Analysis of glyphosate degradation in a soil microcosm

Glyphosate (GLP) herbicide leaching into soil can undergo abiotic degradation and two enzymatic oxidative or hydrolytic reactions in both aerobic and anaerobic conditions; biotic oxidation produces aminomethylphosphonic acid (AMPA). Both GLP and AMPA are phytotoxic. A comprehensive GLP degradation reaction network was developed from the literature to account for the above pathways, and fifteen experimental data sets were used to determine the corresponding Michaelis-Menten-Monod (MMM) kinetic parameters. Various sensitivity analyses were designed to assess GLP and AMPA degradation potential against O2 (aq) and carbon (C) availability, pH, and birnessite mineral content, and showed that bacteria oxidized or hydrolyzed up to 98% of GLP and only 9% of AMPA. Lack of a C source limited the GLP cometabolic hydrolytic pathways, which produces non-toxic byproducts and promotes AMPA biodegradation. Low bacterial activity in O2 (aq)-limited conditions or non-neutral pH resulted in GLP accumulation. Birnessite mineral catalyzed fast GLP and AMPA chemodegradation reaching alone efficiencies of 79% and 88%, respectively, regardless of the other variables and produced non-toxic byproducts. Overall, O2 (aq) and birnessite availability played the major roles in determining the partitioning of GLP and its byproducts mass fluxes across the reaction network, while birnessite, C availability, and pH affected GLP and AMPA biodegradation effectiveness.

Several works evaluated the toxicity of pharmaceutical drugs and climate related changes in invertebrates but few explored the combined effects of both stressors, namely considering their mode of action (MoA). Carbamazepine (CBZ) and cetirizine (CTZ) are pharmaceutical drugs detected in the environment and the toxicity derived from the combined effects of these drugs with ocean acidification (OA) is poorly explored. Thus, the present study investigated the biochemical parameters related to an oxidative stress response and the transcription of genes related to the MoA of CBZ (1.0 μg/L) and CTZ (0.6 μg/L) in the clam Ruditapes philippinarum chronically exposed (28 days) to control (7.8) and low (7.5) pH conditions. The results obtained showed that despite the clams accumulated both drugs, at low pH the clams exposed to CTZ decreased drug concentration and BCF values (CTZ uptake: 2.0 ± 0.5 ng/g fresh weight; BCF: 3.8 ± 0.9) in comparison with clams exposed to control pH (CTZ uptake: 2.9 ± 0.3 ng/g fresh weight; BCF: 5.5 ± 0.6). No oxidative stress was induced by the exposure to CBZ or CTZ at each pH level, but the transcription of several genes related with the MoA (neurotransmission, immunity and biomineralization) was altered by low pH, drug exposure and the combination of both stressors. At both pH conditions, CBZ increased the transcription of GABA receptor gene (neurotransmission) and CTZ led to a decrease of Perlucin gene (biomineralization) transcription. The transcription of MyD88 gene (immunity) decreased at low pH (7.5) combined with drug exposure (CBZ or CTZ). Thus, it was highlighted that the interaction of drug exposure and low pH conditions can change bivalves’ sensitivity to drugs or alter drugs toxicity.

This work investigated the role of natural organic matter (NOM) in the environmental processes of silver nanoparticles (AgNP) and the uptake and accumulation of AgNP in wheat. Different NOMs (Suwannee River humic acids [SRHA], fulvic acid [FA]) and Ag elements (Ag⁽⁰⁾ and Ag⁺) were incubated in a hydroponic media for 15 days. The results showed that the NOM (10 mg C L⁻¹) altered the dissolution, stabilization, uptake and accumulation of AgNP. The dissolution of AgNP declined in the presence of NOM. Compared with FA, the dissolved Ag⁺ decreased much more from 0.30 mg L⁻¹ to 0.10 mg L⁻¹ in the presence of SRHA. The fluorescence quenching results indicated that SRHA exhibited stronger binding to Ag⁺ than that of FA, and the quenching constants Ksv were 0.1309 (SRHA) and 0.0074 (FA), respectively. CO, CH, COC, and MeOH were involved in the interaction between NOM and AgNP. The NOM decreased the accumulated content of Ag in wheat. Hence, NOM alleviated the inhibition of AgNP to wheat growth. SRHA reduced the Ag content of wheat roots approximately 3-fold. These results clearly indicated the importance of NOM on altering the behavior, fate and toxicity of AgNP in an environment.

The neonicotinoid insecticide imidacloprid is used in Bangladesh for a variety of crop protection purposes. Imidacloprid may contaminate aquatic ecosystems via spray drift, surface runoff and ground water leaching. The present study aimed at assessing the fate and effects of imidacloprid on structural (phytoplankton, zooplankton, macroinvertebrates and periphyton) and functional (organic matter decomposition) endpoints of freshwater, sub-tropical ecosystems in Bangladesh. Imidacloprid was applied weekly to 16 freshwater microcosms (PVC tanks containing 400 L de-chlorinated tap water) at nominal concentrations of 0, 30, 300, 3000 ng/L over a period of 4 weeks. Results indicated that imidacloprid concentrations from the microcosm water column declined rapidly. Univariate and multivariate analysis showed significant effects of imidacloprid on the zooplankton and macroinvertebrate community, some individual phytoplankton taxa, and water quality variables (i.e. DO, alkalinity, ammonia and nitrate), with Cloeon sp., Diaptomus sp. and Keratella sp. being the most affected species, i.e. showing lower abundance values in all treatments compared to the control. The observed high sensitivity of Cloeon sp. and Diaptomus sp. was confirmed by the results of single species tests. No significant effects were observed on the species composition of the phytoplankton, periphyton biomass and organic matter decomposition for any of the sampling days. Our study indicates that (sub-)tropical aquatic ecosystems can be much more sensitive to imidacloprid compared to temperate ones.

The isotopic fractionation could contribute to understanding the Cd accumulation mechanisms in plant species. However, there are few of systematical investigations with regards to the Cd isotope fractionation in hyperaccumulator plants. The Cd tolerant Ricinus communis and hyperaccumulator Solanum nigrum were cultivated in nutrient solutions with varying Cd and EDTA concentrations. Cd isotope ratios were determined in the solution, root, stem and leaf. The two investigated plants were systematically enriched in light isotopes relative to their solutions (Δ114/110Cdplant-solution = −0.64‰ to −0.29‰ for R. communis and −0.84‰ to −0.31‰ for S. nigrum). Cd isotopes were markedly fractionated among the plant tissues. For both plant species, an enrichment in light Cd isotopes from solution to root was noted, followed by a slight depletion in light Cd isotopes from root to shoot. Noticeably, the chelation process has caused lighter Cd isotope enrichment in the root of R. communis and S. nigrum. Further, the good fits between △114/110Cdroot-plant and ln Froot (or between △114/110Cdshoot-plant and ln Fshoot) indicate that Cd isotopic signatures can be used to study Cd transportation during the metabolic process of plants. This study suggests that knowledge of the Cd isotope ratios could also provide new tool for identifying the Cd-avoiding crop cultivars.

Organophosphate flame retardants (OPFRs) have been commonly observed in indoor dust, food, and drinking water in China, but little is known about their exposure levels or factors leading to exposure in Chinese children. In this study, we measured eight metabolites of OPFRs (mOPFRs) in 227 urine samples collected from 0- to 5-year-old children in China. The high detection rates of mOPFRs (60%–100%) in the collected urine samples demonstrated the widespread exposure of this population to OPFRs. The median concentrations indicated that bis(2-chloroethyl)phosphate (BCEP, 0.85 ng/mL) and diphenyl phosphate (DPHP, 0.27 ng/mL) were the dominant chlorinated mOPFRs and nonchlorinated mOPFRs, respectively. Interestingly, the median urinary levels of bis(1-chloro-2-propyl)phosphate (BCIPP, 6.48 ng/mL) and bis(2-butoxyethyl)phosphate (BBOEP, 0.31 ng/mL) in inpatient infants were one order of magnitude higher (p < 0.01) than those observed in outpatient infants. For home-stay participants, furthermore, infants (0–1 year) had the highest median levels of BCIPP (0.72 ng/mL) and dibutyl phosphate (DBP, 0.14 ng/mL) among the three age groups (i.e., 0–1, >1–3, and >3–5 years), and significantly (p < 0.05) negative age-related relationships were found for both urinary mOPFRs. Two set of data on estimated daily intakes (EDIs) were calculated based on the fraction of OPFR excreted as the corresponding mOPFR (FUE) in human liver microsomes (EDIHLM) and S9 fraction (EDIS₉) system, respectively. In general, children have relatively high EDIs of tris(2-chloroethyl)phosphate (TCEP: EDIHLM = 485 ng/kg bw/day, EDIS₉ = 261 ng/kg bw/day). Furthermore, 17% or 21% of inpatient infants had EDIs that exceeded the reference dose, whereas this value was reduced to 13% in outpatient infants; and this value decreased with age among all home-stay children (0–5 years). Our results indicated that inpatient and home-stay infants had a higher potential risk of OPFR exposure. To our knowledge, this is the first study to identify the elevated urinary levels of mOPFRs in inpatients.

Sleep disorders may pose a risk to workers in the workplace. We aimed to investigate the associations between metal fume fine particulate matter (PM2.5) and sleep quality in workers. We assessed the effects of personal exposure to metal fume PM2.5 on lung functions, urinary biomarkers, and sleep quality in shipyard welding workers. In total, 96 welding workers and 54 office workers were recruited in the present study; office workers were exposed to 82.1 ± 94.1 μg/m³ PM2.5 and welding workers were exposed to 2166.5 ± 3149.1 μg/m³. Welding workers had significantly lower levels of FEV25-75 than office workers (p < 0.05). An increase in 1 μg/m³ PM2.5 was associated with a decrease of 0.003 ng/mL in urinary serotonin (95% CI = −0.007–0.000, p < 0.05) in all workers and with a decrease of 0.001 ng/mL in serotonin (95% CI = −0.004–0.002, p < 0.05) in welding workers, but these were not observed in office workers. There was no significant association of PM2.5 with urinary cortisol observed in any workers. Urinary serotonin was associated with urinary Cu, Mn, Co, Ni, Cd, and Pb. Urinary cortisol was associated with Cu, Mn, Co, Ni, Cd, and Pb. Sixteen subjects were randomly selected from each of the office and welding workers for personal monitoring of sleep quality using a wearable device. We observed that welding workers had greater awake times than did office workers (p < 0.05). Our study observed that exposure to heavy metals in metal fume PM2.5 may disrupt sleep quality in welding workers.

Organic contaminants such as pharmaceuticals, personal care products (PPCPs) and other emerging contaminants (ECs) are known to persist in the aquatic environment and many are indicated as endocrine, epigenetic, or other toxicants. Typically, the study of PPCPs/ECs in the aquatic environment is limited to their occurrence dissolved in river water. In this study, accumulation and spatial distribution of thirteen PPCPs/ECs were assessed in aquatic sediment (n = 23), periphyton (biofilm, n = 8), plants Callitriche sp. (n = 8) and Potamogeton sp. (n = 7) as well as amphipod crustaceans (Gammarus pulex, n = 10) and aquatic snails (Bithynia tentaculata, n = 9). All samples (n = 65) were collected from the Hogsmill, Blackwater and Bourne Rivers in southern England. Targeted PPCPs/ECs included pharmaceuticals, plasticisers, perfluorinated compounds, illicit drugs and metabolites. Extraction from solid matrices occurred using ultrasonic-assisted extraction followed by an in-house validated method for solid-phase extraction and subsequent liquid-chromatography tandem mass-spectrometry. Field-derived bioconcentration-factors and biota-sediment accumulation-factors were determined for all studied biota. Residues of studied contaminants were found in all sediment and biota. Concentrations of contaminants were generally higher in biota than sediment. Evidence suggests that the studied aquatic plants may effectively degrade bisphenol-A into its main transformation product hydroxyacetophenone, potentially mediated by cytochrome p450 and internalisation of contaminants into the cellular vacuole. A positive association between both hydrophobicity and PFC chain length and contaminant accumulation was observed in this work. Only PFCs, plasticisers and HAP were classified as either ‘bioaccumulative’ or ‘very bioaccumulative’ using BCF criteria established by guidelines of four governments. Contaminants appeared to be differentially bioaccumulative in biota, indicating there may be a need for a species-specific BCF/BSAF classification system. These data form a detailed accounting of PPCP/EC fate and distribution in the aquatic environment highlighting accumulation at lower trophic levels, a potential source for higher organisms.

Drinking water safety is continuously threatened by the emergence of numerous toxic organic pollutants (TOPs) in environmental waters. In this study, an approach integrating in vitro bioassays and chemical analyses was performed to explore toxicological profiles of representative source and drinking water from waterworks of the Yangtze River (Yz), Taihu Lake (Th), and the Huaihe River (Hh) basins in eastern China. Overall, 34 of 96 TOPs were detected in all water samples, with higher concentrations in both source and drinking water samples of Hh, and pollutant profiles also differed across different river basins. Non-specific bioassays indicated that source water samples of Hh waterworks showed higher genotoxicity and mutagenicity than samples of Yz and Th. An EROD assay demonstrated dioxin-like toxicity which was detected in 5 of 7 source water samples, with toxin concentration levels ranging from 62.40 to 115.51 picograms TCDD equivalents per liter of water (eq./L). PAHs and PCBs were not the main contributors to observed dioxin-like toxicity in detected samples. All source water samples induced estrogenic activities of 8.00–129.00 nanograms 17β-estradiol eq./L, and estrogens, including 17α-ethinylestradiol and estriol, contributed 40.38–84.15% of the observed activities in examined samples. While drinking water treatments efficiently removed TOPs and their toxic effects, and estrogenic activity was still observed in drinking water samples of Hh. Altogether, this study indicated that the representative source water in eastern China, especially that found in Hh, may negatively affect human health, a finding that demonstrates an urgent requirement for advanced drinking water treatments.

Heavy metal contamination of source water frequently occurred in developing countries as a result of accidents. To address the problems, most of the previous studies have focused on engineering countermeasures. In this study, we investigated the effects of heavy metals, particularly copper, on the development of antibiotic resistance by establishing a copper shock loading test. Results revealed that co-selection occurred rapidly within 6 h. Copper, at the levels of 10 and 100 mg/L, significantly increased bacterial resistance to the antibiotics tested, including rifampin, erythromycin, kanamycin, and a few others. A total of 117 antimicrobial-resistance genes were detected from 12 types of genes, and the relative abundance of most genes (particularly mobile genetic elements intⅠand transposons) was markedly enriched by at least one fold. Furthermore, the copper shock loading altered the bacterial community. Numerous heavy metal and antibiotic resistant strains were screened out and enriched. These strains are expected to enhance the overall level of resistance. More noticeably, the majority of the co-selected antibiotic resistance could sustain for at least 20 h in the absence of copper and antimicrobial drugs. Resistance to vancomycin, erythromycin and lincomycin even could remain for 7 days. The prominent selection pressure by the copper shock loading implies that a real accident most likely poses similar impacts on the water environment. An accidental release of heavy metals would not only cause harm to the ecological environment, but also contribute to the development of bacterial antibiotic resistance. Broader concerns should be raised about the biological risks caused by sudden releases of pollutants by accidents.
