Multiple shoot induction and plant regeneration of Feronia limonia (L.) Swingle

An in vitro regeneration system was developed for propagation of Feronia limonia (L.) Swingle. Axillary shoot proliferation was induced in the nodal segments on Murashige and Skoog (MS) medium supplemented with N6-benzylaminopurine/kinetin/thidiazuron (BAP/KN/TDZ). Maximum number of shoots (11.33 shoots per explant) were developed on a medium containing 5 μM BAP. Adventitious shoot regeneration occurred from leaf explants on MS media containing BAP/TDZ. The best response in terms of frequency of shoot regeneration (100%) and number of shoots per explant (7.33 shoots per explant) was observed on medium supplemented with 5 μM BAP. A proliferating shoot culture was established by repeatedly sub-culturing the original nodal and leaf explants on shoot multiplication medium after each harvest of newly formed shoots. Thus, 20–32 shoots were obtained in 5 months from a single nodal or leaf explant. Rooting was induced in 83.3% of the regenerated shoots on a half-strength MS medium supplemented with 1 μM α-naphthaleneacetic acid (NAA). The plantlets were acclimatized and established in soil.
