Immunization of young chicks using graded dose of wild strain of <i>Eimeria tenella</i> النص الكامل

A wild strain of Eimeria tenella was isolated and utilized for immunization studies. Its optimal sporulation was attained at room temperature 24-25 °C after 24-48 h. Two groups of chicks were immunized by dosing a graded dose of five oocysts/chick/day for 6 days followed by 50 oocysts/chick/day for 7 days. A third group was not immunized and served as a negative control. Immunized chicks gained mass at the same rate as unimmunized ones, but when challenged with 200 000 oocysts/chick, mass gains declined in the unimmunized group. The growth rate of immunized chicks was not affected by challenge (P > 0.05). Upon challenge, unimmunized chicks produced significantly more oocysts than immunized chicks (P < 0.005). Immunized chicks withstood a challenged with 200 000 oocysts/chick without developing any clinical signs whereas the unimmunized chicks developed typical clinical signs of coccidiosis. Unimmunized chicks had significantly more severe lesions in the caecum than any other group (P > 0.005) and also produced significantly more oocysts than any other group (P > 0.005).

Ingestion of the plant Nolletia gariepina was confirmed as the cause of acute mortalities in cattle in the Kuruman area of the Northern Cape Province of South Africa. The aim of this trial was to investigate the toxic effects of this plant with respect to clinical signs, pathophysiology and pathology using the sheep as a model. At dosages of 1.5 g dried, milled plant material/kg body mass there were no detectable abnormal findings, while at dosages of 2.8-3.0 g/kg most of the animals died acutely. In subacutely affected sheep, depression, inappetance, teeth grinding, tachycardia, weak ruminal movements and recumbency were noticed. The most prominent pathophysiological changes observed, included a sharp rise in non-protein nitrogen substances in the plasma, remarkable decline in glomerular filtration rate, increase in sodium and potassium excretion, and a rise in urine gamma glutamyltransferase activity. Macroscopically a severe nephrosis was present in all the animals. The most important findings detected histologically were necrosis of the proximal convoluted tubular epithelium and large numbers of protein casts in the lumens.

Poisoning with cardiac glycoside-containing plants is collectively the most important plant-associated poisoning of livestock in southern Africa. As a diagnosis of this significant poisoning is currently based on circumstantial evidence, a practical chemical procedure indicating the presence of cardiac glycosides in plants and animal specimens would be of considerable benefit. The fluorescence polarization immunoassay (FPIA) method, used to determine digoxin plasma levels in humans and dogs, was adapted to estimate cardiac glycoside levels in known cardiac-glycoside- containing plants as well as in the rumen and organs of dosed sheep. Positive FPIA values were obtained with bufadienolide-containing plants, while negative results were obtained with plants not known to contain cardiac glycosides. The FPIA has aided in the diagnosis of cardiac glycoside poisoning in livestock and game in 30 outbreaks examined at the Division of Toxicology, Onderstepoort Veterinary Institute. Each outbreak is briefly described. As a result of this assay, a better understanding of cardiac glycoside poisoning has been reached.

Psittacine beak and feather disease (PBFD) is a common viral disease of wild and captive psittacine birds characterized by symmetric feather loss and beak deformities. The causative agent, beak and feather disease virus (BFDV), is a small, circular single-stranded DNA virus that belongs to the genus Circovirus. BFDV can be detected by PCR or the use of haemagglutination (HA) and haemagglutination inhibition (HI) assays that detect antigen and antibodies respectively. Erythrocytes from a limited number of psittacine species of Australian origin can be used in these tests. In South Africa, the high cost of these birds makes them difficult to obtain for experimental purposes. Investigation into the use of erythrocytes from African Grey parrots and Brown-headed parrots yielded positive results showing the haemagglutinating activity of their erythrocytes with purified BFDV obtained from confirmed clinical cases of the disease. The HA activity was further confirmed by the demonstration of HI using BFDV antiserum from three different African Grey parrots previously exposed to the virus and not showing clinical signs of the disease.

Serum samples from 474 domestic pigs (Sus scrofa) from Zimbabwe were tested for anti-Toxoplasma gondii IgG antibodies using the indirect fluorescent antibody test. The results showed that T. gondii infection is widespread in Zimbabwean pigs. Seroprevalence was lowest in fattening pigs from large and small-scale commercial farms that practise good hygiene (19.75 % of 238) and highest in backyard scavenging pigs (35.71 % of 70). Only 11.7 % (11) of the 127 positive samples had titres of > 1:400 and nine (81.82 %) of these 11 originated from pigs reared under poor hygienic conditions. A prevalence of 3.51 % was found in the same group of fattening pigs using an indirect IgG enzyme-linked immunosorbent assay at the single serum dilution of 1:400. The serosurvey shows the importance of modern intensive husbandry systems in reducing the prevalences of T. gondii infection in domestic pigs.

This paper focuses on the geographical distribution and habitats of Bulinus forskalii, the snail intermediate host of the conical fluke of equids, Gastrodiscus aegyptiacus as reflected by the 1 209 samples in the database of the National Freshwater Snail Collection of South Africa. The 362 different loci on record represent an extensive distribution in KwaZulu-Natal Province, the Limpopo Province, the coastal areas of the Eastern Cape Province and the south-eastern part of the North West Province. Although it was recorded from all types of water-body represented in the database, the highest percentages of samples were recovered from dams (30.4 %) and brooks (28.2 %). The majority of samples came from perennial habitats (59.1%), 60.7% from habitats with standing water, 54.0 % from habitats with clear water and 71.8 % from habitats of which the water was described as fresh. The majority of samples (39.5 %) were collected in habitats of which the substratum was recorded as muddy. The highest percentage of samples, by far (81.5 %), was collected in habitats that fell within the mean yearly temperature interval ranging from 15-20 °C. An integrated decision tree constructed from the data in the database indicated that temperature and type of water-body played a decisive role in determining the presence of B. forskalii in a given area. The results of experimental exposure to miracidia of a local strain of both Schistosoma haematobium and Schistosoma mattheei in the laboratory indicated that a local strain of B. forskalii was incompatible with both these strains of parasite. Research to clarify the role of B. forskalii in the transmission of both Calicophoron microbothrium and G. aegyptiacus in South Africa, is recommended.

In this study we assessed the influences of ultraviolet (UV) light B radiation on epidermal cells by apoptotic sunburn cell (SBC) and the effect of green tea treatment on the inhibition of SBC formation in SKH1-hr mouse. The extent of changes following 200 mJ/㎠ (0.5 mW/sec) was studied at 0, 3, 6, 12, 18, 24, 30 or 36 hours after exposure. SBCs were recognized by 3 hours after irradiation. There was tendency to increase from 3 hours to 24 hours and decrease from then to 36 hours after irradiation. The mice that received 0, 25, 50, 100, 200, 400 or 800 mJ/㎠ of UVB were examined 24 hours after irradiation.

To elucidate the roles of phospho-IkB expression in the development and progression of EAE, we investigated the expression of phospho-IkB in the central nervous system (CNS) of rats during experimental autoimmune encephalomyelitis (EAE) using immunohistochemistry and Western blot analysis. In Western blot analysis, the increased expression of phospho-IkB went parallel to severity of EAE. The expression of phospho-IkB increased significantly at the peak stage of EAE followed by gradual decrease. Immunohistochemical studies showed that the phospho-IkB immunoreactivity was mainly expressed in inflammatory cells (macrophages, T cells) and glial cells (astrocytes, microglial cells) at the peak stage of EAE and disappeared at the recovery stage.

The melanocortin 1 receptor (MC1R) plays an important role in regulation of melanin pigment synthesis within mammalian melanocytes. Mutations within the gene encoding MC1R have been shown to explain coat color variations within several mammalian species including cattle. To develope a rapid and accurate method for the identification of Hanwoo meat, we performed a single nucleotide polymorphism (SNP) analysis in Melanocortin 1 receptor (MC1R) gene using TaqMan∨® MGB probe-based real-time PCR. Two specific probes (one for Hanwoo and the other for Holstein and Black angus) were designed.

We report septicemic colibacillosis accompanied by white-spotted kidney in a 30-day old female Holstein-Friesian dairy calf. Grossly, there were numerous white spots sized average 0.5cm in diameter in both kidneys. When sectioned sagittally, there were radially oriented gray streaks extending outward and reaching the renal cortex. The renal papillae were ulcerated, white to gray in color and very friable. Histologically, there was extensive purulent inflammation characterized by severe neutrophilic cellular infiltrations in the tubular lumens and interstitia. In addition, massive coagulative necrosis were found in the apices of papillae.
