhibit that Cu, Zn, As, Se, Ag, Cd, TI, and Pb have unimodal peak in fine particles range (<2.1 μm); Al, Ti, Fe, Sr, Cr, Co, Ni, Mo, and U have unimodal peak in coarse range (>2.1 μm), and Be, Na, Mg, Ca, Ba, Th, V, Mn, Sn, Sb, and K have bimodal profiles with a dominant peak in the fine range and a smaller peak in the coarse range. The total deposition fluxes of trace elements were estimated at 2.1 × 10⁻² – 4.1 × 10³ ng/h by the MPPD model, and the region with the highest contribution was the head region (42% ± 13%), followed by the tracheobronchial region (11% ± 3%) and pulmonary region (6% ± 1%). The daily intake of individual element for humans occurs via three main exposure pathways: ingestion (2.3 × 10⁻⁴ mg/kg/day), dermal contact (2.3 × 10⁻⁵ mg/kg/day), and inhalation (9.0 × 10⁻⁶ mg/kg/day). A further health risk assessment revealed that the risk values for humans were all above the guidelines of the hazard quotient (1) and cancer risk (10⁻⁶), indicating that there are potential non-cancer effects and cancer risks in this area.

Volatile organic compounds (VOCs) emissions from the chimney of a prevailing domestic stove fuelled with raw bituminous coal were measured under flaming and smoldering combustion processes in a farmer's house. The results indicated that the concentrations of VOCs quickly increased after the coal loading and achieved their peak values in a few minutes. The peak concentrations of the VOCs under the smoldering combustion process were significantly higher than those under the flaming combustion process. Alkanes accounted for the largest proportion (43.05%) under the smoldering combustion, followed by aromatics (28.86%), alkenes (21.91%), carbonyls (5.81%) and acetylene (0.37%). The emission factors of the total VOCs under the smoldering combustion processes (5402.9 ± 2031.8 mg kg⁻¹) were nearly one order of magnitude greater than those under the flaming combustion processes (559.2 ± 385.9 mg kg⁻¹). Based on the VOCs emission factors obtained in this study and the regional domestic coal consumption, the total VOCs emissions from domestic coal stoves was roughly estimated to be 1.25 × 10⁸ kg a⁻¹ in the Beijing-Tianjin-Hebei region.

Microbial methylation and demethylation are central to arsenic's (As) biogeochemical cycling. Here, the transformations of monomethylarsonic acid (MMA(V)) (50 mg L−1) for 15 days in cells of As-methylating fungi, Fusarium oxysporum CZ-8F1, Penicillium janthinellum SM-12F4, and Trichoderma asperellum SM-12F1, were evaluated, and trace concentrations of As(III) and As(V) were observed in fungal cell extracts. Trace amounts of DMA(V) were also detected in MMA(V) and P. janthinellum SM-12F4 incubations. In situ X-ray absorption near edge structure (XANES) indicated that after exposure to MMA(V) (500 mg L−1) for 15 days, 28.6–48.6% of accumulated As in fungal cells was DMA(V), followed by 18.4–30.3% from As(V), 0–28.1% from As(III), and 4.8–28.9% from MMA(V). The concurrent methylation and demethylation of As occurs in fungal cells. Furthermore, a majority of proteins involved in metabolism, transport, ATP activity, biosynthesis, signal transduction, DNA activity, translation, and oxidative stress were upregulated in T. asperellum SM-12F1 cells after MMA(V) exposure compared to As(III), As(V), and DMA(V). The detoxification process of T. asperellum SM-12F1 was As species-specific. Methylenetetrahydrofolate reductase (R7YMH0) donation of a methyl group for S-adenosylmethionine (SAM) generation significantly increased following MMA(V) exposure.

The causation between bioavailability of heavy metals and environmental factors are generally obtained from field experiments at local scales at present, and lack sufficient evidence from large scales. However, inferring causation between bioavailability of heavy metals and environmental factors across large-scale regions is challenging. Because the conventional correlation-based approaches used for causation assessments across large-scale regions, at the expense of actual causation, can result in spurious insights. In this study, a general approach framework, Intervention calculus when the directed acyclic graph (DAG) is absent (IDA) combined with the backdoor criterion (BC), was introduced to identify causation between the bioavailability of heavy metals and the potential environmental factors across large-scale regions. We take the Pearl River Delta (PRD) in China as a case study. The causal structures and effects were identified based on the concentrations of heavy metals (Zn, As, Cu, Hg, Pb, Cr, Ni and Cd) in soil (0–20 cm depth) and vegetable (lettuce) and 40 environmental factors (soil properties, extractable heavy metals and weathering indices) in 94 samples across the PRD. Results show that the bioavailability of heavy metals (Cd, Zn, Cr, Ni and As) was causally influenced by soil properties and soil weathering factors, whereas no causal factor impacted the bioavailability of Cu, Hg and Pb. No latent factor was found between the bioavailability of heavy metals and environmental factors. The causation between the bioavailability of heavy metals and environmental factors at field experiments is consistent with that on a large scale. The IDA combined with the BC provides a powerful tool to identify causation between the bioavailability of heavy metals and environmental factors across large-scale regions. Causal inference in a large system with the dynamic changes has great implications for system-based risk management.

Lead (Pb) is a well-known developmental toxicant. The aim of the present study was to analyze the association between maternal serum Pb level and risk of preterm birth in a population-based birth cohort study. The present study analyzed a sub-study of the China-Anhui Birth Cohort that recruited 3125 eligible mother-and-singleton-offspring pairs. Maternal serum Pb level was measured by graphite furnace atomic absorption spectrometry. All subjects were classified into three groups by tertile division according to serum Pb level: Low-Pb (L-Pb, <1.18 μg/dl), Medium-Pb (M-Pb, 1.18–1.70 μg/dl), and High-Pb (H-Pb, ≥1.71 μg/dl). The rate of preterm birth was 2.8% among subjects with L-Pb, 6.1% among subjects with M-Pb, and 8.1% among subjects with H-Pb, respectively. After controlling confounding factors, the adjusted OR for preterm birth was 2.33 (95%CI: 1.49, 3.65) among subjects with M-Pb and 3.09 (95%CI: 2.01, 4.76) among subjects with H-Pb. Of interest, maternal Pb exposure in early gestational stage than in middle gestational stage was more susceptible to preterm birth. Moreover, maternal serum Pb level was only associated with increased risk of late preterm birth. The present study provides evidence that maternal serum Pb level during pregnancy is positively associated with risk of preterm birth in a Chinese population.
