The role of lipid peroxidation products and antioxidant enzymes in the pathogenesis of aseptic and purulent inflammation in cats

Objective: The work aimed to determine the state of lipid peroxidation products and the activity of the antioxidant system in cats with aseptic inflammation and purulent-inflammatory processes of varying severity. Materials and Methods: The intensity of the process of lipid peroxidation- antioxidant system processes in blood plasma was evaluated using commercial kits. The level of diene conjugates (DC), the content of malondialdehyde (MDA), the level of medium-weight molecules were deter¬mined from lipid peroxidation indices. The state of antioxidant protection was assessed by indi¬cators of superoxide dismutase (SOD) activity, catalase (CT), ceruloplasmin (CP) concentration, glutathione peroxidase activity, glutathione reductase (GLR), and total antioxidant activity (AOA). Results: With aseptic inflammation in the blood of cats, a slow activation of peroxidation reactions occurred. The DC indicator increased by 1.4 times, the SOD level rose by 1.38 times, the amount of GLR by 1.04 times, and the activation of total AOA by 9.06. In sepsis, the values of DC, MDA, and medium-weight molecules increased by 4.4, 8.4, and 8.8 times, respectively. With abscesses in the blood of cats, an increase in CT, CP, and GLR is observed against a background of a decrease in glutathione peroxidase. With purulent wounds, the total AOA of plasma increases by 2.5 times; with abscesses, the total AOA increases by 1.9 times, and with sepsis, it decreases by 1.5 times. Conclusion: With surgical infections in cats, deep biochemical changes or irreversible biochemical changes (in sepsis) occur that indicate a significantly enhanced catabolic orientation of metabolic processes and the accumulation of toxic metabolites, which leads to damage and a decrease in tissue regenerative capacity. Objective: This study aimed to salvage the study population from the fatality that occurs due to iatrogenic injury to the thoracic cavitys pleural membrane. Materials and Methods: An experimental study of temporomandibular joint arthroplasty with costochondral graft was carried out on 72 healthy Oryctolagus cuniculus species of male rab¬bits. The rabbits were distributed into two age groups: growing (34 months) and adult (1218 months). All the procedures were carried out under general anesthesia with xylazine hydrochlo-ride and ketamine hydrochloride after calculating the doses, maintained by halothane and O2 inhalations. Out of 72 rabbits, 33 rabbits had accidental perforation of the pleural membrane observed that required a chest drain. Results: In this study, 21 (63.64%) rabbits received chest drain and salvaged. The rest of the rab¬bits (n = 12; 36.36%) that did not receive any chest drain and died. Most of the rabbits (n = 17; 81%) were under the growing group, weighing less than 2 kg and four (19%) were adult rabbits. Conclusion: This manual chest drain is life-saving for rabbits. It is a new addition to the advance¬ment of thoracic surgery on animals. It is cost-effective and safe. The developed customized drain¬age system may make it easier to harvest the costochondral graft-related experiments. [J Adv Vet Anim Res 2021; 8(1.000): 138-145]

Objectives: The study intended to detect the presence and distribution of avian encephalomyeli¬tis virus (AEV)-specific antibodies in Sonali (cross-bred) parent chickens regarding farm location, flock size, and age in Bogura district of Bangladesh, a Sonali chicken belt. Materials and methods: A total of 275 Sonali parent chickens blood samples were collected ran¬domly from 39 flocks during laying age with a healthy and non-vaccination history against AEV. Blood samples were collected aseptically from the wing veins of chickens using 3-ml syringes and sera were separated. Then, the sera were transferred to the laboratory by maintaining a cool chain. Indirect enzyme-linked immunosorbent assay was used to detect the specific antibodies against AEV present in the sera samples. Results: Overall, 70.18% of the chickens were found seropositive for AEV antibodies. Based on the location, the highest seropositivity was recorded in Bogura Sadar [91.30%, confidence inter¬vals (CI) 79.21%97.58%], and the lowest was in the Adomdighi sub-district (45.45%, CI 29.49% 63.08%). For flock size, AEV seropositivity was significantly (p < 0.05) higher in the large flock (82.22%, CI 72.74%89.48%). Regarding age groups, the seropositivity of AEV was significantly (p < 0.05) increased with chickens age. Higher seropositivity was noted in chickens aged >51 weeks (89.32%, CI 81.69%94.55%). Conclusion: The results indicate that AEV is circulating in the environment, and chickens were exposed to the field strain of AEV. To the best of our knowledge, this is the first report on AEV in chickens in Bangladesh. Proper vaccination and standard farm biosecurity practice could minimize AEV infection in chickens. A detailed epidemiology study, detection, and characterization of the AEV would be essential for effective AEV infection control. [J Adv Vet Anim Res 2021; 8(1.000): 78-83]

Objective: The objective of this study was to estimate the heritability (h2 ), repeatability (r), and correlations (r yi yi ) in some traits of zootechnical interest in a population of New Zealand white rabbits of a tropical dry forest area. Materials and Methods: Three mating groups were formed, each one of 1 male and 70 females. The traits evaluated were littersize at birth (LB), born alive (BA), born dead (BD), litter weight born alive (LW), litter weight at weaning (LWW), weaning weight (WW), slaughter weight (SW), and carcass weight (CW). Weaning took place at 42 days, and the fattening phase lasted 60 ± 3 days. A descriptive statistical study was carried out on the study variables. Paternal heritability was esti mated (h2 f ) and maternal (h2 m), repeatability, rabbit index IC, and Pearsons correlations (r yi yi ) between traits. The descriptive statistics showed high variation for the BD traits. Results: The values of the productivity found were similar to those presented in studies around the world. h2 presented magnitudes between low and medium. h2 f ranged between 0.09 and 0.42 and between 0.11 and 0.45 for h2 m. In general, the values of h2 m were higher than the values of h2 f . The r valuesforthe traits LB, BA, LW, LWW, and SW presented low magnitude, while it was medium for WW and CW. From the values of r, IC was calculated for each of the rabbits, allowing their categorization, which will be used in future selection plans. r yi yi among the variables ranged from −0.01 to 0.860. They were generally positive and mostly not significant (p > 0.05); they took a magnitude from low to moderate, except for the correlation between LB and BA. Conclusion: The production of rabbits under tropical conditions is similar to other reports. The genetic parameters evaluated were medium-to-low, indicating a robust non-additive gene and/ or environmental effect. [J Adv Vet Anim Res 2021; 8(3.000): 471-478]

Objective: The purpose of this study was to determine the effects of the ethanolic extract of avocado seed flour on the estrous cycle characteristics, the concentrations of reproductive hormones [luteinizing hormone (LH) and estradiol], and the activities of some tissues (ovarian and uterine) that are markers of oxidative stress in female guinea pigs (Cavia porcellus). Materials and Methods: Twenty-four female cavies with normal estrous cycles and equivalent body weights (464.25 and 71.88 gm) were randomly assigned to four groups, each with six females. The control group received 1 ml of distilled water orally, whereas the EE100, EE200, and EE400 groups received 100, 200, and 400 mg/kg bw of ethanolic extract of Persea americana seed, respectively. Following that, three consecutive estrous cycles were observed using vaginal smears. After the trial, the females were slaughtered, and blood and organs were gathered for study. Results: The mean duration of the estrus phase is considerably (p < 0.05) longer in group EE100 animals than in control animals. LH concentrations were substantially (p < 0.05) higher in females in group EE200 than in controls. Total cholesterol levels typically dropped in females in the extract groups, but only significantly (p < 0.05) in those in group EE100 compared to the control group. Serum estradiol levels increased considerably (p < 0.05) in treated females compared to controls. Catalase activity rose considerably (p < 0.05) in the ovaries of group EE400 when compared to the control. Superoxide dismutase activity rose considerably (p < 0.05) in the uterus of female cavies given the extract compared to the control. Conclusion: Avocado seed ethanolic extract prolongs the estrus phase, increases estradiol and LH levels, and protects the uterus from oxidative stress in female cavies. Objective: Sertoli cells (SCs) are important sustentacular cells in the seminiferous tubules of the testis. Isolation and identification of SCs are the premise for studying their functions. Since New Zealand rabbit is a stable strain which is widely used for biomedical research and animal farming, this study aimed to develop a simple and effective protocol for SC isolation in New Zealand rabbits. Materials and Methods: The SCs of three 30-day-old New Zealand rabbits were isolated by incu¬bation with enzymatic digestion I (Dulbeccos modified Eagle medium supplemented with 1 mg/ ml collagenase IV and 50 μg/ml DNase I) and digestion II (digestion I + 1 mg/ml hyaluronidase + 1 mg/ml trypsin), as well as differential plating. The cells were enriched and identified by using immunocytochemical staining and reverse transcription polymerase chain reaction analysis. Results: Homogeneous cells were obtained. They presented the typical large cell body and an irregular pyramidal shape after differential plating and passaging. These cells expressed mRNA of the SC marker sex-determining region Y-box 9 (SOX9) instead of the Leydig cell marker StAR. Immunocytochemically, they are positive of SOX9, GATA binding protein 4, and androgen-binding protein. Conclusion: The SCs were enriched from the testicular tissues of prepubertal New Zealand rabbits by a simple and effective protocol, which provides a basis for further theoretical researches and practical applications. Objective: The research was conducted to study the effect of a complex antimicrobial drug with an anti-inflammatory effect and an antimicrobial drug with an immunostimulating effect on the parameters of nonspecific resistance in calves. Materials and Methods: Two groups (n = 5 each) of sick calves with respiratory pathology were selected for this study. For the treatment of the first experimental group, a complex antimicrobial drug Sulfetrisan® was used. The second experimental group of the calves was intramuscularly injected with the experimental drug gentaaminoseleferon (GIA). To assess the cellular component of immunity in the blood before and after treatment, the number of white blood cells, T-lymphocytes, B-lymphocytes, phagocytic activity of leukocytes, phagocytic number, and phagocytic index (PhI) were determined. In addition, for assessing the humoral component, serum complement activity (SCA), serum lysozyme activity, serum bactericidal activity (SBA), circulating immune complexes (CIC), and total immunoglobulins (total Ig) were measured. The results were compared with the baseline parameters of healthy calves of the control group. Results: When studying the parameters of the humoral and cellular components of nonspecific resistance, it was found that in sick animals, compared with healthy ones, respiratory pathology was accompanied by an imbalance in the immune system. In the process of recovery in animals of the experimental groups under the effect of the drugs, positive changes occurred. However, many of the studied parameters did not reach the values of healthy animals. In the group of calves that received GIA, compared with the calves given Sulfetrisan®, a significant increase in PhI (p < 0.05), SBA (p < 0.006), SCA (p < 0.05), total Ig (p < 0.0005), and CIC (p < 0.05) was observed, which indi¬cated an increase in natural resistance due to the immunostimulating action. Conclusion: The use of GIA in sick animals added to an increase in the general nonspecific cellular and humoral resistance of calves, which made it possible to increase therapeutic efficacy and shorten their recovery time. Objective: The current study was undertaken to evaluate the seasonal dependency and prevalence of gastrointestinal roundworms (nematodes) infecting domestic fowls (Gallus gallus domesticus) in Kashmir. Materials and Methods: From August 2017 through July 2019, the investigation was undertaken during each of the four seasons. We tested 400 guts obtained from varied places around the Kashmir valley for nematode infestation. The nematodes found within the digestive tract were collected and identified using a variety of identification keys under the microscope. Statistical Package for the Social Sciences was used to analyze the data (version 20). Chi-square (χ2) test was carried out to analyze the sample data. Results: 196 hosts were infected with various nematodes, indicating an overall prevalence of 49% (196/400). The findings revealed that the prevalence of Ascaridia galli was 32.97% (61/185) in the first year and 35.34% (76/215) in the second year. Heterakis gallinarum had a prevalence rate of 20.80% (38/185) in the first year and 24.18% (52/215) in the second year, whereas Capillaria spp. had a prevalence rate of 10.81% (20/185) in the first year and 12.55% (27/215) in the second year. The overall prevalence of A. galli was determined to be 34.25% in both years (August 2017July 2019), with a mean intensity of 4.86. Summer months had the highest parasitic load. Heterakis gallinarum had a prevalence rate of 22.5% and a mean intensity of 26.83. Summer was shown to have the most considerable parasitic burden. Capillaria spp. had an overall prevalence of 11.75% and a mean intensity of 4.59; autumn had the highest parasite load. The most abundant species was identified as A. galli. It was shown that there is a significant (p < 0.01) link between seasonal¬ity and helminth parasite prevalence. Conclusion: The studys findings indicate that these gastrointestinal nematodes are ubiquitous throughout the year, but are particularly abundant in the summer and fall seasons among domes¬tic poultry in the study area. This study on the prevalence of gastrointestinal nematodes in Gallus gallus domesticus demonstrates the seasonality of infection rates and also offers various methods and techniques for framing effective strategies for controlling these helminthes to maximize profit from backyard chicken farming. Objective: This research investigated the immunomodulatory potentials of two medicinally important wild epiphytic Bangladeshi orchids Cymbidium aloifolium and Papilionanthe teres using Swiss albino mice. Materials and Methods: Orchid extracts were prepared using a cold methanol extraction proce¬dure. To assess the immunomodulatory action, Swiss albino mice of either sex weighing 2535 gm were divided into five groups each with six animals. Sheep red blood cells (SRBC) of 0.5 × 109 cells/ml were used to immunize all mice on the 7th day, and a booster dose of the same quan¬tity of SRBC was given on the 11th day of the experiment. After 14 days of oral treatment with 100 and 200 mg/kg bw of orchid extract, the mice were sacrificed to collect serum and organs. Hematological assays, delayed type of hypersensitivity assays, phagocytic index (PI), and histo¬pathological investigations were used to assess in vivo immunomodulatory efficacy. Results: The body weight changes of the experimental animals were considerably greater at 100 mg/kg bw than at a higher dose (200 mg/kg bw). There was a substantial improvement of relative organ weights of the thymus and spleen at the low dose, but no effect on kidney weights was evi¬dent. The liver weight increased significantly (p < 0.05) at both doses. Total neutrophil, leukocyte, and lymphocyte counts, hemoglobin percentage, delayed hypersensitivity reaction, and PI were all significantly (p < 0.05) increased in mice receiving the lower dose. In contrast to the control group, the higher dose reduced immunological response, suggesting the negative influence of a higher dose of extracts on the immune reaction. Conclusions: The results demonstrate that orchid extracts can potentially modulate the innate immune system in the experimental animal. [J Adv Vet Anim Res 2021; 8(3.000): 479-488]

Objective: This work aimed to detect the frequency of pathogenic bacteria of zoonotic impor¬tance in ducks dropping, their surrounding environment, and farmworkers in contact with them. Furthermore, the susceptibility pattern of isolated bacteria to antimicrobial drugs and the effi¬ciency of disinfectants (CID 20, Durak® plus, and hydrogen peroxide (H2O2), nano zinc oxide (ZnO NPs), and hydrogen peroxide loaded nano zinc oxide (H2O2/ZnO NPs) composites against isolated bacteria were evaluated. Materials and Methods: A total of 271 samples were collected from duck pens, including 35 fecal droppings, 200 environmental samples, and 36 from the hands of pen workers for isolation and identification of bacterial strains using standard microbiological procedures. After that, the antibi¬otic sensitivity testing of 40 bacterial isolates was carried out using disk diffusion assay. ZnO NPs and H2O2/ZnO NPs were characterized using Fourier-transform infrared spectrum and high-reso-lution transmission electron microscopy. The efficacy of disinfectants and nanocomposites was evaluated against enteropathogenic bacteria using the broth macro-dilution method. Results: The results showed that the overall prevalence of pathogenic bacteria in duck pens was 62.73. The highest isolation rate was detected in duck fecal droppings (100%), while Escherichia coli was found to be the most isolated pathogen (56.47%), followed by Pseudomonas aeruginosa (21.8%), Proteus mirabilis (15.29), and Salmonella species (6.47%). Multidrug resistance (MDR) was detected in the majority of bacterial isolates. The efficiency of CID 20 and Durak® plus dis¬infectants against all bacterial isolates was highly susceptible (100%) after 120 min of exposure time compared to the effectiveness of H2O2 on enteropathogenic bacteria which did not exceeded 60% at 5% concentration. Meanwhile, the sensitivity of Salmonella spp. to Durak® plus did not exceeded 80%. Conclusion: The duck fecal droppings are the primary source of bacterial isolates. MDR isolates were susceptible to both CID 20 and Durak® plus disinfectants after 120 min of exposure time at a concentration of 1:100 ml. Besides, H2O2/ZnO NPs composite proved its lethal effect against all testing strains at 0.02 mg/ml after 120 min of exposure. Strict biosecurity guidelines are required to mitigate and prevent the transmission of potentially zoonotic pathogens through the farm envi¬ronment and/or duck droppings.
