Psychoactive pharmaceuticals in aquatic systems: A comparative assessment of environmental monitoring approaches for water and fish

Environmental monitoring and surveillance studies of pharmaceuticals routinely examine occurrence of substances without current information on human consumption patterns. We selected 10 streams with diverse annual flows and differentially influenced by population densities to examine surface water occurrence and fish accumulation of select psychoactive medicines, for which consumption is increasing in the Czech Republic. We then tested whether passive sampling can provide a useful surrogate for exposure to these substances through grab sampling, body burdens of young of year fish, and tissue specific accumulation of these psychoactive contaminants. We identified a statistically significant (p < 0.05) relationship between ambient grab samples and passive samplers in these streams when psychoactive contaminants were commonly quantitated by targeted liquid chromatography with tandem mass spectrometry, though we did not observe relationships between passive samplers and tissue specific pharmaceutical accumulation. We further observed smaller lotic systems with elevated contamination when municipal effluent discharges from more highly populated cities contributed a greater extent of instream flows. These findings identify the importance of understanding age and species specific differences in fish uptake, internal disposition, metabolism and elimination of psychoactive drugs across surface water quality gradients.

Hydroquinone (HQ), one of the main metabolites of benzene, is a well-known human leukemogen. However, the specific mechanism of how benzene or HQ contributes to the development of leukemia is unknown. In a previous study, we demonstrated the upregulation of DNA methyltransferase (DNMT) expression in HQ-induced malignant transformed TK6 (HQ-TK6) cells. Here, we investigated whether a regulatory loop between the long noncoding RNA FAS-AS1 and DNMT3b exists in HQ-TK6 cells and benzene-exposed workers. We found that the expression of FAS-AS1 was downregulated in HQ-TK6 cells and workers exposed to benzene longer than 1.5 years via histone acetylation, and FAS-AS1 expression was negatively correlated with the time of benzene exposure. Restoration of FAS-AS1 in HQ-TK6 cells promoted apoptosis and inhibited tumorigenicity in female nude mice. Interestingly, treatment with a DNMT inhibitor (5-aza-2-deoxycytidine), histone deacetylase inhibitor (trichostatin A), or DNMT3b knockout led to increased FAS-AS1 through increased H3K27ac protein expression in HQ-TK6 cells, and DNMT3b knockout decreased H3K27ac and DNMT3b enrichment to the FAS-AS1 promoter region, which suggested that DNMT3b and/or histone acetylation involve FAS-AS1 expression. Importantly, restoration of FAS-AS1 resulted in reduced expression of DNMT3b and SIRT1 and increased expression of FAS in both HQ-TK6 cells and xenograft tissues. Moreover, the average DNMT3b expression in 17 paired workers exposed to benzene within 1.5 years was decreased, but that of the remaining 103 paired workers with longer exposure times was increased. Conversely, DNMT3b was negatively correlated with FAS-AS1 expression. Both FAS-AS1 and DNMT3b influenced the enrichment of H3K27ac in the FAS promoter region by regulating the expression of SIRT1, consequently upregulating FAS expression. Taken together, these observations demonstrate crosstalk between FAS-AS1 and DNMT3b via a mutual inhibition loop and indicate a new mechanism by which FAS-AS1 regulates the expression of FAS in benzene-related carcinogenesis.

Polycyclic Aromatic Hydrocarbons are strongly associated with agricultural, residential, transportation, and industrial activities. This study determined by GC-MS the concentration of 15 PAHs in soil and sediments at different sites from the Awotan-Asunle dumpsite area in the Southwestern region of Nigeria, which is one of the largest dumpsites in Africa. The sources of contamination, toxicity and associated risks for human health were also evaluated. Total PAHs concentrations were from 489 to 5616 μg kg⁻¹, and 642–2159 μg kg⁻¹, for soil and sediment, respectively. For soils, the highest values were observed for indeno[1,2,3-c,d]pyrene, coronene, and phenanthrene, while for sediments, the most abundant species were pyrene, fluoranthene and phenanthrene. Diagnostic ratios were used to determine the sources of PAHs and suggested that the compounds were mainly emitted from non-traffic sources. The total BaP-TEQ and BaP-MEQ for soils did not exceed the value recommended by the Canadian guideline since the country does not present guidelines. The analysis of incremental lifetime cancer risk was high mostly for dermal and ingestion exposures in the population. This study might provide valuable information regarding exposure to PAHs in soils of a Nigerian community.

The aim of the research was to study short-term variations in concentrations of 17 elements in two widely distributed natural plant species (couch grass and plantain) and in the rhizosphere soil of the plants. The plant and soil samples were collected in a field from a small site over a daytime. In the course of the day, the variations of the total amounts of C, N, and H in the rhizosphere soil were rather marked and different for the soils taken from roots of plantain and couch grass. The concentrations of some other elements in the rhizosphere soil of the plants varied in a similar way. The short-term variations of element concentrations in roots and leaves of the plants were also rather large. In many cases, a decrease of element concentration in roots correlated with an increase of its concentration in leaves. Although couch grass and plantain were collected simultaneously and from the same site, mean concentrations of many elements in the two plant species were statistically significantly different. This may be result of the fact that the plants belong to different clades. The differences between concentrations of most part of elements in roots and leaves of the plants were also statistically significant. The concentrations of many trace elements were higher in roots than in leaves, while the concentrations of essential plant nutrients were often higher in leaves compared to roots. The distribution of elements between different plant parts were not the same in couch grass and plantain.

Microplastic (MPs) pollution in the aquatic and terrestrial environments has caught many attentions in the scientific literatures. Currently, no information is available about MPs pollution in Caspian Sea, the largest lake in the world. This study indicates the first report on the MPs pollution in the sediments of the southern Caspian coastal zones, northern Iran. Density separation method was conducted on 17 surficial sediments. The combination of observation techniques including SEM-EDS analysis, polarized light microscopy and Raman micro-spectroscopy were used to identify MPs. The abundance and size of microplastics in the samples ranged between 25 and 330 items/kg and 250–500 μm, respectively. Fibers constituted the most common MPs shape and polystyrene (PS) and polyethylene (PE) were major polymer types in the samples. The distribution of MPs in the study area reflected a patchy and irregular spatial pattern implying that the higher MPs concentration are near mouth of permanent rivers and in the regions with higher level of the fishing and tourism activities. The results showed the wide occurrence of MPs in the sediments of the world’s largest lake which extend the knowledge on MPs pollution in the marine system. We also recommend further research on microplastics in different compartments of Caspian Sea to inform policy discussions and the development of appropriate management responses.

Evolution of the microbial community structure in crude oil contaminated marine sediments was assessed under aerobic biodegradation during wet (18 °C) and dry (28 °C) seasons experiments, to account for seasonal variations in nutrients and temperature, under biostimulation and natural attenuation conditions. NMDS showed significant variation in the microbial communities between the wet and the dry season experiments, and between the biostimulation and the natural attenuation treatments in the dry season microcosms. No significant variation in the microbial community and oil biodegradation was observed during the wet season experiments due to high background nitrogen levels eliminating the effect of biostimulation. Larger variations were observed in the dry season experiments and were correlated to enhanced alkanes removal in the biostimulated microcosms, where Alphaproteobacteria dominated the total microbial community by the end of biodegradation (54%). Many hydrocarbonoclastic bacterial genera showed successive dominance during the operation affecting the ultimate performance of the microcosms.

New analytical methods are needed to efficiently measure the growing list of priority pharmaceuticals in environmental samples. In this regard, a rapid, sensitive, and robust method was developed for quantitation of 168 pharmaceuticals and pharmaceutical metabolites using solid-phase extraction (SPE) and liquid chromatography with tandem mass spectrometry. The extraction protocol and instrumental efficiency were specifically addressed to increase analytical workload and throughput. The optimized protocols, which are five times more efficient than US EPA Method 1694, enabled analyte recoveries that ranged from 77% to 117% for 162 analytes with method quantitation limits (MQLs) as low as 0.1 ng L⁻¹. To verify the suitability of the improved analytical method for environmental samples, 24-h composite samples of raw wastewater and wastewater effluent, along with downstream surface water, were analyzed. Overall, 143/168 target compounds were identified in at least one of the samples, and 130/168 analytes were present at concentrations above their MQLs. The total mass concentration of the measured analytes decreased by 93% during wastewater treatment. The analyte concentrations in the wastewater effluent were comparable to those measured in surface water 1 km downstream of the wastewater discharge point. Ultimately, the comprehensive method will serve as an important tool to inform the occurrence, fate, transport, and toxicity of a large suite of priority pharmaceuticals and pharmaceutical metabolites in natural and engineered systems.

The individual effects of biological constituents of particulate matter (PM) such as fungal spores, on lung function in children are not well known. This study investigated the seasonal short-term effect of daily variation in Alternaria and Cladosporium fungal spores on lung function in schoolchildren.This panel study evaluated 313 schoolchildren in informal settlements of the Western Cape of South Africa, exposed to spores of two commonly encountered fungi, Alternaria and Cladosporium species. The children provided forced-expiratory volume in 1-s (FEV₁) and peak-expiratory flow (PEF) measurements thrice daily for two consecutive school-weeks in summer and winter. Daily PM₁₀ levels, from a stationary ambient air quality monitor and fungal spore levels using spore traps were measured in each study area throughout the year. The effects of Alternaria and Cladosporium spores, on lung function were analysed for lag periods up to five-days, adjusting-for PM₁₀, other pollen exposures, study area, and other host and meteorological factors. Same-day exposure-response curves were computed for both fungal species.There was more variability in Alternaria spores level with noticeable peaks in summer. There were consistent lag-effects for Alternaria on PEF compared to Cladosporium, with the largest PEF deficit observed in winter (mean deficit: 13.78 L/min, 95%CI: 24.34 to −3.23 L/min) per 10spores/m³ increase in Alternaria spores on lag day-2. Although there were no observable lag-effects for Alternaria and Cladosporium on FEV₁, same-day effects of Cladosporium spores on FEV₁ was present across both seasons. Threshold effects of Alternaria on both PEF and FEV₁ deficits were apparent at levels of 100 spores/m³, but could not be explored for Cladosporium beyond the levels observed during the study.The study provides evidence for the independent effects of daily exposure to ambient fungal spores of Alternaria and Cladosporium on lung function deficits, more especially in winter for PEF.

Interactions between the intestine and the liver, the so-called ‘gut-liver axis’, play a crucial role in the onset of hepatic steatosis and non-alcoholic fatty liver disease. However, not much is known about the impact of environmental pollutants on the gut-liver axis and consequent hepatic steatosis. Bisphenol A (BPA), a widely used plasticiser, is an important environmental contaminant that affects gut microbiota. We hypothesised that BPA induces hepatic steatosis by promoting gut microbiota dysbiosis and activating the gut-liver axis. In this study, male CD-1 mice were fed with diet containing BPA (50 μg/kg body weight/day) for 24 weeks. Dietary exposure to BPA increased lipid contents and fat accumulation in the liver. Analysis of 16 S rRNA gene sequencing revealed that the diversity of gut microbiota reduced and the composition of gut microbiota was altered in the BPA-fed mice. Further, the abundance of Proteobacteria, a marker of dysbacteria, increased, whereas the abundance of Akkermansia, a gut microbe associated with increased gut barrier function and reduced inflammation, markedly decreased. Expression levels of intestinal tight junction proteins (zona occludens-1 and occludin) also decreased drastically, leading to increased intestinal permeability and elevated levels of endotoxins. Furthermore, BPA up-regulated the expression of Toll-like receptor 4 (TLR4) and phosphorylation of nuclear factor-kappa B (NF-κB) in the liver and increased the production of inflammatory cytokines, including interleukin-1β, interleukin-18, tumour necrosis factor-α, and interleukin-6. Take together, our work indicated that dietary intake of BPA induced hepatic steatosis, and this was closely related to dysbiosis of gut microbiota, elevated endotoxin levels, and increased liver inflammation through the TLR4/NF-κB pathway.

Air pollution is a major public health challenge in the highly urbanized megacities of China. However, knowledge on exposure to ambient unregulated air pollutants such as black carbon (BC) and ultrafine particles (UFP) among the Chinese population, especially among urban high school students who may have highly variable time-activity patterns, is scarce. To address this, the personal exposures to BC and UFP of high school students (aged 17 to 18) in Chengdu, China were measured at 1-min intervals via portable samplers. Monitoring lasted for 2 consecutive 24-h periods with days classified as “school days” or “non-school days”. Time-activity diaries and measurements were combined to explore spatial, temporal, and behavioral factors that contribute to different exposure profiles. The overall geometric means of BC and UFP were 3.60 μg/m³ and 1.83 × 10⁴p/cm³, respectively with notable spatiotemporal variation in exposures observed. In general, the household and transport microenvironments were the predominant contributors to total BC (74.5%) and UFP (36.5%) exposure. However, the outdoor public microenvironment was found to have significantly higher overall average levels of BC than the household and transport microenvironments (p < 0.001) while also presenting the greatest exposure dose intensity (EDI – a measure of exposure in a microenvironment in proportion to time spent in that environment) of 4.79. The largest overall average level of UFP occurred in the indoor public microenvironment followed by transport. The outdoor public microenvironment also presented the greatest EDI of UFP (4.17). This study shows notable spatiotemporal variety in exposure patterns and will inform future exposure and population health studies. The high EDI outdoors may mean that health positive activities, such as exercise, may be being undermined by ambient pollution.
