Toxicological effects of polystyrene microplastics on earthworm (Eisenia fetida)

Microplastics are plastic fragments of particle sizes less than 5 mm, which are widely distributed in marine and terrestrial environments. In this study, earthworms Eisenia fetida were exposed to 100 and 1000 μg of 100 nm and 1300 nm fluorescent polystyrene microplastics (PS-MPs) per kg of artificial soil for 14 days. Uptake or accumulation of PS-MPs in earthworm intestines, histopathological changes, oxidative stress, and DNA damage were assessed to determine the toxicological effects of PS-MPs on E. fetida. The results showed that the average accumulated concentrations in the earthworm intestines were higher for 1300 nm PS-MPs (0.084 ± 0.005 and 0.094 ± 0.003 μg/mg for 100 and 1000 μg/kg, respectively) than for 100 nm PS-MPs (0.015 ± 0.001 and 0.033 ± 0.002 μg/mg for 100 and 1000 μg/kg, respectively). In addition, histopathological analysis indicated that the intestinal cells were damaged after exposure to PS-MPs. Furthermore, PS-MPs significantly changed glutathione (GSH) level and superoxide dismutase (SOD) activity. The GSH levels were 86.991 ± 7.723, 165.436 ± 4.256–167.767 ± 18.642, and 93.590 ± 4.279–173.980 ± 15.523 μmol/L in the control, 100 nm, and 1300 nm PS-MPs treatment groups. In addition, the SOD activities were 10.566 ± 0.621, 9.039 ± 0.787–9.408 ± 0.493, and 7.959 ± 0.422–9.195 ± 0.327 U/mg protein for the control, 100 nm, and 1300 nm PS-MPs treatment groups, respectively, indicating that oxidative stress was induced after PS-MPs exposure. Furthermore, the comet assay suggested that exposure to PS-MPs induced DNA damage in earthworms. Overall, 1300 nm PS-MPs showed more toxic effect than 100 nm PS-MPs on earthworms. These findings provide new insights regarding the toxicological effects of low concentrations of microplastics on earthworms, and on the ecological risks of microplastics to soil animals.

In this study, by products such as red mud, phosphorus gypsum and fly ash were used as binders, and are compared with Portland cement (PC) in immobilizing Cu, Pb and Zn. Cu, Pb and Zn -doped pastes and mortars were prepared with a metal to binder ratio at 1%. Samples were cured for 7d, 14d and 28d. The unconfined compressive strength (UCS) test, a batch leaching test along with scanning electron microscopy (SEM) and thermogravimetric analysis (TGA) were applied for the testing of Cu, Pb and Zn-doped pastes and mortars. The UCS results show that red mud-phosphorus gypsum treated samples produce higher strength than these treated by red mud-fly ash, or PC. The results of leaching test revealed that the immobilization degree of heavy metals from these pastes depends on the leachate pH of these pastes. With the aid of the cement, red mud-phosphorus gypsum-cement pastes leached less metals compared to that of red mud-phosphorus gypsum pastes. The leachate concentrations of Cu, Pb and Zn from red mud-phosphorus gypsum-cement pastes are 1.5 mg/L, 1 mg/L, and 3 mg/L respectively. They are able to meet the China Ministry of Environment Protection (MEP) regulatory limit. With the increase of the curing time, the unconfined compressive strength and the leaching concentrations of these pastes showed a slightly increasing trend. In addition, SEM and TGA analyses show that the major hydration product is ettringite.

Di (2-ethylhexyl) phthalate (DEHP), a widely spreading environmental endocrine disruptor, has been confirmed to adversely affect the development of animals and humans. The formation of neutrophil extracellular traps (NETs) termed NETosis, is a recently identified antimicrobial mechanism for neutrophils. Though previous researches have investigated inescapable role of the immunotoxicity in DEHP-exposed model, relatively little is known about the effect of DEHP on NETs. In this study, carp peripheral blood neutrophils were treated with 40 and 200 μmol/L DEHP to investigate the underlying mechanisms of DEHP-induced NETs formation. Through the morphological observation of NETs and quantitative analysis of extracellular DNA, we found that DEHP exposure induced NETs formation. Moreover, our results proved that DEHP could increase reactive oxygen species (ROS) levels, decrease the expression of the anti-autophagy factor (mTOR) and the anti-apoptosis gene Bcl-2, and increase the expression of pro-autophagy genes (Dynein, Beclin-1 and LC3B) and the pro-apoptosis factors (BAX, Fas, FasL, Caspase3, Caspase8, and Caspase9), thus promoting autophagy and apoptosis. These results indicate that DEHP-induced ROS burst stimulates NETs formation mediated by autophagy and increases apoptosis in carp neutrophils.

Environmental pollution from plastic debris is a major global concern, being a potential threat to marine organisms and ecosystems. The accumulation of microplastics (MPs) in the oceans has notable ecological implications due to their long persistence, their potential ecotoxicity, and their ability to adsorb other pollutants and act as vectors of pathogens. Nevertheless, whereas the number of investigations documenting the presence of MPs in wild fish has increased, less studies have addressed the toxicological effects associated with the ingestion of MPs in long-term laboratory conditions. The aim of the present study was to assess the physiological response of gilthead seabream (Sparus aurata) exposed to low-density polyethylene (LDPE) MPs during a 90-day exposure followed by an extra 30 days of depuration through the application of oxidative stress biomarkers in the gut. No changes were observed in the Fulton condition factor of fish associated with MP intake. The activities of antioxidant enzymes and glutathione s-transferase and the levels of reduced glutathione progressively increased throughout the study in the MPs-fed group compared to the control group, reaching the highest values at 90 days. Similarly, the activity of the pro-inflammatory enzyme, myeloperoxidase, and the levels of oxidative damage markers -malondialdehyde and protein carbonyls-also increased after 90 days of exposure to an enriched diet with MPs. During the 30-day depuration period, all the biomarkers analysed tended to normalize, with the majority recovering values similar to those of the control group. In conclusion, MPs exposure during 90 days to S. aurata induced oxidative stress and a pro-inflammatory response in gut, and were able to recover after the exposure to MPs was removed.

Indoor plants can be used to monitor atmospheric particulates. Here, we report the label-free detection of combustion-derived particles (CDPs) on plants as a monitoring tool for indoor pollution. First, we measured the indoor CDP deposition on Atlantic ivy leaves (Hedera hibernica) using two-photon femtosecond microscopy. Subsequently, to prove its effectiveness for using it as a monitoring tool, ivy plants were placed near five different indoor sources. CDP particle area and number were used as output metrics. CDP values ranged between a median particle area of 0.45 × 10² to 1.35 × 10⁴ μm², and a median particle number of 0.10 × 10² to 1.42 × 10³ particles for the indoor sources: control (greenhouse) < milling machine < indoor smokers < wood stove < gas stove < laser printer. Our findings demonstrate that Atlantic ivy, combined with label-free detection, can be effectively used in indoor atmospheric monitoring studies.

Chemical warfare agents (CWA) dumped worldwide in all types of aquatic reservoirs pose a potential environmental hazard. Leakage of CWAs from eroding containers at dumping sites had been observed, and their presence in the tissues of aquatic animals was confirmed. However, the ecological effects of CWA have not yet been studied. In standardized laboratory bioassays, we tested if sublethal concentration of Clark I, an arsenic based CWA, can affect life histories (somatic growth rate, fecundity, size at maturity), population growth rate and stable isotope signatures of a keystone crustacean grazer Daphnia magna. We found that the life histories and fitness of daphnids reared in the presence of Clark I differed from those reared in Clark-free conditions. The effects were observed when Clark I concentrations were no less than 5 μg×L⁻¹. With increasing concentrations of the tested CWA, all of the tested parameters decreased linearly. The finding indicates that even sublethal concentrations of Clark I can affect crustacean populations, which should be taken into account when assessing the environmental risks of this particular CWA. We found intraspecific diversity in susceptibility to Clark I, with some clones being significantly less vulnerable than others. We also found that in the presence of Clark I, the ratio of heavy and light isotopes of nitrogen in the bodies of daphnids was affected – daphnids exhibited δ¹⁵N enrichment with increasing concentrations of this CWA. The isotopic composition of carbon was not affected by the presence of Clark I. The nitrogen isotopic signature may be used as an indicator of stress in zooplankton exposed to the presence of toxic xenobiotics.

Fourteen perfluoroalkyl substances (PFASs) in fishery organism, surface seawater, river water, rainwater, and wastewater samples collected from Jiaozhou Bay (JZB) in China and its surrounding area were determined to understand their contamination status, sources, health risk, and causes of spatiotemporal variations in the aquatic environment of a temperate bay adjacent to a metropolis. The total concentration of PFASs in 14 species of fishery organisms ranged from 1.77 ng/g to 31.09 ng/g wet weight, and perfluorooctane sulfonate (PFOS) was the dominant PFAS. ∑PFASs concentration in surface seawater ranged from 5.54 ng/L to 48.27 ng/L over four seasons, and dry season (winter and spring) had higher levels than wet season (summer and autumn). Perfluorooctanoic acid (PFOA) was the predominant individual PFAS in seawater, indicating that notorious C8 homologs remained the major PFASs in this region. The seasonal variation in seawater concentrations of three major PFASs, namely, PFOA, perfluoroheptanoic acid, and perfluorononanoic acid, was similar to that of ∑PFASs. However, the seasonal variation of PFOS concentration was different from that of ∑PFASs, with the lowest in winter and the highest in spring. In general, seasonal variations of terrigenous input and water exchange capacity were the main reasons for the spatiotemporal variation of PFASs in the aquatic environment of JZB. Moreover, bioselective enrichment for individual PFAS affected the partition of PFASs in different environment medium. Wet precipitation, sewage discharge, and surface runoff were the main sources of PFASs in this area. Nevertheless, the contribution of different sources to individual PFAS indicated a clear difference, and wastewater and river water were not consistently the most important source for every PFAS. Preliminary risk assessment revealed that the consumption of seafood, especially fish, from JZB might pose a certain extent of health risk to local consumers based on their estimated daily intake of PFASs.

Like their parent polycyclic aromatic hydrocarbons (PAHs), substituted polycyclic aromatic hydrocarbons (SPAHs), including methyl PAHs (MPAHs), oxygenated PAHs (OPAHs), and chlorinated PAHs (ClPAHs), exist ubiquitously in urban and agricultural rivers. Although laboratory studies have found the biological toxicities of certain SPAHs to be higher than that of their parent PAHs, the ecological risk of SPAHs in rivers has been largely ignored. Here, we studied the distribution, source and transport of PAHs and SPAHs as well as ecological risks in the Chaobai River System, which experiences a high level of anthropogenic activity. The results show that the concentration of ΣOPAHs (321 ± 651 ng/L) was higher than that of ΣPAHs (158 ± 105 ng/L), ΣMPAHs (28 ± 22 ng/L), and ΣClPAHs (30 ± 12 ng/L). We also found that (S)PAHs in Chaobai River mainly originated from Beiyun River (53%–65%), which receives considerable municipal wastewater treatment plant effluent from Beijing. The major transport pathway of (S)PAHs from Chaobai River was likely for irrigation (83%–86%) and transportation into Yongdingxin River (13%–16%), which finally merged into the Bohai Sea. The mixed chronic risk of (S)PAHs (risk quotient = 45 ± 53) was higher than the mixed acute risk (risk quotient = 1.9 ± 1.4), with all sites facing chronic risk and 90% of sites experiencing acute risk. Although the chronic and acute risks of (S)PAHs to plants, invertebrates, and vertebrates were mainly from PAHs (97.5% to chronic risk and 96.5% to acute), SPAHs still posed a chronic risk to invertebrates and vertebrates (risk quotient > 1). Accordingly, the ecological risk of (S)PAHs in Chaobai River should be taken into consideration for ecosystem protection. The transmission of PAHs and SPAHs from Chaobai River may also pose potential risks to farmland through irrigation, as well as to the Bohai Sea via river water discharge.

Concentrations of 99 volatile organic compounds (VOCs) were continuously measured online at an urban site in Beijing, China, in January, April, July, and October 2016. Characterization and sources of VOCs and their related changes during days with heavy ozone (O₃) pollution were analysed. The total observed concentration of VOCs (TVOCs) was 44.0 ± 28.9 ppbv. The VOC pollution level has decreased in Beijing but remains higher than in other Chinese cities. Alkanes comprised the highest proportion among seven major sampled VOC groups. The concentrations and sources of ambient VOCs showed obvious temporal variations. Six emission sources were identified by the positive matrix factorization (PMF), including biomass burning, coal combustion, gasoline vehicles, diesel vehicles, solvent usage, and biogenic + secondary emissions. The combustion source was the key control factor for VOC reduction in Beijing. From the potential source contribution function (PSCF) and concentration-weighted trajectory (CWT) model, Beijing, Tianjin, Hebei, Shanxi, Inner Mongolia, Shandong, and Henan were identified as major potential source regions of ambient VOCs. O₃ formation was sensitive to VOCs in Beijing according to the VOC/NOx ratio (ppbC/ppbv, 8:1 threshold). High- and low-O₃ days in July were identified, and high O₃ levels were due to both enhanced VOC emission levels and meteorological conditions favourable to the production of O₃. These findings provide evidence that the fuel combustion and regional transport have a great impact on concentrations and sources of VOCs in urban Beijing.

High phosphorus (P) load and consequent algal bloom are critical issues because of their harmful effects to aquatic ecosystems. The organic phosphorus (Po) cycling and hydrolyzation pathway in the sediments of a hypereutrophic lake area with high algae biomass were investigated using stable isotopes (δ¹³C and δ¹⁵N) along with C/N ratios, a sequential extraction procedure, ³¹P NMR spectrum, and alkaline phosphatase activity (APA) was measured simultaneously. C/N ratios lower than 10 combined with lighter δ¹³C (−23.5 to −25.2‰) and δ¹⁵N values (3.7–9.5‰) indicated that endogenous algal debris contributed to the predominant proportions of P-containing organic matter in the sediments. Sequential extraction results showed that Po fractions decreased as nonlabile Po > moderately labile Po > biomass-Po. Decreasing humic-associated Po (HA-Po) in sediments downward suggested the degradation of high-molecular-weight Po compounds on the geological time scale to low-molecular-weight Po including fulvic-associated Po (FA-Po), which is an important source of labile Po in the sediment. An analysis of the solution ³¹P NMR spectrum analysis showed that important Po compound groups decreased in the order of orthophosphate monoesters > DNA-Po > phospholipids. The significant correlation indicated that orthophosphate monoesters were the predominant components of HA-Po. Rapid hydrolysis of labile orthophosphate diesters further facilitated the accumulation of orthophosphate monoesters in the sediments. Additionally, the simultaneously upward increasing trend demonstrated that APA accelerated the mineralization of Po into dissolved reactive phosphorus (DRP), which might feed back to eutrophication in algae-dominant lakes. The significantly low half-life time (T₁/₂) for important Po compound groups indicated faster metabolism processes, including hydrolysis and mineralization, in hypereutrophic lakes with high algae biomass. These findings provided improved insights for better understanding of the origin and cycling processes as well as management of Po in hypereutrophic lakes.
