Relationship between microbial community dynamics and process performance during thermophilic sludge bioleaching 全文

Heavy metals can be removed from the sludge using bioleaching technologies at thermophilic condition, thereby providing an option for biotreatment of wasted sludge generated from wastewater treatment. The purposes of this study were to establish a molecular biology technique, real-time PCR, for the detection and enumeration of the sulfur-oxidizing bacteria during the thermophilic sludge bioleaching. The 16S rRNA gene for real-time PCR quantification targeted the bioleaching bacteria: Sulfobacillus thermosulfidooxidans, Sulfobacillus acidophilus, and Acidithiobacillus caldus. The specificity and stringency for thermophilic sulfur-oxidizing bacteria were tested before the experiments of monitoring the bacterial community, bacterial number during the thermophilic sludge bioleaching and the future application on testing various environmental samples. The results showed that S. acidophilus was identified as the dominant sulfur-oxidizing bacteria, while A. caldus and S. thermosulfidooxidans occurred in relatively low numbers. The total number of the sulfur-oxidizing bacteria increased during the thermophilic bioleaching process. Meanwhile, the decrease of pH, production of sulfate, degradation of SS/VSS, and solubilization of heavy metal were found to correlate well with the population of thermophilic sulfur-oxidizing bacteria during the bioleaching process. The real-time PCR used in this study is a suitable method to monitor numbers of thermophilic sulfur-oxidizing bacteria during the bioleaching process.

Heavy metals cause serious problems in the environment, and they can be accumulated in organisms, especially in the higher fungi. The concentration of Ni, Cr, Pb, Cd, and Hg in 10 species of edible mushrooms in Medvednica Nature Park, Croatia was therefore determined. In addition, the similarity between the studied species was determined by cluster analysis based on concentrations of the aforementioned metals in the fruiting bodies. The contents of nickel, chromium, lead, cadmium, and mercury in the fruiting bodies of mushrooms were obtained by X-ray fluorescence spectrometry. The highest concentrations of Ni (3.62 mg kg⁻¹), Cr (3.01 mg kg⁻¹), and Cd (2.67 mg kg⁻¹) were determined in Agaricus campestris. The highest concentration of Pb (1.67 mg kg⁻¹) was determined in Macrolepiota procera, and the highest concentration of Hg (2.39 mg kg⁻¹) was determined in Boletus edulis. The concentration of all heavy metals significantly differed (p < 0.001) between examined saprophytic and ectomycorrhizal mushrooms. Considering anatomical part of the fruiting body (cap-stipe), a considerably higher concentration of the analyzed elements was found in the cap for all mushroom species. According to calculated bioconcentration factors, all the examined species were found to be bioexclusors of Ni, Cr, and Pb and bioaccumulators of Cd and Hg. Cluster analysis performed on the basis of the accumulation of the studied metals revealed great phenotypic similarity of mushroom species belonging to the same genus and partial similarity of species of the same ecological affiliation.

Effluents after biochemical treatment contain pollutants that are mostly non-degradable. Based upon previous pilot-scale test results, an industrial-scale electro-oxidation device was built to decompose these refractory materials in the effluent from a park wastewater treatment plant. The electro-oxidation device comprised a ditch-shaped plunger flow electrolysis cell, with mesh-plate Ti/PbO₂ electrodes as the anode and the same size mesh-plate Ti as the cathode. Wastewater flowed vertically through electrodes; the effective volume of the cell was 2.8 m³, and the surface-to-volume ratio was 17.14 m² m⁻³. The optimal current density was 100 A m⁻², and a suitable flow velocity was 14.0 m h⁻¹. The removal efficiencies for chemical oxygen demand and color in the effluent were over 60.0 and 84.0 %, respectively. In addition, the electro-oxidation system offered a good disinfection capability. The specific energy consumption for this industrial-scale device was 43.5 kWh kg COD⁻¹, with a current efficiency of 32.8 %, which was superior to the pilot-scale one. To meet the requirements for emission or reuse, the operation cost was $0.44 per ton of effluent at an average price for electricity of $0.11 kWh⁻¹.

Acid rain can directly or indirectly affect plant physiological functions, especially photosynthesis. The enzyme ATP synthase is the key in photosynthetic energy conversion, and thus, it affects plant photosynthesis. To clarify the mechanism by which acid rain affects photosynthesis, we studied the effects of acid rain on plant growth, photosynthesis, chloroplast ATP synthase activity and gene expression, chloroplast ultrastructure, intracellular H⁺ level, and water content of rice seedlings. Acid rain at pH 4.5 remained the chloroplast structure unchanged but increased the expression of six chloroplast ATP synthase subunits, promoted chloroplast ATP synthase activity, and increased photosynthesis and plant growth. Acid rain at pH 4.0 or less decreased leaf water content, destroyed chloroplast structure, inhibited the expression of six chloroplast ATP synthase subunits, decreased chloroplast ATP synthase activity, and reduced photosynthesis and plant growth. In conclusion, acid rain affected the chloroplast ultrastructure, chloroplast ATPase transcription and activity, and P ₙ by changing the acidity in the cells, and thus influencing the plant growth and development. Finally, the effects of simulated acid rain on the test indices were found to be dose-dependent.

The potential of microalgae consortia used in dairy wastewater treatment combined with microalgae biodiesel feedstock production was evaluated by comparing the nutrient removal of dairy wastewater, the growth of cells, and the lipid content and composition of biomass between monoalgae and microalgae consortia cultivation system. Our results showed that higher chemical oxygen demand (COD) removal (maximum, 57.01–62.86 %) and total phosphorus (TP) removal (maximum, 91.16–95.96 %) were achieved in almost microalgae consortia cultivation system than those in Chlorella sp. monoalgae cultivation system (maximum, 44.76 and 86.74 %, respectively). In addition, microalgae consortia cultivation except the mixture of Chlorella sp. and Scenedesmus spp. reached higher biomass concentration (5.11–5.41 g L⁻¹), biomass productivity (730.4–773.2 mg L⁻¹ day⁻¹), and lipid productivity (143.7–150.6 mg L⁻¹ day⁻¹) than those of monoalgae cultivation (4.72 g L⁻¹, 674.3, and 142.2 mg L⁻¹ day⁻¹, respectively) on the seventh day. Furthermore, the fatty acid methyl ester (FAME) profiles indicated the lipids produced from microalgae consortia cultivation system were more suitable for biodiesel production. The microalgae consortia display superiority in dairy wastewater treatment and the getting feedstock for biodiesel production.

Thirteen particle-phase PAHs, including nine >4-ring congeners [Benz[a]anthracene (BaAn), Chrysene (Chry), Benzo[b]fluoranthene (BbF), Benzo[k]fluoranthene (BkF), Benzo[e]pyrene (BeP), Benzo[a]pyrene (BaP), Dibenzo[a,h]anthracene (dBaAn), Benzo[g,h,i]perylene (BghiPe), Indeno(1,2,3-c,d)pyrene (IP)], listed by IARC (International Agency for Research on Cancer) as class 1, class 2A, and 2B carcinogens, plus four ≤4-ring congeners [Phenanthrene (Ph), Anthracene (An), Fluoranthene (Fl), Pyrene (Py)], were concurrently measured in inhalable and respirable particle fractions (PM₁₀ and PM₂.₅) at a heavy-traffic and an urban background site in Thessaloniki, northern Greece, during the warm and the cold period of the year. Carcinogenic and mutagenic potencies of the PAH-bearing particles were calculated, and the inhalation cancer risk (ICR) for local population was estimated. Finally, Chemical Mass Balance (CMB) modeling was employed for the source apportionment of ambient PAH levels and the estimated lung cancer risk. Resulted inhalation cancer risk during winter was found to be equivalent in the city center and the urban background area suggesting that residential wood burning may offset the benefits from minor traffic emissions.

Sediments function both as a sink and a source of pollutants in aquatic ecosystems and may impose serious effects on benthic organisms and human health. As one of the largest estuaries in the world, the Yangtze River estuary suffers from abundant wastewater from the coastal cities. In this study, the zebrafish (Danio rerio) embryos were employed in the fish embryo test and a comet assay to evaluate the embryotoxicity and genotoxicity of the sediments from the Yangtze River estuary, respectively. Results showed that the sediments from the Yangtze River estuary significantly increased mortality, induced development abnormalities, and reduced hatching rate and heart rate of zebrafish embryos after 96 h of exposure. Significant genotoxicity was observed in the samples relative to the controls. Relatively low-level embryotoxicity and genotoxicity of sediments were found in the Yangtze River compared with other river systems. Toxic responses were also discussed in relation to the analyzed organic contaminants in sediments. More attention should be paid to non-priority pollutant monitoring in the Yangtze River estuary.

The present review focused on the levels and toxicological status of heavy metals especially chromium (Cr) in the exposed workers from different occupational settings around the globe and in Pakistan. It was found that exposed workers from leather tanning and metal plating units showed elevated levels of Cr than the workers from other occupational settings. Cr and other heavy metals level in biological matrices of the exposed workers in different occupational settings revealed that developing countries are severely contaminated. Occupational settings from the Sialkot district, Pakistan exhibited elevated level of Cr in biological entities of the exposed workers. Review suggested that higher level of Cr exposure to the workers enhance the oxidative stress (reactive oxygen species (ROS) and hydroxyl (OH) radical generation) which may cause; cellular and molecular damage such as genotoxicity and chromosomal aberration formations, and carcinogenic effects. This review will help to understand the Cr contamination mechanisms and associated health implications in different occupational settings around the globe in general and particularly to Pakistan. This study will also assist occupational health and safety management authorities to devise or change the Cr recommended exposure limits (REL) for different occupational settings.

Monitoring the levels of trace elements in hair can allow estimating the effects of the geographical location and also can provide a notion of the metal body burden. However, the use of human hair is controversial due to the different confounding factors that could affect the presence of trace elements in hair. As a result, a comprehensive monitoring study was performed in Alcalá de Henares, one of the major cities in the Madrid region, Spain. Trace elements have been monitored in urban topsoils and in human hair of two well-defined and healthy groups of population: children (6–9 years) and adolescents (13–16 years). The city was divided into four areas or zones with different characteristics to assess the possible effect of area of residence and age in the presence of Al, As, Be, Cd, Cr, Cu, Hg, Mn, Pb, Sn, Ti, Tl and Zn in soils and hair. There is no current hypothesis that explains the possible effect of the area of monitoring in the distribution of Be, Cr, Ni, Sn and Ti found in these urban soils, maybe because urban soils receive high disturbance, and there are many factors involved. The presence of most of the trace elements monitored was significantly higher in the hair of the children population, except for Sn and Zn. This could be attributed mainly to dietary habits. Other factors influencing metal content in hair such as environmental factors would have had a minimal effect in the population groups here studied. Finally, none of the levels of trace elements studied in hair were significantly correlated with levels measured in the topsoils of public parks in Alcalá de Henares, with the exception of Pb in adolescent participants.

The effects of modified attapulgite (MA) on the dissipations of the plasticizers di-n-butyl phthalate (DBP) and di-(2-ethylhexyl) phthalate (DEHP) in soil, as well as on the composition of soil microbial community, were studied. DBP, DEHP (50 mg kg⁻¹ in soil, respectively), and MA (1, 5, and 10 % in soil) were mixed thoroughly with soil and incubated for 60 days. DBP- and DEHP-contaminated soils without MA were used as the controls. Both of DBP and DEHP residues in bulk soils and four soil fractions were measured at five incubation times 1, 7, 15, 30, and 60 days, and their dissipation kinetic equations were analyzed. The microbial phospholipid fatty acid (PLFA) concentrations were also measured at the end of experiment. Our results showed that the effect of modified attapulgite on DBP dissipation was related to its dosage in soil. The DEHP dissipation was both inhibited by MA at the 5 and 10 % rates in soils. The application of MA changed the content percentages but did not change the concentration order of phthalate acid esters (PAEs) in soil particle-size fractions. The total microbial PLFA content was significantly increased by 5 and 10 % MA in the contaminated soils. Meanwhile, the gram-negative (GN)/gram-positive (GP) ratios increased when MA was applied at the dosages of 5 and 10 % in DBP and 10 % in DEHP-contaminated soils. Principal component analysis (PCA) indicated that the change of bacteria PLFA, especially the GN bacterial PLFA, depended on the dosages of MA added into soil. The application of MA into soil has a positive effect on reducing the eco-toxicity of PAEs in soil based on the analysis of the soil microbial PLFA.
