Pendimethalin induces apoptosis in testicular cells via hampering ER-mitochondrial function and autophagy

Pendimethalin (PDM) is a dinitroaniline crop pesticide that is extensively utilized worldwide. However, the reproductive toxicity and cellular mechanisms of PDM have not been identified. Therefore, we elucidated the adverse effects of PDM on the reproductive system using mouse testicular Leydig and Sertoli cells (TM3 and TM4 cells, respectively). Our results demonstrated that PDM suppressed the viability and proliferation of TM3 and TM4 cells. Additionally, PDM induced cytosolic calcium upregulation and permeabilization of mitochondrial membrane potential in both TM3 and TM4 cells. We also verified that PDM activates the endoplasmic reticulum (ER) stress pathway and autophagy. Furthermore, we confirmed that activation of ER stress and autophagy were blocked by 2-aminoethoxydiphenyl borate (2-APB) treatment. Finally, we confirmed PDM-induced cell cycle arrest and apoptosis in TM3 and TM4 cells. Thus, we first demonstrated that PDM impedes the survival of testis cells, and further, their function.

Sedum alfredii is a Cd/Zn hyperaccumulator native to China, which was collected from a mined area where Mn content in soil was extremely high, together with Zn and Cd content. We investigated the tolerance and accumulation ability of Mn and its possible association with Cd hyperaccumulation in this plant species by using MP-AES, SR-μ-XRF, and RT-PCR. The results showed that the hyperaccumulating ecotype (HE) S. alfredii exhibited high tolerance to Mn and accumulating around 10,000 and 12,000 mg kg⁻¹ Mn in roots and shoots, respectively, without exhibiting toxicity under 5000 mg kg⁻¹ Mn treatment for 4 weeks. Exposure to Cd significantly reduced plant uptake of Mn. In contrast, exogenous Mn application significantly improved root uptake and root-to-shoot translocation of Cd, resulting in the increased Cd accumulation in the shoots of HE S. alfredii. SR-μ-XRF analysis demonstrated that high Mn (20 μM) exposure resulted in higher intensities of Cd localized in both stem vascular bundles and cortex, as well as leaf mesophyll cells, than in those treated with low Mn levels (0.2 μM or 2.0 μM). RT-PCR analysis of several genes possibly involved in Mn/Cd transportation showed that expression of SaNramp3 in roots was significantly reduced under high Mn exposure. These results suggested a significant interaction between Cd and Mn in the HE S. alfredii plants, possibly through their competition for transporters and theoretically provided a strategy to improve the efficiency of Cd extraction from polluted soils by this plant species, after using appropriate nutrient management of Mn.

Effluent is often treated with ozone before being discharged into a natural water environment. This process will change the interaction between effluent organic matter and pollutants in aquatic environment. The impact of ozonation on complexation between dissolved organic matter in such wastewater and sulfadimidine often found in natural water was studied in laboratory experiments using four types of real wastewater. Ozonation was found to decrease the proportion of organic matter with a molecular weight greater than 5 kDa as well as protein-like, fulvic-like and humic-like components, but except the proportion of hydrophilic components. The aromaticity of the dissolved organic matter was also reduced after ozonation. The complexation of tryptophan and tyrosine with sulfadimidine mainly depends on their hydrophobicity and large molecular weight. Ozonation of fulvic and humic acid tends to produce small and medium molecular weight hydrophilics. The complexation of humic and fulvic acids with sulfadimidine was enhanced by ozonation. Dissolved organic matter, with or without oxidation, were found to weaken sulfadimidine’s inhibition of microbial growth, especially for Aeromonas and Acinetobacter species. This finding will expand our understanding about the impact of advanced treatment processes on the dissolved organic matters’ properties in effluent.

The distribution of freshwater and marine microplastics (MPs) varies due to the difference in fresh and seawater densities and MP sources. This study aims to investigate the abundance of MPs and their possible sources in surface waters of different ecosystems, such as sea, lagoon, and lake. We classified MPs in terms of their color and type and established the relationship between the MPs in surface waters with different characteristics. The mean MP abundance (33 particles L⁻¹) detected herein was higher than that in the previously conducted studies. Fragment particles (37.95%) were determined to be the dominant MP type, and the predominant MP color was blue (75.28%). As for the seasonal MP distribution, its highest content (48.03 particles L⁻¹) was observed in autumn, unlike that reported by other studies. The findings of this study reveal the effects of wastewater treatment plant (WWTP) discharge and current flow on the MP distribution in the study area. This study aims to provide representative data on the MP abundance and distribution, as well as MP-affecting parameters for similar aquatic areas in other parts of the world.

Ciprofloxacin (CFX) and ofloxacin (OFX) are two of the most often used fluoroquinolone antibiotics, and their residues are found in large amounts in various aquatic settings. However, the toxicity tests of CFX using eukaryotic organisms such as Daphnia magna are inadequate, and the test result of OFX is currently unknown. Therefore, the chronic toxicity test for D. magna was performed during 42 days under exposure to CFX and OFX concentrations of 50, 500, and 5000 μg L⁻¹. All exposure conditions did not cause mortality for D. magna. CFX exposure at 500 μg L⁻¹ resulted in an earlier oogenesis date and increased brood size in the second birth. The Poisson-based generalized linear mixed-effects model revealed that the reduction of fertility was statistically significant for the CFX and OFX exposures at 5000 μg L⁻¹. On the other hand, the production of dead eggs as offspring degradation was also found significantly as maternal D. magna exposed to antibiotics at 5000 μg L⁻¹. In addition, following long-term exposure to antibiotics, maternal adaptation to antibiotics was established for offspring deterioration and fertility. However, the OFX exposure showed that the fertility-suppressed effects continued for a longer period than the CFX exposure. Although no rational explanation has yet been given for the more substantial effect of OFX on reducing fertility than CFX, molecular cell biology and symbiotic microbial flora derived from previous studies could explain our ecotoxicological results. This study is the first report for the OFX chronic toxicities on D. magna by comparing it to the toxicity of CFX. Our study contributes to guiding the future impact assessment of fluoroquinolone antibiotic pollution on ecosystems, including the need for new statistical methods in ecotoxicological studies.

Pollution damages ecosystems around the globe and some forms of pollution, like oil pollution, can be either man-made or derived from natural sources. Despite the pervasiveness of oil pollution, certain organisms are able to colonise polluted or toxic environments, yet we only have a limited understanding of how they are affected by it. Here, we analysed phenotypic responses to oil pollution in guppies (Poecilia reticulata) living in oil-polluted habitats across southern Trinidad. We analysed body-shape and life-history traits for 352 individuals from 11 independent populations, six living in oil-polluted environments (including the naturally oil-polluted Pitch Lake), and five stemming from non-polluted habitats. Based on theory of, and previous studies on, responses to environmental stressors, we predicted guppies from oil-polluted waters to have larger heads and shallower bodies, to be smaller, to invest more into reproduction, and to produce more but smaller offspring compared to guppies from non-polluted habitats. Contrary to most of our predictions, we uncovered strong population-specific variation regardless of the presence of oil pollution. Moreover, guppies from oil-polluted habitats were characterised by increased body size; rounder, deeper bodies with increased head size; and increased offspring size, when compared to their counterparts from non-polluted sites. This suggests that guppies in oil-polluted environments are not only subject to the direct negative effects of oil pollution, but might gain some (indirect) benefits from other concomitant environmental factors, such as reduced predation and reduced parasite load. Our results extend our knowledge of organismal responses to oil pollution and highlight the importance of anthropogenic pollution as a source of environmental variation. They also emphasise the understudied ecological heterogeneity of extreme environments.

This study investigates the occurrence and distribution of microplastics in water, sediment, and crayfish samples within pond and rice-crayfish co-culture breeding modes in Jianli prefecture, China. Microplastics in environmental and biological samples were systematically extracted by CaCl₂ solution, digested by H₂O₂ and KOH, and identified by μ-FTIR. A cleansing treatment for crayfish was performed in pure water before dissection and microplastic accumulation in different tissues (gill, stomach, gut, and flesh) of non-cleansed and cleansed crayfish were compared. The average microplastic abundances were 1.3 ± 0.1–2.5 ± 0.1 particles/L, 0.03 ± 0.01–0.04 ± 0.02 particles/g, and 0.17 ± 0.07–0.92 ± 0.19 particles/individual in water, sediment, and crayfish samples, respectively. Microplastics were detected in all studied crayfish tissues, except the flesh. There were no significant differences in microplastic abundances in water (P = 0.82), sediment (P = 0.90), and crayfish (P = 0.47 for non-cleansed samples; P = 0.30 for cleansed samples) between two breeding modes despite the detection of relatively higher microplastic abundances in the samples from the pond breeding mode. Microplastic accumulation in non-cleansed crayfish stomachs and guts (0.71 ± 0.18 particles/individual) was higher (P < 0.01) than that recorded in their gills (0.13 ± 0.06 particles/individual). Moreover, microplastics present in the stomachs and guts of cleansed crayfish were significantly less abundant (P < 0.01) than in non-cleansed crayfish, although this was not observed in the gills (P = 0.99). The majority of microplastic particles in this study had fiber-like shapes, blue and transparent colors, a size smaller than 1 mm, and polymer types of PP:PE and PE. The results demonstrate that microplastics in the environment can accumulate in the internal tissues of crayfish, which may pose a potential risk to humans through food consumption without the removal of the gills, stomach, and guts. This study provides valuable information for understanding microplastic accumulation in the different tissues of crayfish and the potential risk of human exposure to microplastics from crayfish as a food supplement.

Delivering quality-changed water often contributes to the biological instability of drinking water distribution systems (DWDS). However, the potential effects of quality-changed water on the biostability within DWDS are not well understood, especially after water switching to quality-improved water. The objective of this study was to investigate the effects of quality-improved water on DWDS, focusing on the stability of biofilm. The practical aged-pipe was assembled into pipe reactors to simulate the effect of switching to quality-improve water. The adenosine triphosphate (ATP) concentration of bulk water in the pipe reactors increased from ∼1.2 ng/L to almost above 5 ng/L when fed water switching to TP 2. Biomass quantified by measuring ATP concentration confirmed that the risk of biofilm release through aged cast-iron (CI) pipe surfaces after water source switching. The changes in water characteristics due to quality-improved water source could cause bacteria release in DWDS at the initial period (at the first 7 days). However, the DWDS can establish the new stable phase after 42 days. Over time, biomass in the bulk water of the distribution system decreased significantly (The ATP concentration in the bulk maintains around 3 ng/L) after 42 days, indicating the improvement of water quality. The biofilm was dominated by bacteria related to iron-cycling process, and at the genus level, Desulfovibrio had the highest relative abundance, however, it decreased significantly (from 48% to 9.3%) after water source switching. And there was a slightly increase in the fraction of iron-oxidizing bacteria (IOB) and siderophore-producing bacteria (SPB), but a relatively higher increase in nitrate-reducing bacteria (NRB), nitrobacteria (NOB), and iron-reducing bacteria (IRB) was observed. Taken together, these results and the corrosion morphology, indicate that pipe biofilm and corrosion were chemically and microbially stable after re-stability under water source switching. In addition, the bulk water environment showed a marked decrease in selected bacteria at genus level, including pathogenic species, indicating the improvement of quality in drinking water.

140 contaminants belonging to various classes (organochlorine and organophosphorus pesticides, pyrethroid insecticides, carbamates, fungicides, acaricides, herbicides, synergists, insect growth regulators, polychlorobiphenyls, polycyclic aromatic hydrocarbons) were simultaneously analysed by GC-MS/MS in marine sediments, aquatic plant leaves and fish tissues samples. A total of 260 samples from five stations along the coast of Tunisia were evaluated. The results highlight that only 28 residues (12 polychlorobiphenyls, 8 organochlorine pesticides, 7 polycyclic aromatic hydrocarbons and triphenyl phosphate) were detected at levels higher than relative LOQ values. The amounts in sediment samples were compared with Sediment Quality Guidelines (SQGs) showing that the values are acceptable and no toxic effect is expected on aquatic organisms. A little variation of contaminant residues in sediment samples among coastal stations was recorded. Namely, with respect to almost all polychlorobiphenyls and organochlorine pesticides, higher values were recorder in summer. With respect to almost all polycyclic aromatic hydrocarbons, higher values were recorder in autumn. Aquatic plant leaves showed a residue accumulation higher than that of other compartments of marine system. The data about fish samples (Sparus aurata and Sarpa salpa, the two most frequently caught fish species at five sites on the central coast of Tunisia) do not pose direct hazard to human health because values were lower than protection limits.

The abnormal elevation of cyanobacterial density and total phosphorus concentration after the reduction of exogenous pollutants in Lake Taihu is still an open question. An in-situ light-dark bottle method was used to investigate the spatiotemporal differences of phosphorus release potential of bloom-forming cyanobacteria (BFC) in Lake Taihu. Generalized additive model analysis (GAM) of field data revealed that the phosphorus release potential of BFC increased with the upregulation of Chlorophyll a (Chl-a) content per cell, which was further validated by the laboratory experiment results. We deduced that the accumulation of Chl-a content per cell might be an essential index of high phosphorus release potential of BFC. The phosphorus release potential of BFC was much higher in summer and autumn than that in spring and winter, while the phosphorus absorption potential increased with the rising of temperature. The distinct physiological status of BFC at different seasons brought about their variation in phosphorus release potential. Additionally, high phosphorus release potential of BFC region mainly concentrated in the eastern and the central, northwest, western, and the south of Lake Taihu in spring, summer, autumn, and winter, respectively. Further studies showed that the spatial differences in phosphorus release potential of BFC were most probably due to the horizontal drift of BFC driven by the prevailing wind. Collectively, the synergism of BFC’s physiological status and horizontal drift determined the spatiotemporal differences of phosphorus release potential of BFC in Lake Taihu. Moreover, apparent spatiotemporal differences in phosphorus release potential of BFC were essential factors that induced the distinct distribution of total phosphorus in Lake Taihu. This study provides insight for exploring the reason for the constant increase of total dissolved phosphorus concentration and cyanobacterial density in Lake Taihu for the past 5 years.
