Expression of zinc and cadmium responsive genes in leaves of willow (Salix caprea L.) genotypes with different accumulation characteristics

Salix caprea is well suited for phytoextraction strategies. In a previous survey we showed that genetically distinct S. caprea plants isolated from metal-polluted and unpolluted sites differed in their zinc (Zn) and cadmium (Cd) tolerance and accumulation abilities. To determine the molecular basis of this difference we examined putative homologues of genes involved in heavy metal responses and identified over 200 new candidates with a suppression subtractive hybridization (SSH) screen. Quantitative expression analyses of 20 genes in leaves revealed that some metallothioneins and cell wall modifying genes were induced irrespective of the genotype's origin and metal uptake capacity while a cysteine biosynthesis gene was expressed constitutively higher in the metallicolous genotype. The third and largest group of genes was only induced in the metallicolous genotype. These data demonstrate that naturally adapted woody non-model species can help to discover potential novel molecular mechanisms for metal accumulation and tolerance.

Urban particulate air pollution is a serious health issue. Trees within cities can remove fine particles from the atmosphere and consequently improve air quality and human health. Tree effects on PM2.5 concentrations and human health are modeled for 10 U.S. cities. The total amount of PM2.5 removed annually by trees varied from 4.7 tonnes in Syracuse to 64.5 tonnes in Atlanta, with annual values varying from $1.1 million in Syracuse to $60.1 million in New York City. Most of these values were from the effects of reducing human mortality. Mortality reductions were typically around 1 person yr−1 per city, but were as high as 7.6 people yr−1 in New York City. Average annual percent air quality improvement ranged between 0.05% in San Francisco and 0.24% in Atlanta. Understanding the impact of urban trees on air quality can lead to improved urban forest management strategies to sustain human health in cities.

Metal contamination from mining activity is of great concern because of potential health risks to the local inhabitants. In the present study, we investigated the levels of Cd, Cu, As, Pb, and Zn in environmental samples and foodstuffs grown in the vicinity of the mines in Goseong, Korea, and evaluated potential health risks among local residents. Soils near the mines exceeded the soil quality standard values of Cu, As, and Zn contamination. The concentrations of Cd, Cu, Pb, and Zn in crop samples collected from the study area were significantly higher than those of the reference area. Some rice samples collected from the study area exceeded the maximum permissible level of 0.2 mg Cd/kg. The intake of rice was identified as a major contributor (≥75%) to the estimated daily intake among the residents. The average estimated daily intakes of metals were, however, below the provisional tolerable daily intake.

The occurrence and temporal variation of 18 perfluoroalkyl substances (PFASs) and 8 polybrominated diphenyl ethers (PBDEs) in the European Alps was investigated in a 10 m shallow firn core from Colle Gnifetti in the Monte Rosa Massif (4455 m above sea level). The firn core encompasses the years 1997–2007. Firn core sections were analyzed by liquid chromatography–tandem mass spectrometry (PFASs) and gas chromatography–mass spectrometry (PBDEs). We detected 12 PFASs and 8 PBDEs in the firn samples. Perfluorobutanoic acid (PFBA; 0.3–1.8 ng L−1) and perfluorooctanoic acid (PFOA; 0.2–0.6 ng L−1) were the major PFASs while BDE 99 (<MQL–4.5 ng L−1) and BDE 47 (n.d.–2.6 ng L−1) were the major PBDEs. This study demonstrates the occurrence of PFASs and PBDEs in the European Alps and provides the first evidence that PFASs compositions may be changing to PFBA-dominated compositions.

The adverse health effects of high concentrations of ground-level ozone are well-known, but estimating exposure is difficult due to the sparseness of urban monitoring networks. This sparseness discourages the reservation of a portion of the monitoring stations for validation of interpolation techniques precisely when the risk of overfitting is greatest. In this study, we test a variety of simple spatial interpolation techniques for 8-h ozone with thousands of randomly selected subsets of data from two urban areas with monitoring stations sufficiently numerous to allow for true validation. Results indicate that ordinary kriging with only the range parameter calibrated in an exponential variogram is the generally superior method, and yields reliable confidence intervals. Sparse data sets may contain sufficient information for calibration of the range parameter even if the Moran I p-value is close to unity. R script is made available to apply the methodology to other sparsely monitored constituents.

Methane (CH4) emission from constructed wetland has raised environmental concern. This study evaluated the influence of mono and polyculture constructed wetland and seasonal variation on CH4 fluxes. Methane emission data showed large temporal variation ranging from 0 to 249.29 mg CH4 m−2 h−1. Results indicated that the highest CH4 flux was obtained in the polyculture system, planted with Phragmites australis, Zizania latifolia and Typha latifolia, reflecting polyculture system could stimulate CH4 emission. FISH analysis showed the higher amount of methanotrophs in the profile of Z. latifolia in both mono and polyculture systems. The highest methanogens amount and relatively lower methanotrophs amount in the profile of polyculture system were obtained. The results support the characteristics of CH4 fluxes. The polyculture constructed wetland has the higher potential of global warming.

The extraction of bitumen from the Alberta oil sands using in-situ technologies is expanding at a rapid rate; however, investigations into the environmental impacts of oil sands development have focused on surface mining in the Athabasca region. We measured polycyclic aromatic hydrocarbons (PAH) in soils, spruce needles, and lake sediment cores in the Cold Lake oil sands region to provide a historical and spatial perspective on PAH contamination related to in-situ extraction activities. A pronounced increase in PAH concentrations was recorded in one of two study lakes (Hilda Lake) corresponding to the onset of commercial bitumen production in ∼1985. Distance from extraction rigs was not an important predictor of PAH concentrations in soils, although two samples located near installations were elevated in alkyl PAHs. Evidence of localized PAH contamination in Hilda Lake and two soil samples suggests that continued environmental monitoring is justified to assess PAH contamination as development intensifies.

Ozone concentration spatial patterns remain largely uncharacterized across the extensive wilderness areas of the Sierra Nevada, CA, despite being downwind of major pollution sources. These natural areas, including four national parks and four national forests, contain forest species that are susceptible to ozone injury. Forests stressed by ozone are also more vulnerable to other agents of mortality, including insects, pathogens, climate change, and ultimately fire. Here we analyze three years of passive ozone monitor data from the southern Sierra Nevada and interpolate landscape-scale spatial and temporal patterns during the summer-through-fall high ozone concentration period. Segmentation analysis revealed three types of ozone exposure sub-regions: high, low, and variable. Consistently high ozone exposure regions are expected to be most vulnerable to forest mortality. One high exposure sub-region has been documented elsewhere as being further vulnerable to increased drought and fire potential. Identifying such hot-spots of forest vulnerability has utility for prioritizing management.

Concerns over exposure to airborne particulate matter (PM) are on the rise. Currently monitoring of PM is done on the basis of interpolating a mass of PM by volume (μg/m3) but has the drawback of not taking the chemical nature of PM into account. Here we propose a method of collecting PM at its emission source and employing automated analysis with scanning electron microscopy associated with EDS-analysis together with light scattering to discern the chemical composition, size distribution, and time and space resolved structure of PM emissions in a heavily trafficated roundabout in Sweden. Multivariate methods (PCA, ANOVA) indicate that the technogenic marker Fe follows roadside dust in spreading from the road, and depending on time and location of collection, a statistically significant difference can be seen, adding a useful tool to the repertoiré of detailed PM monitoring and risk assessment of local emission sources.

A consortium consisting of para-nitrophenol utilizer Pseudomonas sp. strain WBC-3, meta-nitrophenol utilizer Cupriavidus necator JMP134 and ortho-nitrophenol utilizer Alcaligenes sp. strain NyZ215 was inoculated into soil contaminated with three nitrophenol isomers for bioaugmentation. Accelerated removal of all nitrophenols was achieved in inoculated soils compared to un-inoculated soils, with complete removal of nitrophenols in inoculated soils occurring between 2 and 16 days. Real-time polymerase chain reaction (PCR) targeting nitrophenol-degradation functional genes indicated that the three strains survived and were stable over the course of the incubation period. The abundance of total indigenous bacteria (measured by 16S rRNA gene real-time PCR) was slightly negatively impacted by the nitrophenol contamination. Denaturing gradient gel electrophoresis profiles of total and group-specific indigenous community suggested a dynamic change in species richness occurred during the bioaugmentation process. Furthermore, Pareto–Lorenz curves and Community organization parameters indicated that the bioaugmentation process had little impact on species evenness within the microbial community.
