Chinese. 探讨常温下适宜箭杆杨试管苗腋芽增殖、生根的最佳培养条件。以MS培养为基本培养基,附加不同浓度的吲哚丁酸(IBA)、萘乙酸(NAA)以及6-氨苄基腺嘌呤(6-BA)组成试验培养基,对箭杆杨组织进行培养,观察其生根情况和愈伤组织诱导情况。箭杆杨脱分化最佳培养基为:MS+1.0 mg/L 6-BA+0.1 mg/L IBA,该条件下的愈伤组织诱导率为90%;不定芽分化诱导培养基为:1/2MS+0.1 mg/L IBA+0.5 mg/L 6BA,该条件下的诱导分化率为67.1%;芽增殖培养基为:MS+1.0 mg/L 6-BA+0.1mg/L IBA,该条件下的增殖数为5.8;最佳生根培养基为:1/2MS+1.0 mg/L IBA,该条件下的生根率为81.3%。建立了箭杆杨组织培养再生体系,为其种质资源的试管保存奠定了基础。
Show more [+] Less [-]English. The study is to explore the best culture condition of axillary buds mutiplication and rooting in Populus nigra var thevestina tissue culture under normal temperature. With the basic MS medium added with different concentrations of IBA, NAA and 6-BA, the rooting and callus induction were observed. The best medium for dedifferentiation was MS + 6-BA 1.0 mg/L + IBA 0.1 mg/L, the frequency of callus induction was 90%; The ideal medium for induction and differentiation of the callus was 1/2MS + IBA 0.1 mg/L + 6-BA 0.5 mg/L, induction and differentiation rate was 67.1%; The medium for shoot proliferation was MS + 6-BA 1.0 mg/L + IBA 0.1mg/L, proliferation number was 5.8; The optimum medium for producing roots was 1/2MS + IBA 1.0 mg/L with 81.3% rooting ratio. The tissue culture regeneration system was established, which laid a basis for the storage invitro of germplasm resources.
