Impacts of selenium supplementation on soil mercury speciation, and inorganic mercury and methylmercury uptake in rice (Oryza sativa L.) 全文

Rice grain is known to accumulate methylmercury (MeHg) and has been confirmed to be the major pathway of MeHg exposure to residents in mercury (Hg) mining areas in China. Selenium (Se) supplementation has been proven to be effective in mitigating the toxicity of Hg. To understand how Se supplementation influences soil Hg speciation, a wide range of Se (0–500 mg/kg) was applied to Hg polluted paddy soils in this study, which decreased MeHg concentration in soil from 2.95 ± 0.36 to 0.69 ± 0.16 μg/kg (or 77%). After Se addition, humic acid state Hg (F4) was transformed into strong-complexed state Hg (F5), indicating that Hg bound up to the non-sulfur functional groups of humic acid (non-RSH) was released and reabsorbed by strong binding Se functional group (F5). As a result, inorganic Hg (IHg) was reduced by >48%, 18%, and 80% in root, stem, and grain, respectively, however, the reduction was not apparent in leaf. Substantial reductions were also found for MeHg in grain and root, but not in stem and leaf. Soil is suggested to be the main source of both MeHg and IHg in rice grain. Such a finding may provide an idea for improving Hg-polluted paddies through controlling soil IHg and MeHg. Further research on the molecular structure of the strong-complexed Hg in F5 should be conducted to elucidate the mechanism of Hg-Se antagonism.

Schizophrenia is a devastating neuropsychiatric disorder with increasing concern. Limited studies have been conducted to assess the relationship between short-term exposure to ambient air pollution and schizophrenia attacks. This study aimed to investigate the associations between short-term air pollution exposure and schizophrenia outpatient visits based on a time-series study performed in China. Daily data of schizophrenia outpatient admissions and air pollution from 1 October 2010 to 31 December 2013 were collected in Xi'an, a heavily-polluted city in China. We utilized a time-series Poisson regression model to examine the associations between short-term air pollution and schizophrenia outpatient visits with different lag days. A total of 34,865 outpatient-visits for schizophrenia were identified. A 10 μg/m³ increase of PM₁₀, SO₂, and NO₂ concentrations corresponded to 0.289% (95% Cl: 0.118%, 0.460%), 1.374% (95% Cl: 0.723%, 2.025%), and 1.881% (95% Cl: 0.957%, 2.805%) elevation in outpatient-visits for schizophrenia at lag 0, and the associations appeared to be stronger, although not statistically significantly, in females and in middle and older age adults (40 and over). The most significant associations were observed on the concurrent day in different lag models. In conclusion, short-term exposure to ambient air pollution (PM₁₀, SO₂, and NO₂) can be associated with increased risk of daily outpatient visits for schizophrenia, which may contribute to the further understanding of the potential adverse effects of air pollution in schizophrenia and other neuropsychiatric disorders.

Arsenic (As) tends to mobilize in flooded paddy soil due to the reductive dissolution of the iron (oxyhydr)oxides to which As sorbs, resulting in elevated As accumulation in rice that poses a potential risk to the food safety and human health. Microbial sulfate reduction is an important biogeochemical process in paddy soils, but its impact on As mobilization remains poorly understood. In this study, we incubated eight As-contaminated paddy soils under flooded conditions to investigate the effect of sulfate addition on As mobility. Porewater Fe and As concentrations and As species were determined. Among the eight soils, an addition of 50 mg S kg⁻¹ as sodium sulfate decreased porewater arsenite only in two soils, which also showed a high mobilization of Fe²⁺. Further experiments showed that the addition of sulfate to these two soils stimulated microbial sulfate reduction but decreased porewater concentrations of both arsenite and Fe²⁺. Additionally, the supply of sulfate increased the fractions of As associated with acid volatile sulfides in the solid phase and decreased As uptake by rice in pot experiments under similar conditions. The effect of sulfate addition on porewater As was diminished by the addition of molybdate, an inhibitor of sulfate reducing bacteria. These results suggest the formation of secondary FeS minerals which co-precipitate or sorb arsenite as a likely mechanism of As immobilization, which was also supported by thermodynamic modeling of the pore water. Thus, sulfate additions can immobilize As and reduce its availability to rice plants in paddy soils containing a high potential for microbial Fe reduction, providing an efficient way to mitigate the As transfer to the food chain.

Though the toxicity of strobilurins on non-target aquatic organisms has been characterized, the associated toxic mechanisms have not been fully explored. The present study showed that the larval stage was the most sensitive developmental stage in zebrafish, and pyraclostrobin (PY) had the highest acute toxicity to embryos, larvae, juvenile and adult with 96 h-LC₅₀ at 0.048 mg/L, 0.029 mg/L, 0.039 mg/L, 0.031 mg/L respectively, when compared with the toxicity of trifloxystrobin (TR), kresoxim-methyl (KM) and azoxystrobin (AZ) at corresponding developmental stage. Then we investigated the transcriptomics and developmental toxicity of TR, KM, AZ and PY on zebrafish embryos after 72 h exposure. RNA-seq revealed that the pathways related to cell apoptosis and cancer, and cellular components organelle membrane and mitochondrion, were markedly affected after TR, KM, AZ and PY exposure during zebrafish early life stages. The results were further confirmed by the induction of antioxidant enzymes catalase (CAT) and superoxide dismutase (SOD) activities, the elevation of H₂O₂, malondialdehyde (MDA) and reactive oxygen species (ROS) level, as well as the reduction of intracellular calcium ions (Ca²⁺) and mitochondrial membrane potential (MMP), which indicated that strobilurins could cause mitochondrial dysfunction and cell apoptosis. The present study was performed a systematic analysis of strobilurins to zebrafish at multi-levels, which provided suggestions for further investigation of molecular mechanisms underlying the toxicity induced by strobilurins on aquatic organisms.

Immobilization of enzymes on carriers have been pursued to make the enzyme stable, reusable and obtaining even better enzyme activity. Due to the highly stable two-dimensional layer structure, large surface area and pore volume, graphene materials were seemed as ideal carrier for enzyme immobilization. In this paper, pristine few layer graphene (FLG) was applied to interact with laccase to synthesize laccase-graphene composite and the results of AFM, FT-IR and adsorption isotherm suggested that laccase was loaded on the FLG with a very high loading dosage (221.1 mg g⁻¹). Based on the measured interaction force and binding type between laccase and graphene, we proposed that the great enzyme loading on FLG is likely due to the non-covalent π-π stacking in addition to the large surface area of FLG. The composite has better stability to the variance of pH and storage temperature than free laccase. The synthesized composite can effectively transform beta-blocker labetalol with an enhanced efficiency, though the possible reaction pathways kept not changing. We further performed molecular simulation study on the crystal structure variation of laccase binding on FLG and proposed that catalytic activity enhancement may be attributed to the more exposure extent of the catalytic center of laccase. In addition, the laccase-graphene composite can be reused more than ten times in catalyzing the labetalol removal.

The cell-free dithiothreitol (DTT) assay is widely used and the DTT consumption rate is interpreted to assess the oxidative potential (OP). Most researchers use an experimental procedure developed by Cho et al. (2005) while some adopt a procedure by Li et al. (2009). The key difference between the two procedures is the initial DTT concentration, 100 μM used in the former and 20 μM in the latter, raising an unaddressed issue of comparability. We examine in this work this issue using metal-free humic-like substance (HULIS) samples isolated from ambient aerosol and two metals (i.e. copper and manganese). We found that higher initial DTT concentrations led to higher DTT consumption rates for both HULIS and metals. For HULIS, the increase in DTT consumption rate was proportional to the initial DTT concentration (i.e., roughly by 5-fold), allowing correction of the concentration effect and direct comparison of results from the two protocols. However, the proportionality did not hold for the metals or metal-organic mixtures. The increase was much lower than the proportionality of 5 and metal concentration-dependent, specifically, 1.2–1.3 for Cu and from negligible to 2.0 for Mn. For six water extracts of ambient aerosol samples, in which HULIS and metals co-exist, the proportionality ranged from 1.3 to 2.2. This deviation from a linear dependence on initial DTT concentration, plausibly due to metal-DTT binding, impedes assessing and comparing OP of metals and metal-organic mixtures using different implementations of the DTT assay. Considering the different antioxidants concentrations in real human lung fluid, this work raises caution about using the DTT assay to assess metal-containing mixtures, such as ambient aerosol samples.

Nickel is a ubiquitous heavy-metal pollutant in lakes and severely affects aquatic organisms. Aquatic plants are often initially linked to having heavy metal contents and further are proposed as phytoremediation agent to remove heavy metal from water. Although the toxic effects of nickel on aquatic plants are thoroughly explored, the effective investigation to increase Ni tolerance is still in its infancy. The role of rare-earth elements (REEs) in plant resisting heavy-metal pollution has recently received considerable interest. To explore the physiological effects of REEs on Potamogeton crispus under Ni stress, we explored whether or not the additive exposure to low-dose yttrium (Y; 2.5 μM) promotes the polyamine metabolism, antioxidation, and photosynthesis performance of P. crispus under Ni stress values of 0, 50, 100, 150, and 200 μM. Results showed that Y exposure did not influence Ni bioaccumulation in P. crispus. Furthermore, Y exposure alleviated the adverse effects of Ni stress to convergent degrees because Y positively converts putrescine into spermidine and spermine, inhibits oxidative stress, increases the total chlorophyll content, and maximum/potential quantum efficiency of photosystem II. We concluded that low-dose Y can positively regulate polyamine transformation, inhibit oxidative stress, stimulate photosynthesis, and finally promote the resist ability of P. crispus to nickel stress. Thus, REEs have potential to be applied in regulating submerged plant tolerance to aquatic heavy-metal pollution.

In this work PM₂.₅ was collected during winter and summer in a Sardinian village (Gonnostramatza, Italy) highly affected by biomass burning emissions. A multi-technique approach was adopted for the complete PM chemical characterization. The bulk characterization was performed by IC (Ion Chromatography), HPAEC (High-Performance Anion-Exchange Chromatography), TOT (Thermal Optical Transmittance) and ED-XRF (Energy-Dispersive X-Ray Fluorescence) while XPS (X-ray Photoelectron Spectroscopy) was used for the surface characterization. Using levoglucosan as specific tracer of biomass burning emissions, the assessment of the impact of this source was carried out and it represent the major PM source at the investigate site during winter. In winter the average levoglucosan concentration is 2096 ± 324 ng/m³ while during summer its concentration is negligible (18 ± 7 ng/m³). Levoglucosan content in PM₂.₅ during winter is on average 13.7%; it is estimated that 65% of PM₂.₅ is due to wood burning. XPS has been exploited in this work aiming at highlighting possible differences between surface and bulk composition of PM₂.₅. The surface of the particulate matter resulted enriched in carbon compared to the bulk. Among the components of XPS C1s signals recorded on the samples collected during winter, it was found that the signal at 286.5 eV, which is due to the presence of COH, reflects the bulk composition of levoglucosan.

Airborne lower-chlorinated PCBs are vulnerable to metabolization to PCB sulfates through further sulfation of the hydroxylated metabolites (OH-PCBs). However, studies on the toxic effects and mechanisms of PCB sulfates are still very limited. Here, we investigated for the first time the potential endocrine disruption effects of PCB sulfates through estrogen-related receptor γ (ERRγ) in comparison with their OH-PCBs precursors and PCB parent compounds. The binding affinity of thirteen PCBs/OH-PCBs/PCB sulfates was measured by using fluorescence competitive binding assays based on fluorescence polarization (FP). All of the tested chemicals could bind to ERRγ with the Kd (dissociation constant) values ranging from not available (NA) to 3.2 μM 4′–OH–PCB 12 showed the highest binding affinity with Kd value of 3.2 μM, which was comparable to that of a synthetic ERRγ agonist GSK4716. The effects of the thirteen chemicals on the ERRγ transcriptional activity were determined by using the luciferase reporter gene assay. We found the PCBs/OH-PCBs/PCB sulfates acted as agonists for ERRγ, with the lowest observed effective concentration reaching 3 μM. The binding affinity and agonistic activity of PCBs towards ERRγ were both enhanced after hydroxylation, while further sulfation of OH-PCBs decreased the activity instead. Molecular docking simulation showed that OH-PCBs had lower binding energy than the corresponding PCBs and PCB sulfates, indicating that OH-PCBs had higher binding affinity theoretically. In addition, OH-PCBs could form hydrogen bonds with amino acids Glu316 and Arg247 while PCBs and PCB sulfates could not, which might be the main factor impacting the binding affinity and agonistic activity. Overall, ERRγ is a novel target for lower-chlorinated PCBs and their metabolites.

In order to investigate the relationship between air pollution and the respiratory tract microbiota, 114 healthy volunteers aged 18–21 years were selected during the winter heating period in Northeast China; 35 from a lightly polluted region (group A), 40 from a moderately polluted region (group B) and 39 from a heavily polluted region (group C). Microbial genome DNA was extracted from throat swab samples to study the oral flora composition of the volunteers by amplifying and sequencing the V3 regions of prokaryotic 16S rRNA. Lung function tests were also performed. The relative abundance of Bacteroidetes and Fusobacteria were significantly lower and Firmicutes Proteonacteria and Actinobacteria higher in participants from polluted regions. Within bacteria classes, Bacterioida abundance was lower and Clostridia abundance higher in polluted areas, which was also reflected in the order of abundance. In samples from region C, the abundance of Prevotellaceae, Veillonellaceae, Porphyromonadaceae, Fusobacteriaceae Paraprevollaceae and Flavobacteriaceae were lowest among the 3 regions studied, whereas the abundance of Lachnospiraceae and Ruminococcaceae were the highest. From group A to group C, the relative class abundances of Prevotella, Veillonella, Fusobacterium, Camphylobacter and Capnocytophaga Porphyromonas, Peptostreptococcus and Moraxella became lower in polluted areas.Pulmonary function correlated with air pollution and the oropharyngeal microbiota differed within regions of high, medium and low air pollution. Thus, during the winter heating period in Northeast China, the imbalance of the oropharyngeal microbiota might be caused by air pollution and is likely associated with impairment of lung function in young people.
