Water-based gas purge microsyringe extraction coupled with liquid chromatography for determination of alkylphenols from sea food Laminaria japonica Aresh النص الكامل

A novel organic solvent-free mode of gas purge microsyringe extraction, termed water-based gas purge microsyringe extraction, was developed. This technique can directly extract target compounds in wet samples without any drying process. Parameters affecting the extraction efficiency were investigated. Under optimal extraction conditions, the recoveries of alkylphenols were between 87.6 and 105.8%, and reproducibility was between 5.2 and 12.1%. The technique was also used to determine six kinds of alkylphenols (APs) from samples of Laminaria japonica Aresh. The OP and NP were detected in all the samples, and concentrations ranged from 26.0 to 54.5ngg⁻¹ and 45.0–180.4ngg⁻¹, respectively. The 4-n-butylphenol was detected in only one sample and its concentration was very low. Other APs were not detected in L. japonica Aresh samples. The experimental results demonstrated that the technique is fast, simple, non-polluting, allows for quantitative extraction, and a drying process was not required for wet samples. Since only aqueous solution and a conventional microsyringe were used, this technique proved affordable, efficient, and convenient for the extraction of volatile and semivolatile ionizable compounds.

Seaweeds are rich in bioactive compounds which have well-documented antioxidant properties. They also have antimicrobial activities against food pathogenic microorganisms. This study uses an extract of the brown seaweed, Saccharina (Laminaria) japonica, produced by subcritical water hydrolysis (SWH) for investigating its potential to inhibit bacteria. De-oiled S. japonica was obtained by supercritical carbon dioxide extraction. The reaction temperatures for hydrolysis of raw and de-oiled S. japonica were maintained from 200 to 280 °C. The experiment was done with condition 1.3–6.0 MPa for the reaction pressure and 1:10 (w/v) for the ratio of material to water. The antibacterial activities of raw and de-oiled S. japonica produced by SWH were determined by using the agar diffusion method. Antibacterial activity was tested against two Gram-negative (Escherichia coli and Salmonella typhimurium) and two Gram-positive bacteria (Staphylococcus aureus and Bacillus cereus). The antibacterial activities of hydrolysate water with catalyst at 240 °C showed better bacterial inhibition than the others. Strong antibacterial activity was found using de-oiled material with acetic acid added, with a zone of inhibition of S. typhimurium (14.33 ± 0.06 mm) and E. coli (13.00 ± 0.09 mm). On the other hand, the weakest antibacterial inhibition was found for S. aureus (12.83 ± 0.10 mm) and B. cereus (12.50 ± 0.09 mm).

Enhanced hydrolysis and methane yield by applying microaeration pretreatment to the anaerobic co-digestion of brown water and food waste النص الكامل

Microaeration has been used conventionally for the desulphurization of biogas, and recently it was shown to be an alternative pretreatment to enhance hydrolysis of the anaerobic digestion (AD) process. Previous studies on microaeration pretreatment were limited to the study of substrates with complex organic matter, while little has been reported on its effect on substrates with higher biodegradability such as brown water and food waste. Due to the lack of consistent microaeration intensities, previous studies were not comparable and thus inconclusive in proving the effectiveness of microaeration to the overall AD process. In this study, the role of microaeration pretreatment in the anaerobic co-digestion of brown water and food waste was evaluated in batch-tests. After a 4-day pretreatment with 37.5mL-O2/LR-d added to the liquid phase of the reactor, the methane production of substrates were monitored in anaerobic conditions over the next 40days. The added oxygen was consumed fully by facultative microorganisms and a reducing environment for organic matter degradation was maintained. Other than higher COD solubilization, microaeration pretreatment led to greater VFA accumulation and the conversion of other short chain fatty acids to acetate. This could be due to enhanced activities of hydrolytic and acidogenic bacteria and the degradation of slowly biodegradable compounds under microaerobic conditions. This study also found that the nature of inoculum influenced the effects of microaeration as a 21% and 10% increase in methane yield was observed when pretreatment was applied to inoculated substrates, and substrates without inoculum, respectively.

Annexes not included | This study examined on-going processes of climate change and adaptation strategies in six village development committees (VDCs) along the Gandak transect of central Nepal. Administrative boundaries (VDC’s) and hydrological sub-watershed boundaries do not correspond. Using participatory GIS, the research team updated maps of all six study VDCs and produced a bathymetric survey of Rupa Lake. The Mid-Hills ecological region, most vulnerable to climate change was the focus of the study. In assessing vulnerability, researchers identified crucial questions: “At what scale will climate change impacts occur?” “Where will they be most severe?” and “Who will be most affected by them?”

The objective of this work was to study the microbial community and reactor performance for the anaerobic co-digestion of brown water and food waste in single- and two-phase continuously stirred tank reactors (CSTRs). Bacterial and archaeal communities were analyzed after 150days of reactor operation. As compared to single-phase CSTR, methane production in two-phase CSTR was found to be 23% higher. This was likely due to greater extent of solubilization and acidification observed in the latter. These findings could be attributed to the predominance of Firmicutes and greater bacterial diversity in two-phase CSTR, and the lack of Firmicutes in single-phase CSTR. Methanosaeta was predominant in both CSTRs and this correlated to low levels of acetate in their effluent. Insights gained from this study would enhance the understanding of microorganisms involved in co-digestion of brown water and food waste as well as the complex biochemical interactions promoting digester stability and performance.

Anopheles gambiae larvae have frequently been observed to dive, but the ecology of this behavior has not been extensively examined. We manipulated food level, water depth, and temperature for individually-reared larvae and observed diving activity. Larvae dived more often under low food, which suggests that they dive to forage. There was only weak evidence for effects of water depth or temperature on diving. Experimental results are discussed in the context of energy budgets. Understanding larval ecology of this species is important for predicting how it will respond to environmental change. Further study is needed to assess the role that larval diving plays in both feeding ecology and thermal regulation of this and other medically important species.

Disease models which take explicit account of heterogeneities in the risk of infection offer significant advantages over models in which the risk of infection is assumed to be uniform across all hosts. However, estimating the incidence rate (force of infection) in the different at-risk (exposure) groups is no easy matter. Classically, epidemiologists differentiate groups of hosts with different infection-risks according to their exposure to putative explanatory risk factors. The importance of these risk factors is assessed by case-control studies, in which the measure of effect (the difference in disease occurrence between one population and another) is the odds ratio. This paper describes for the first time how – and under what circumstances – the incidence in these different exposure groups can be estimated from odds ratios derived from case control studies in which controls have been selected by density sampling. This new estimation technique can be applied to any transmission modality but is especially useful in the case of models of food- and water-borne disease for which the case control literature represents a vast and, as yet, untapped resource. The paper finishes with a worked example using one of the most common of all food- and water-borne pathogens, Toxoplasma gondii.

An on-line flow injection pre-concentration-flame atomic absorption spectrometry method was developed to determine trace zinc in water (tap, dam, and well water), biological (hair and nail), and liver samples. As a solid phase extractant, a synthesized new chelating resin, poly(2-thiozylmethacrylamide-co-divinylbenzene-co-2-acrylamido-2-methyl-1-propane sulfonic acid) was used. The resin was characterized by Fourier transform infrared spectroscopy, elemental analysis, and surface area by nitrogen sorption. A pre-concentration factor of 40-fold for a sample volume of 12.6mL was obtained by using the time-based technique. The detection limit for the pre-concentration method was found to be 2.2μgL⁻¹. The precision (as RSD,%) for 10 replicate determinations at the 0.04μgmL⁻¹ Zn concentration was 1.2%. The calibration graph using the pre-concentration system for zinc was linear with a correlation coefficient of 0.998 in the concentration range from 0.005 to 0.05μgmL⁻¹. The applicability and accuracy of the developed method were estimated by the analysis spiked water, biological, liver samples (83–105%), and also certified reference material TMDA-70 (fortified lake water) and SPS-WW1 Batch 111-Wastewater. The results were in agreement with the certified values.

Hydrothermal (HT) reactions of agricultural and food-processing waste have been proposed as an alternative to conventional waste treatment technologies due to allowing several improvements in terms of process performance and energy and economical advantages, especially due to their great ability to process high moisture content biomass waste without prior dewatering. Complex structures of wastes and unique properties of water at higher temperatures and pressures enable a variety of physical–chemical reactions and a wide spectra of products. This paper’s aim is to give extensive information about the fundamentals and mechanisms of HT reactions and provide state of the research of agri-food waste HT conversion.
