Three diagnostic tests for thiamine deficiency were studied for the assessment of thiamine status in 23 consecutive alcoholics (alcoholic intake over 50 g/day for more than 1 year; 20 men, 3 women; ages 25-84) and in 21 healthy controls (14 men, 7 women; ages 40-75). The study led to the development of a hypothetical model incorporating all 3 tests that may unequivocally distinguish intermediate grades of thiamine status, ranging from normal status to severe deficiency. The 3 tests were: (1) erythrocyte transketolase activity (ETKA); (2) ETKA stimulated in vitro with thiamine pyrophosphate (TPP); and (3) the percent increase in ETKA after TPP in vitro stimulation.(wz)
