Evaluation of some commercial pectinases

An evaluation procedure was developed for some commercial pectinases. The procedure included assay of pectolytic and non-pectolytic activities, determination of protein, total carbohydrates and reducing sugars, and fractionation with affinity chromatography. Ultrazym 100, Rapidase and pectinase contained 132.86, 3.57 and 138.72 polygalacturonase units/mg protein, respectively. It was found that Ultrazym 100 and Rapidase possesed cellulase, amylase, invertase and Beta-galactosidase activity while pectinase contained the first three activities only. The protein content of the three preparations was in the range of 1.6 - 22.5%. Total carbohydrates content of pectinase, Ultrazym 100 and Rapidase solutions (5 mg/ml) was 66, 76 and 36%, respectively. Separation of exo-and endo-polygalacturonases (present in Ultrazym 100) was achieved by affinity chromatography using cross-linked sodium pectate. It was further confirmed by analysis of end products
