Influence of low temperature on amino acid pool in the eggs of the silkworm, Bombyx mori

A series of experiments was conducted to determine whether the amino acid pool in the eggs undergoing diapause in the silkworm, Bombyx mori was influenced by low temperatures (5 and 0 degrees C). Amounts of alanine (Ala), proline (Pro) and glutamate (Glu) were different among the eggs kept at different temperatures. In the eggs continuously kept at 25 degrees C after oviposition, the amounts of glu and Pro increased until day 160 after oviposition and reached a level of 20 micro-moles/g eggs. Ala accumulated at a level of 55 micro-moles up to day 30 after oviposition, and then the level increased gradually. Chilling (5 or 0 degrees C) of the eggs induced a decline in the Ala content during diapause maintenance. In the eggs that had completed the diapause termination as were transferred to 5 degrees C, the decrease continued but was limited in the diapausing eggs transferred to 0 degrees C. On the other hand, the Glu content decreased around the diapause termination in the eggs transferred to 5 degrees C, but continued to increase in the diapausing eggs kept at 0 degrees C. In contrast, the level of Pro accumulation was higher in the eggs kept at 5 degrees C than at 0 degrees C. These results suggest that a low temperature of 0 degrees C but not 5 degrees C suppresses the metabolic pathway of Glu to Pro in Bombyx eggs
