Strain characterization of selected isolates of sugarcane mosaic virus

Sugarcane mosaic is one of the most widely distributed diseases of sugarcane caused by four species of Potyvirus. Sugarcane Mosaic Potyvirus (SCMV) has been found to infect a wide range of hosts such as sugarcane and oil palm. Mechanical inoculation of virus-infected sugarcane and oil palm on sorghum was performed. Total RNA was extracted from field collected symptomatic sugarcane and oil palm, and mechanically inoculated sorghum using CTAB and TRizol method. CDN was synthesized using M-MLV RT and amplified using virus-specific primers (SCMV F3, SCMV R3). Amplicon was digested using restriction enzymes Apo1, Hinf1, Taq1, and Xba 1. Preliminary results on strain identification of restriction fragment length polymorphism of amplification products show variation among isolates.
