Genome modifications include potential about providing significant advantages on increasing yield performance and developing resistance to diseases. Gene editing methods that provides silencing or expressing of a gene which is an individual already has, have important potential for improving genetic structure without environmental effects. In recent times, new gene editing systems were developed. These are ZFNs (Zinc Finger Nucleases), TALENs (Transcription Activator-like Effector Nucleases) and CRISPR/Cas nuclease systems. CRISPR/Cas system is a microbial immune system that uses RNA guided nucleases for destroying genetic materials and its potential usage like a simple and efficient gene editing mechanism in animals is being evaluated recently. In this study it was aimed to determine the microbial diversity and level of antibiotic resistance patterns of Gram-negative bacterial isolates from the hospital sewages. The 219 Gram-negative bacterial isolates to 16 different antibiotics (belonging 10 classes), was investigated by agar diffusion method. A total of 18 species of bacteria were isolated: the most common strains isolated from all samples were Klebsiella oxytoca (27.4%), Klebsiella pneumoniae (20.5%) and Escherichia coli (20.1%). There was a high incidence of resistance to ampicillin (98.6%), streptomycin (95.9%) and erythromycin (90.0%), and a low incidence of resistance to cefepim (13.2%), imipenem (5.0%) and meropenem (3.2%). 35.6% of all bacteria isolated from hospital sewage were resistant to 9 different antibiotics. The multiple antibiotic resistances (MAR) index ranged from 0.25 to 0.94. Results show that hospital sewages have a significant proportion of antibiotic resistant Gram-negative bacteria, and these bacteria constitute a potential risk for public health.

This study was conducted in 2013 to determine the biochemical properties of 10 different monofloral honey types produced across Turkey. For the study, honey samples were obtained from beekeepers and the identity and relative percentage of dominant pollen types in samples was determined through microscopic analysis. Pollen analysis revealed that honey types chaste tree (Vitex agnus-castus) and sunflower (Helianthus annuus) contained high proportions of each respective pollen type. These proportions reached as high as 86%, while other honey types such as Anason had low Anasom pollen content, occurring at 45% in whole honey samples. Biochemical analysis revealed that the average honey sample quality characteristics were as followings: ash 0.37±0.04, electricity conductivity 0.79±0.07 mS/cm, moisture 19.15±0.09%, pH 3.97±0.02, acidity 29.07±0.39 meq/kg, hydroxymethylfurfural (HMF), 6.17±0.76 mg/kg, diastase 22.05±0.00, invert sugar 69.72±0.49% and sucrose 2.90±0.30%. These analyses confirmed that honey samples met standards outlined by the Turkish Food Codex Honey Standard, European Union Honey Standard and Codex Standards respectively.

Cocoa, Theobroma cacao L. is one of the main tropical industrial crops. Cocoa has a very high level of interfering substances, such as polysaccharides and phenolic compounds that could prevent the isolation of suitable protein. Efficient methods of protein extraction are a priority to successfully apply proteomic analyses. We compared and evaluated two methods (A and B) of tissue preparation for total protein extract by phenol/SDS extraction protocol. The difference in the application of the two methods was that extensively washed dry powder of pod tissue were made in Method A, whereas that crude extract were prepared Method B. Extracted proteins were examined using one-dimensional electrophoresis (1-D). Results show that each extraction method isolated a unique subset of cocoa pod proteome. Principal component analysis showed little variation in the data obtained using Method A, while that in Methods B showed no low reproducibility, thus demonstrating that Method A is a reliable for preparing cocoa pod proteins. The protocol is expected to be applicable to other recalcitrant plant tissues and to be of interest to laboratories involved in plant proteomics analyses. A combination of extraction approaches is recommended for increasing proteome coverage when using gel-based isolation techniques.

This study was aimed to examine the cotton production techniques and determined of input use in the cotton production. The primary data used in research was obtained from 94 cotton-growing farms by surveying method in the Antalya province. The data belonged to 2011 production period. In the research area 95.74% of agricultural farms made conventional farming. It was determined that cotton seed was used 2.43 kg per decares and 27.7% of farmers had soil analysis and 24.5% of them applied fertilizer according to the analyses. It was determined that the average fertilizer applications per decares were 24.9 kg nitrogen, 17.2 kg phosphorus, 8.2 kg potassium and 0.5 kg sulfur fertilizer. It was calculated that the average number of plant spraying 5.4, the number of irrigation 4.53 in the cotton cultivation. The application level of water-saving irrigation techniques of farmers was low(2.12% of farmers used drip and sprinkler irrigation system). Labor force was used as 53.95 hours and machine was calculated as 3.06 hours per decares. Mechanization of the cotton harvest increased. It was determined that 47.87% of farmers did not receive consultancy services; 22.34% of farmers were knowledgeable about the concept of integrated pest management and 41.5% of farmers were knowledgeable about good agricultural practices in the region. In order to increase the production of cotton in the region; new varieties which will be compatible to the region, higher levels of yield, resistant to disease and pests should be developed and farmers should be informed about technical applications by extension personnel.

Genetic variation is prerequisite for plant breeding. Nothing information existed in the literature for available diversity of Coriander accession in Turkey. Plant breeding activities are negligible in Turkey. So in order to start effective plant breeding program in Turkey, information on the available genetic diversity is viable. Therefore we planned to study the genetic variation and population structure of 29 Coriander accessions by seed storage protein (SDS). SDS analysis elaborated the lack of population structure and genetic bottleneck in the Coriander accessions in Turkey. Based on the results of this study, it was clear that sampling strategy was not appropriate and plant introduction should be made from different sources and diverse genotypes should be used as parents to initialize the effective Turkish Coriander breeding program.

Deficiency of sulphur (S) is an important limiting factor of plant growth for sustainable agricultural production. The decline in sulphur dioxide emission, decrease in S-containing fertilizer consumption due to the high cost of S-fertilizers, breeding of new high yielding species are the well known causes of S-deficiency. A greenhouse experiment was conducted to investigate the effects of several doses of K2SO4-S, CaSO4-S and elemental-S applied on growth, shoot dry matter yield, S and N concentrations of wheat cultivar. The experiments were conducted in three soils differed from available S concentrations. Effects of different S-treatments (0, 25, 50 and 100 mg S kg-1) and S-forms had significant effects on shoot dry matter yields of plants. Sulphur from different S-sources did not increase shoot S-concentrations in Eskisehir and Konya soils, but increase was significant obtained in the Harran soil. Shoot S-concentration in Harran soil for zero K2SO4 treatment was 0.09%, the values were 0.22, 0.26 and 0.27% respectively for 25, 50 and 100 mg kg-1 treatments. The results indicated significant effects of S-treatments on plant growth and yield mostly based on soil properties, especially the available S-levels

The present study is designed to attract attention to macroscobic and microscobic findings, causes and economic losses of wooden breast which is a significant muscle problem leading to visual and sensory differences in poultry breast meat quality. Wooden Breast is a crucial problem in poultry industry. It has started to seriously affect poultry production in European and other countries, and especially in United States of America. Woody breast formation is a situation characterized with completely hardened breast fillet or with swelling easily understood when touched. Surface of the fillet contains exudate with petechial bleeding or non-bleeding viscous gelatine. Woody structure called “wooden” can occur not only in breast meat but also in legs. Growth rate, gender, energy level of the feed, amino acid level, and selenium supplement are the factors affecting wooden breast-woody breast meat formation. Shaded colour, surface leak and abnormally hard tissue are seen in macroscopic image of woody breast. In microscopic findings, muscle fibers are less and round. Lesions don’t contain pathogenic micro-organisms, they are aseptic. Colour of the meat of woody breast is brighter and whiter, and (L* value) This muscle problem affects muscles visually and sensorial; but it doesn’t have a negative effect on human health. The more white lines and woody structure in breast meat, the less they are accepted among European consumers.

In the present study, it was aimed to determinate the effects of different substrates on mycelial growth, fructification, yield, sizes and colours of fruit bodies of Hericium isolates (HE-Ankara, HE-Denizli, HE, HE-Trabzon, HE-İzmit, HC, HE-Amerika). Experiments were conducted with 7 different substrates prepared with oak sawdust (MT), wheat bran (BK), cotton seed hulls (PK) and olive press cake (ZP) (80MT:20BK, 90MT:10PK, 80MT:20PK, 70MT:30PK, 90MT:10ZP, 80MT:20ZP, 70MT:30ZP). 1 kg (wet weight) of substrates were packed in polypropylene autoclaveable bags of 25x45 cm and sterilized in autoclave at 121oC during 90 minutes. Sterilized substrate was inoculated and then carried to growing room at 25±2ºC. After full colonization, the bags were exposed to 400 lux for a 12 hours photoperiod at 20±2ºC with a humidity of 80-90% in a cropping room. The best yield and BE were detected from oak sawdust medium supplemented with 20 and 30% cotton seed hulls on HE-Ankara, HE-Denizli, HE, HE-İzmit, HC, HE-Amerika isolates while the best yield and BE were detected from 70MT:30ZP on Trabzon isolate. Significant differences were found among substrates regarding yield, BE, average mushroom weigh, fruit body size and colour of Hericium isolates.

Like the whole sub-Sahara Africa, rainfall in Fina reserve is subject of strong inter-annual variability. This paper assesses farmers’ perception on land use utilised in the Fina biosphere reserve and their adaptation measures to climate variability. The statistical methods (descriptive and inferential analysis) are used in this study to determine farmers’ perceptions and the adaptation measures in the Fina reserve. Results reveal that 75.5% of the farmers noticed an increase in temperature and decrease in rainfall ignoring the recent recovery observed in the annual rainfall. The length of rainy season is considered to be shorter according to 77.6% of farmers involved to the investigation. However, all the farmers underlined frequent and longer dry spell. Bush fire is considered by 10.2% of farmers as the major factor affecting the natural resources of the reserve and only about 10% of farmers accorded much importance to agricultural land as contributor to reserve degradation. Most of the farmers are limited by lack of manpower and not by the reserve rule to increase their farmlands. The major adaptation measures are the methods of shifting cultivation and improving seed which are practiced by 51% and 87.8% of farmers, respectively. New adaptation strategies such as adoption of planting pits and stone-bunds for water retention need to be practiced by farmers.

In this study macrofungi specimens have been collected from Eucalyptus forests in Muğla district (Ula, Köyceğiz, Marmaris, Dalaman, Fethiye) in the years of 2013-2014. As a result of the field and fungarium studies, Totally 30 taxa belonging to 2 classes and 14 families have been identified. 2 of these taxa belong to Ascomycota, and the other 28 taxa were belong to Basidiomycota.
