The "Gallina Valenciana de Chulilla" is an endangered autochthonous chicken breed from the Comunidad Valenciana which is currently being preserved. The objective of this study was to compare the in vitro quality (sperm motility and plasma membrane integrity) of sperm cryopreserved with two different protocols (each of them used a different cryoprotectant: glycerol or dimethylacetamide-DMA). Samples cryopreserved with glycerol exhibited higher in vitro sperm quality after cryopreservation than samples cryopreserved with DMA (67.5+/-1.7% vs. 25.4+/-1.7% total motile sperm and 80.7+/-1.8% vs. 39.9+/-1.8% sperm with intact plasma membrane). In conclusion, the protocol where glycerol was used preserved better the in vitro sperm quality after thawing than the DMA protocol.
