Comparison of alpha-galactosidase activity in cells and cultivation medium of lemon balm (Melissa officinalis L.)

A rapid, simple and reproducible method for the identification of extracelullar alpha-galactosidase has been developed. The alpha-galactosidase secreted by callus culture or by roots of seedlings hydrolyses the substrate (1-naphthyl-alpha-D-galactopyranoside) in alpha-D-galactose and 1-naphthol, the latter forming slightly soluble reddish brown or reddish violet azo-dye after simultaneous azocoupling with hexazonium p-rosaniline or Fast Blue B. Colour changes under callus culture and around it as well as those in seedling roots are a demonstration of the activity of extracellular alpha-galactosidase by the subjects under study. The intensity of colouring is a measure of their enzymatic activity.
