Maize Ac/Ds controlling elements a molecular viewpoint

English. The molecular basis of a Ds controlling element mutation at the Adh1 locus of maize is related to the genetic properties of the Ac/Ds controlling element system. The Ds mutation is due to the insertion of a short DNA sequence located in the 5' non-coding region. The sequence of this element, its effect on transcription and the production of reversion mutations by its excision are outlined. Other Ds controlling elements of maize differ substantially in their internal sequences and it is proposed that the only common property of different families of Ds elements may be the presence of a specific terminal 11 bp inverted repeat sequence. The potential of the Ac/Ds elements in gene identification and gene transfer is considered.

Italian. La base molecolare della mutazione al locus Adh1 indotta da Ds e in relazione alle proprieta genetiche del sistema di controlling elements Ac/Ds. La mutazione indotta da Ds e dovuta all'inserzione di una corta sequenza di questo elemento, il suo effetto sulla trascrizione e la produzione di mutazioni da reversione attraverso la sua excisione. Altri elementi Ds del mais differiscono sostanzialmente nelle sequenze interne; viene proposto che la sola proprieta comune a differenti famiglie di elementi Ds possa essere la presenza di una specifica sequenza di 11 basi ripetuta in senso inverso. Vengono considerate le potenzialita degli elementi Ac/Ds nella identificazione e trasferimento di geni.
