Agrobacterium tumefaciens-mediated transformation and regeneraqtion of carnation (Dianthus caryophyllus L.)
2001
Rafiqi, M.
The work described in this thesis comprises the recovering of transgenic carnation (Dianthus caryophyllus L.) plantlets on Kanamycin-containing medium after co-cultivation of leaf explants with armed strain of Agrobacterium tumefaciens containing a vector pBINPLUS carrying the neomycin phosphotransferase II (NPTII) and Ö-glucuronidase (GUS) genes. Transgenic procedure andpatterns of GUS expression in transgenic plants are described. Additional experiments were carried out to investigate factors affecting the regeneration of transgenic shoots. The GUS activity was easily determined histochemically on the regenerated shoots of different age
