Neuroimaging signatures of brain plasticity in adults with prenatal exposure to polychlorinated biphenyls: Altered functional connectivity on functional MRI

Prenatal exposure to polychlorinated biphenyls (PCBs), persistent organic pollutants in food chains and environment, exerts negative effects on children's cognitive function. To study the long-term effects, we examined cognitive functions in the male children of women with substantial PCB exposure in Taiwan during 1978–1979 and investigated neural basis of cognitive function changes through structural magnetic resonance imaging (MRI) and functional MRI (fMRI), which included resting-state and task-activated fMRI with two paradigms: a 2-back task and a picture rotation task. Ten men aged 30.0 ± 3.7 years with prenatal exposure to PCBs and 11 unexposed controls aged 28.1 ± 3.1 years participated. Both groups had similar cognitive phenotypes and behavioral results. Structural MRI analysis results showed that the PCB group had increased cortical thickness over the right inferior parietal lobule. In the resting-state study, the PCB group showed alterations in the default mode network. During the tasks, the PCB group showed decreased task-induced deactivation signals in cognition–associated brain areas during the 2-back task but enhanced deactivations during the picture rotation task. This study demonstrated altered structural MRI as well as resting and task-related fMRI in men with prenatal PCB exposure, suggesting altered brain plasticity and compensatory neuropsychological performance.

There is increasing concern about phenolic environmental estrogens (PEEs) in river systems, especially in economically developed regions, because of their potential to impact ecological systems. We studied the distribution of, ecological risks from, and factors that influenced PEEs in the sediments from the Duliujian River in the Beijing-Tianjin-Hebei Urban Agglomeration and the Pearl River in Guangdong Province in China. The three target PEEs, nonyl phenol (4-NP), octyl phenol (4-t-OP), and bisphenol A (BPA), were detected in the sediments at concentrations ranging from 204.4 to 12604.3, 32.6 to 297.3, and from 12.8 to 298.4 ng g−1 in the Pearl River, and from 153.5 to 3614.9, 90.7 to 990.0, and 83.5–913.3 ng g−1 in the Duliujian River, respectively. The PEE concentrations were significantly and positively correlated with total organic carbon in the river sediments (p < 0.1). Urbanization influenced the distribution of PEEs and applications and discharges of PEEs were associated with large populations and industries. Rainfall and wastewater discharge patterns also influenced how PEEs were distributed in river sediments. The potential ecological risks from 4-NP, 4-t-OP, and BPA in these two rivers were high. Measures should be put in place to control the transport and storage of these compounds in river systems.

Nearshore environments are a critical transitional zone that connects the marine and terrestrial/freshwater ecosystems. The release of anthropogenic chemicals into nearshore ecosystems pose a human and environmental health risk. We investigated the microbial diversity, abundance and function in metal-contaminated sediments collected from the Rongjiang, Hanjiang and Lianjiang River estuaries and adjacent coastal areas using high throughput sequencing. The concentration of nutrients (NO3-N, NO2-N, NH4-N, PO4-P) and metal (Cu, Zn, Cd, Pb, As, Hg) contaminants were higher at the mouth of the rivers compared to the coastal lines, and this was confirmed using cluster analysis. Estimates obtained using geoaccumulation index showed that about 38.9% of the sites were contaminated with Pb and the pollution load index showed that sediment from the mouth of Hanjiang River Estuary was moderately polluted with metals. In the nearshore sediment samples collected, Proteobacteria, Bacteroidetes, Planctomycetes, Chloroflexi, Acidobacteria were the dominant phylum with relative abundances of 46.6%, 8.05%, 6.47%, 5.26%, and 4.59%, respectively. There was no significant correlation between environmental variables and microbial abundance and diversity except for total organic carbon (TOC) (diversity; r = 0.569, p < 0.05) and Cr (diversity; r = 0.581, p < 0.05). At phyla level, Nitrospirae had a significant negative correlation with all metals except Cr, while OD1 had a significant positive correlation with all the metals. Overall, changes in nearshore sediment microbial communities by environmental factors were observed, and these may affect biogeochemical cycling.

Nanoparticles (NPs), heavy metal and natural organic matter (NOM) may simultaneously exist in the aquatic environment, where they will affect the behavior of each other and may enhance their toxicities. Studies on the influences of interactions between NOM and heavy metal ions on the behavior of NPs are scarce. In this study, combined effects of Pb²⁺ and HA on the aggregation behavior of TiO₂ NPs in water environment were investigated by Dynamic light scattering (DLS) and Nanoparticle tracking analysis (NTA). The results illustrated that interactions between Pb²⁺ and HA could case the aggregation of TiO₂ NPs obviously. The concurrence of Pb²⁺ and HA resulted in decreased critical coagulation concentration (CCC) and increased attachment efficiencies. Meanwhile, we found that the addition sequences of HA and heavy metal clearly influenced the aggregation kinetics of TiO₂ NPs. At different addition sequences, the complex reaction between Pb²⁺ and HA changed the surface charge of TiO₂ NPs, and caused the different aggregation behavior which depended on the complex locations and complex sites. Furthermore, the excitation-emission-matrix (EEM) fluorescence spectra was used to verify the significant effects of the complex interactions between Pb²⁺ and HA on the aggregation of TiO₂ NPs. Our results would be significant for interpreting TiO₂ behavior in the complicated water system.The complexation between Pb²⁺ and HA promoted the aggregation of TiO₂ NPs, meanwhile, complex locations and complex sites played an important role.

Cadmium (Cd), a ubiquitous environmental pollutant, is known to impair placental development. However, the underlying mechanisms remain unclear. The present study used in vivo and in vitro models to investigate the effects of Cd on apoptosis and autophagy in placental trophoblasts and its mechanism. Pregnant mice were exposed to CdCl₂ (4.5 mg/kg) on gestational day (GD) 9. Human JEG-3 cells were exposed to CdCl₂ (0–40 μM) for different time points. Gestational Cd exposure obviously lowered the weight and diameter of mouse placentas. Number of TUNEL-positive cells was markedly elevated in Cd-administered mouse placentas and JEG-3 cells. Correspondingly, Cd significantly up-regulated cleaved caspase-3 protein level, a key indicator of apoptosis, in murine placentas and JEG-3 cells. Simultaneously, Cd also triggered autophagy, as determined by an elevation of LC3B-II and p62 protein, and accumulation of LC3-positive puncta, in placental trophoblasts. Chloroquine an autophagy inhibitor, obviously aggravated Cd-induced apoptosis in JEG-3 cells. By contrast, rapamycin, a specific autophagy inducer, significantly alleviated Cd-triggered apoptosis in JEG-3 cells. Mechanistically, autophagy inhibited Cd-induced apoptosis mainly via degrading caspase-9. Co-localizations of p62, a classical autophagic receptor, and caspase-9 were observed in Cd-stimulated human JEG-3 cells. Moreover, p62 siRNAs pretreatment markedly blocked the degradation of caspase 9 proteins via Cd-activated autophagy in JEG-3 cells. Collectively, our data suggest that activation of autophagy inhibits Cd-induced apoptosis via p62-mediated caspase-9 degradation in placental trophoblasts. These findings provide a new mechanistic insight into Cd-induced impairments of placental and fetal development.

Anthropogenic activities including coastal industries, urbanization, extensive agriculture and aquaculture as well as their cumulative impacts represent major sources of perturbation of marine coastal systems. Macrobenthic communities are useful ecological indicators for monitoring the health status of marine environments (or polluted environments). The present study reports, for the first time, the response of benthic macrofauna sampled during two years survey (2015–2016) to multiple anthropogenic pressures on the coastal zone south of Sfax (Tunisia). A total of 12 stations were monitored seasonally at locations downstream from the main potential sources of disturbance. 106 macrobenthos taxa, belonging to six animal phyla and 70 families, were identified with a dominance of polychaetes (42%), crustaceans (35%) and molluscs (18%). We used an ANOVA test and cluster analysis to identify spatial gradient linked to environmental and anthropogenic factors, including depth, sedimentary texture and anthropogenic activities (i.e. phosphogypsum discharges).The macrofauna present lowest species number and abundance on stations undergoing anthropogenic inputs, which are extremely polluted by heavy metals (Cd, Cu, F and N) and excess of organic matter. Univariate parameters reveal a general trend of increasing species diversity with increasing distance from the pollution source. The polluted stations are strongly dominated by carnivores, and selective deposit feeders, and more closely linked to the availability of trophic resources than to anthropogenic constraints. The seasonal changes in macrobenthic abundance, diversity indices and community structure are mainly linked to the biological cycle (e.g. recruitment events) of the dominant species. Biotic indices (AMBI and BO2A) classified the coastal zone south of Sfax as moderate and good ecological status. This study suggests that initiating a long-term monitoring programme would improve our understanding of the temporal changes of macrobenthic communities of this ecosystem, contributing to the assessment of effective management and conservation measures in this disturbed area.

In this study, various remote sensing data, modeling data and emission inventories were integrated to analyze the tempo-spatial distribution of biomass burning in mainland Southeast Asia and its effects on the local ambient air quality from 2001 to 2016. Land cover changes have been considered in dividing the biomass burning into four types: forest fires, shrubland fires, crop residue burning and other fires. The results show that the monthly average number of fire spots peaked at 34,512 in March and that the monthly variation followed a seasonal pattern, which was closely related to precipitation and farming activities. The four types of biomass burning fires presented different tempo-spatial distributions. Moreover, the monthly Aerosol Optical Depth (AOD), concentration of particulate matter with a diameter less than 2.5 μm (PM₂.₅) and carbon monoxide (CO) total column also peaked in March with values of 0.62, 45 μg/m³ and 3.25 × 10¹⁸ molecules/cm², respectively. There are significant correlations between the monthly means of AOD (r = 0.74, P < 0.001), PM₂.₅ concentration (r = 0.88, P < 0.001), and CO total column (r = 0.82, P < 0.001) and the number of fire spots in the fire season. We used Positive Matrix Factorization (PMF) model to resolve the sources of PM₂.₅ into 3 factors. The result indicated that the largest contribution (48%) to annual average concentration of PM₂.₅ was from Factor 1 (dominated by biomass burning), followed by 27% from Factor 3 (dominated by anthropogenic emission), and 25% from Factor 2 (long-range transport/local nature source). The annually anthropogenic emission of CO and PM₂.₅ from 2001 to 2012 and the monthly emission from the Emission Database for Global Atmosphere Research (EDGAR) were consistent with PMF analysis and further prove that biomass burning is the dominant cause of the variation in the local air quality in mainland Southeast Asia.

Environmental reservoirs of antibiotic resistance (AR) are a growing concern that are gathering more attention as potential sources for human infection. Carbapenem-resistant Enterobacteriaceae (CRE) are extremely dangerous, as carbapenems are often drugs of last resort that are used to treat multi-drug resistant infections. Among the genes capable of conferring carbapenem resistance to bacteria, the most transferrable are those that produce carbapenemase, an enzyme that hydrolyzes carbapenems and other β-lactam antibiotics. The goal of this review was to comprehensively identify global environmental reservoirs of carbapenemase-producing genes, as well as identify potential routes of transmission to humans. The genes of interest were Klebsiella pneumoniae carbapenemase (KPC), New Delhi Metallo-β-lactamase (NDM), Oxacillinase-48-type carbapenemases (OXA-48), and Verona Integron-Mediated Metallo-β-lactamase (VIM). Carbapenemase genes have been reported in the environment on almost every continent. Hospital and municipal wastewater, drinking water, natural waterways, sediments, recreational waters, companion animals, wildlife, agricultural environments, food animals, and retail food products were identified as current reservoirs of carbapenemase-producing bacteria and genes. Humans have been recorded as carrying CRE, without recent admittance to a hospital or long-term care facility in France, Egypt, and China. CRE infections from the environment have been reported in patients in Montpellier, France and Cairo, Egypt. This review demonstrates the need for 1) comprehensive monitoring of AR not only in waterways, but also other types of environmental matrices, such as aerosol, dusts, periphyton, and surfaces in indoor environments; and 2) action to reduce the prevalence and mitigate the effects of these potentially deadly resistance genes. In order to develop an accurate quantitative model for environmental dimensions of AR, longitudinal sampling and quantification of AR genes and bacteria are needed, using a One Health approach.

A recent data set for 22 poly- and per-fluorinated compounds (PFASs) in Ponar grab samples of surface sediments and cores from the Great Lakes of North America was examined for concentrations, loads, correlations with geographical coordinates and depth (time), and for sources. Correlations were determined by multivariate regression analyses. Source apportionment of PFASs was carried out by positive matrix factorization (PMF) for two cores from Lake Ontario. For the five lakes together, the total load of PFASs in sediments was estimated to be 245 ± 24 tonnes, which is about half the load for total PCBs. The recent annual loading was 1812 ± 320 kg/yr. Concentrations and inventories of PFASs were greatest in Lakes Erie and Ontario. Since 1947, concentrations of perfluorooctane sulfonic acid (PFOS) in ten cores have increased exponentially as a function of time with doubling times between 10 and 54 yr and have leveled off in three cores since 2000. PMF demonstrated an effective grouping of two particle-associated factors, characterized mainly by longer-chain PFASs (C ≥ 8) and two other factors of mainly shorter-chain compounds (C ≤ 6). Two factors feature only one dominant compound: factor 1, PFOS, and factor 3, perfluorobutane sulfonic acid (PFBS). Of all factors, factor 3 with PFBS has the largest contribution (47.8%). Significant scores for perfluorohexane sulfonic acid (PFHxS) and PFBS, along with flat or decreasing PFOS contributions since 2003, indicate that the replacement of PFOS with these compounds is beginning to take effect in the environment.

Metals are among the most toxic pollutants in urban stormwater. To investigate the concentration of dissolved and particulate fractions, the temporal variation during rain events, the effect of wash-off surface, and to assess the pollution status of metals in urban runoff, a total of 155 samples were collected mainly from trafficked areas, roofs and parking lots in Beijing from March to November 2015. Most of the metals were found mainly in the particulate fraction (68–96%) from trafficked surfaces, while for roof runoff Cd, Fe, Mn and Zn were found more equally in dissolved and particulate fractions. Metal concentrations were higher during start of a rain event than later (p < 0.05), and also were higher the longer the period of antecedent dry days. The mean concentration of all metals in trafficked areas exceeded both the Chinese standard Level III (swimming and fishery waters) and the European standards (surface water). Mean concentrations of Cd, Mn, Zn, Al, Fe, Pb and Ni from trafficked areas were 2–10 times higher due to higher traffic intensity and substantial atmospheric deposition, while Sb was 20 times higher than in any other reported data for urban runoff. Cluster analysis (CA) and principal component analysis (PCA) together with Pearson's correlation co-efficient suggested that Cd, Cr, Cu, Mn, Ni, Pb, and Zn mainly originates from vehicular activities, while Mn and Zn in roof runoff is due to atmospheric deposition. The geo-accumulation and pollution indices show that runoff from trafficked areas are moderately to heavily polluted by most metals, except Cu and Zn. Thus, Beijing urban runoff presents an environmental risk towards lakes, bathing water and drinking water. The results can be used as basis for development of stormwater and pollution control strategies.
