Evaluation of patatin content in proteins of potato genotypes grown in Latvia

Potato proteins contains essential amino acids in considerably high concentration, therefore potatoes are considered to be one of the most valuable plant origin food for human consumption. Patatin forms one of the largest group of potato proteins with high potential to be used in food industry as a novel food. This study has been performed to approbate patatin determination method for evaluation of protein quality of potato genotypes, as well as evaluate patatin relative abundance (PRA) for breeding programmes to create in the future potato cultivars with higher value and potential to develop new products. The evaluation of patatin was performed in following steps– extraction proteins from potato, determination of patatin concentration and calculation of its relative abundance in proteins. Separation of patatin from potato tubers was made using extraction by SDS extraction buffer and determination of patatin in organically and conventionally (with differnt N suply) grown samples of 20 potato genotypes. The results of one-year study showed that patatin relative abundance of different cultivars varied from 1.65% to 50.2% and it was significantly different among genotypes. The nitrogen content of soil and maturity type of potato did not affect PRA significantly. Results provide impetus for further research.

The kinetics of plasma nitriding of CoCr alloy below temperatures of nitrides formation and mechanisms of nitrogen penetration are analyzed by proposed kinetic modeling in this article. Proposed nitrogen diffusion model is based on the trapping – detrapping (TD) model and developed taking into account the effect of the concentration dependent diffusivity of nitrogen, nitrogen adsorption on the surface of alloy and surface swelling process. The model indicates the influence of chromium atoms to nitrogen atoms diffusivity. The model consists of time and depth dependent diffusion, which is described by a partial differential equation, and it is solved by using Crank – Nicolson finite difference method. By fitting of experimental nitrogen depth profiles, it is shown that nitrogen diffusion coefficient varies with nitrogen concentration according to Einstein-Smoluchowski relation. Nitrogen depth profiles in plasma nitrided medical grade CoCr alloy (ISO 5831 – 12) at T = 400 °C for 1, 4 and 20 hours calculated on the basis of this model are in good agreement with experimental nitrogen profiles. Furthermore, the swelling process is showed and analyzed, derived the dependency of swelling rate on nitriding duration – the swelling rate is inversely proportional to the square root of nitriding duration. The obtained diffusion coefficient value and the swelling process rates satisfy the experimental data form Ref. The derived model explains physical processes during plasma nitriding and allows obtaining nitrogen depth profiles for any requisite nitriding duration.

Nitrogen management strategy in plant growth period based on chlorophyll content evaluation in plant can improve nitrogen usage efficiency and reduce environmental contamination. This study is aimed to determine the impact of different nitrogen and sulphur fertilizers rates on dynamics of chlorophyll content in winter wheat during vegetative growth and to determine the relationship between nitrogen and chlorophyll content and grain yield of winter wheat. Field trial involving a winter wheat (Triticum aestivum L.) variety ‘Kranich’ was conducted at the LUA Research and Study Farm Vecauce during a three-year period (2012-2015). The treatments were 0, 85, 153, 175+S21, 175 (in 2015), 187 N kg haE−1 and different nitrogen norms according to chlorophyll meter Yara N-tester (Konica Minolta Ltd.) data: 180, 150, 205 N kg haE−1 depending on the year. The results of the trial show that the maximum chlorophyll content in different plant parts was observed at the end of flowering stage. The chlorophyll content depended on the level of mineral fertilisation. The highest chlorophyll content in leaves, stems and ears was obtained by using additional sulphur in two trial years. Usage of chlorophyll meter Yara N-tester obtained the highest chlorophyll content in all analysed plant parts in one trial year. Chlorophyll content was significantly dependant on plant growth stage in stems in all trial years, in leaves in two trial years, and in ears in one year. Nitrogen fertilization significantly affected chlorophyll content in leaves and stems in one trial year. Close positive correlation was observed between grain yield and wheat plant chlorophyll content and average nitrogen concentration at the end of flowering stage in all three trial years.
