An effective way of preventing undesirable boar taint in pork meat caused by the presence of androstenone, skatole and indole is surgical castration of piglets. This, however, arouses growing social opposition. An alternative method of inhibiting the development of unpleasant odour is immune castration. The aim of the study was to compare the effectiveness of both methods of castration for the elimination of the compounds responsible and to assess the suitability of oral fluid for pre-slaughter predictive testing for boar taint. The research material was pooled oral fluid and fat samples taken from gilts and surgically and immunologically castrated piglets. The samples were tested with a liquid chromatography– tandem mass spectrometry method developed in this research. The compounds giving rise to boar taint were found only sporadically above the accepted limits; only one sample of oral fluid contained skatole at a concentration above 200 μg L⁻¹ and one contained indole more concentrated than 100 μg L⁻¹. Indole above the limit value was also detected in one fat sample. In none of the tested samples was androstenone found. The results indicate the similar effectiveness of both methods of piglet castration on the reduction of compounds generating boar taint. The usefulness of testing oral fluid for the ante-mortem prediction of boar taint has not been fully confirmed and further investigation is needed.

The plate counting method widely used at present to discern viable from non-viable Brucella in the host or cell is time-consuming and laborious. Therefore, it is necessary to establish a rapid, simple method for detecting and counting viable Brucella organisms. Using propidium monoazide (PMA) to inhibit amplification of DNA from dead Brucella, a novel, rapid PMA-quantitative PCR (PMA-qPCR) detection method for counting viable Brucella was established. The standard recombinant plasmid with the target BCSP31 gene fragment inserted was constructed for drawing a standard curve. The reaction conditions were optimised, and the sensitivity, specificity, and repeatability were analysed. The optimal exposure time and working concentration of PMA were 10 min and 15 μg/mL, respectively. The correlation coefficient (R²) of the standard curve was 0.999. The sensitivity of the method was 10³ CFU/mL, moreover, its specificity and repeatability also met the requirements. The concentration of B. suis measured by the PMA-qPCR did not differ significantly from that measured by the plate counting method, and the concentrations of viable bacteria in infected cells determined by the two methods were of the same order of magnitude. In this study, a rapid and simple PMA-qPCR counting method for viable Brucella was established, which will facilitate related research.

This paper provides an overview of the current knowledge of chlamydiae. These intracellular microorganisms belonging to the Chlamydiaceae family are widely distributed throughout the world. Constant development of culture-independent approaches for characterisation of microbial genomes enables new discoveries in the field of Chlamydia. The number of new taxa is continuously increasing as well as the range of hosts. New species and genotypes are constantly being discovered, particularly new avian and reptilian agents, which are discussed in this article. Interestingly, wild animals are the main hosts for new Chlamydia species including different species of bird, turtle and snake. The availability of next-generation sequencing opens up a new prospect for research and leads to deeper knowledge of these interesting microorganisms about which much is still to discover.

Marek’s disease (MD) is a tumourous disease caused by Marek’s disease virus (MDV) and most commonly described in poultry. The aim of the study was to determine the occurrence of Marek’s disease virus infections in Poland and analyse clinical cases in the years 2015–2018. The birds for diagnostic examination originated from 71 poultry flocks of various types of production. Birds were subjected to anatomopathological examination post mortem, during which liver and spleen sections and other pathologically changed internal organs were taken. These sections were homogenised with generally accepted methods, then total DNA was isolated and amplified with a real-time PCR. A pair of primers complementary to the MDV genome region encoding the meq gene were used. MDV infection was found predominantly in broiler chicken flocks (69.01%), and also in layer breeder (9.85%) and commercial layer flocks (7.04% each). The results of research conducted in the years 2015–2018 clearly indicate that the problem of MDV infections is still current.

The objective of this research was to evaluate the antibody response to multiple doses of an inactivated mixed vaccine against Histophilus somni, Pasteurella multocida, and Mannheimia haemolytica, and to investigate the influence of age at time of vaccination in the field. Healthy female Holstein calves received the vaccine at the age of 5–12 days and 2, 3, or 4 weeks later in the first experiment or at 1, 2, or 3 weeks of age and 4 weeks later in the second. Blood samples were collected at each vaccination and 3 weeks after the booster dose. Based on the antibody titres after the vaccinations, calves were divided into positive and negative groups for each of the bacteria. Calves in the control group were vaccinated only once at the age of 19–26 days. Antibody titres against H. somni and P. multocida were significantly increased by the booster. After the second vaccinations, the titres against each bacterium were higher than those of the control group, and the M. haemolytica-positive percentage in calves with high maternal antibody levels (MAL) exceeded that in calves with low MAL. In the first experiment, a majority of the M. haemolytica-positive calves tended to have received the primary dose at seven days of age or older. A booster dose of the inactivated bacterial vaccine in young Holstein calves increased antibody production and overcame the maternal antibodies. Calves should be vaccinated first at seven days of age or older.

Candida species are a natural component of the intestinal tract microflora, but in favourable conditions they can cause superficial, mucosal, or even systemic candidiasis. Poultry production might be a source of human drug-resistant yeast infections, including Candida spp. The limited data concerning the antifungal susceptibility of poultry Candida isolates prompted us to carry out research to determine the susceptibility of isolates from turkey intestinal tracts. The beak cavity, crop and cloaca were swabbed of 580 turkeys from 58 flocks in western Poland. The susceptibility tests were conducted using the E-test method with amphotericin B, fluconazole, itraconazole, and voriconazole on 52 isolates of C. albicans, C. catenulata, C. glabrata, C. palmioleophila, C. rugosa, C. krusei and C. lusitaniae. All isolates were susceptible to voriconazole. According to the MIC values obtained for amphotericin B and fluconazole, all Candida spp. isolates were classified as susceptible according to the described breakpoints except for C. krusei, which was the only isolate that was amphotericin B-, fluconazole- and itraconazole-resistant. The susceptibility to itraconazole varied: 11 of the Candida isolates were susceptible (21.1%), 29 were dose-dependently susceptible (55.8%), and 12 isolates were resistant (23.1%). There are few resistant strains of Candida in turkeys, and the drug resistance varies. When Candida passes from turkeys to humans, there is a wide range of antifungal treatment options.

With the advancement of research and development of medical technology, there is an increase in the number of animals used in research. As millions of experimental animals are being used in different experiments worldwide, there is a lot of debate about the pain, distress, and death experienced by animals during scientific experiments. Experimental animals also require skilled manpower, time-consuming protocols and are costly. There are various alternatives to animal testing to overcome the drawbacks of animal experiments and avoid unethical procedures. Russell and Burch published principles of the Humane Experimental Technique which includes a strategy of 3 Rs (i.e. reduction, refinement, and replacement) in 1959. They introduced and defined the terms replacement, reduction, and refinement, which subsequently have become known as ‘alternatives’ or ‘alternative methods’ for minimizing the potential for animal pain and distress in biomedical research. Therefore, different methods and different alternative organisms are being used to implement the 3 Rs strategy. So, a brief account of these alternatives and the advantages associated is discussed in this review with examples. An integrated application of these approaches would give insight into the minimum use of animals in scientific experiments.

As of 2016, a total of 38 research projects have been undertaken, with the involvement of 38 principal researchers and 266 co-researchers from various Malaysian universities, research institutes and government agencies, in collaboration with industry partners, to help solve some of the issues faced by EBN industry. In the early stage, research areas were focused onfundamental works which are important to provide the scientific basis for some of the claims made by producers, assisting in issues raised by ranchers and industry, and to gather new knowledge in swiftlet and EBN. The researchers worked mainly based on the problems faced by the industry. Besides publications in journals, proceedings and books, more than 30 Masters, PhDs and technical experts have been trained under these projects. In addition, several patents have been filed, social innovations and ideas have been shared with the community and industry. A few products are ready to go to the next level for commercialisation. Industry partners are involved in many of these stepsand outputs. Some of the issues faced by the industry have been partially solved and are in the process of refinement. The engagement with the industry will be further strengthened by getting more industry partners to be involved in the relevantprojects specific to problems faced by the industry, for the benefits of EBN industry and nation’s wealth creation.

The study was conducted toknow the anatomy of the brachial plexus ofWhite New Zealand Rabbit (Oryctolaguscuniculus). Ten healthy male and femaleWhite New Zealand rabbits were dissectedin this study. It was found that the brachialplexus of the White New Zealand Rabbitwas formed by ventral branches of C5,C6, C7, C8, T1 and T2 spinal nerves.The cranial trunk was formed by thecaudal branch of C5, C6 and caudal trunkformed by rami ventralis of C7, C8, T1and the cranial branch of ventral ramusof T2. Cranial pectoral nerve originatedfrom the caudal trunk spreading into thepectoral muscles. The musculocutaneousnerve innervates into the brachial muscleand the axillary nerve into the subscapularmuscle. The radial nerve was dividedinto two branches as ramus profundusand ramus superficial then divided intothe digital dorsal common III and IV.The thoracodorsal nerve innervates thelatissimusdorsi muscle. The median nervewas divided into digital dorsal commonI, II, III and IV nerves. The ulnar nerveformed the caudal cutaneous antebrachialthen digital dorsal common IV and Vnerves. Lateral thoracic and caudal pectoralnerves originated from the caudal trunk.The origin and distribution of brachialplexus resemble that of porcupines butdiffer from other mammals.

The objective of this study was to evaluate the endocrine and immune responses of steers challenged with infectious bovine rhinotracheitis virus (IBRV). For the study, twelve crossbred beef steers weighing approximately 228.82 kg were fitted with indwelling rectal temperature monitoring devices and randomly assigned to a Control (CON) or IBRV treatments. Immune challenged steers received an intra-nasal dose of IBRV (4 ml total volume; 2ml/nostril) and CON steers received an intra-nasal dose of saline (2 ml/nostril). On day 0, steers were challenged and placed into isolated paddocks. At 72 hours post-inoculation, steers were fitted with indwelling jugular catheters and placed into individual stanchions. Blood samples were intensively collected on days 4 through 8 post-inoculation. Serum was analyzed for cortisol, interleukin-6, interferon-gamma, tumor necrosis factor-alpha, growth hormone, and insulin-like growth factor-1. On day 2, IBRV challenged steers had increased rectal temperature compared to CON steers (P < 0.05); the greatest rectal temperatures were observed on day 4, after which rectal temperatures returned to baseline by day 6. Serum concentrations of cortisol, interferon-gamma, and growth hormone exhibited a similar response pattern increasing by day 2 for the IBRV challenged steers, with the greatest increases observed on day 4, and subsiding on day 6. There was a decrease (P = 0.04) in growth hormone production in IBRV challenged steers, but no difference in insulin-like growth factor-1. Collectively, the data revealed that alterations in the somatotrophic axis were not associated with large increases in circulating pro-inflammatory cytokines. Results suggest that the dose of the virus used in the present study, while sufficient to elicit a febrile response, was not enough to elicit a robust pro-inflammatory immune response.
