Formation of five classes of morphological mutants of Penicillium camembertii at a high frequency by the action of nitrous acid

Spores of Penicillium camembertii were mutated with nitrous acid. A very high portion of the resulting colonies showed morphological alterations. These morphological mutants appeared at a frequency of 2.35%, with five main classes of variation, namely: flat 0.53%; wrinkled 0.45%; cross 0.23%; powdery 0.76%; and compact 0.38% according to their morphological characteristics. After subculturing of the mutants on medium containing the haploidizing agent p-fluoro-DL-phenylalanine, reversion to the wild type morphology could be observed within nearly all classes of mutants except compact. Penicillium camembertii strains not treated with nitrous acid showed no morphological alterations even when a high number of single spore colonies were checked. These results suggest that chromosomal mutations instead of point mutations cause the morphological alterations and that nitrous acid has in addition to its deaminating activity an effect on the chromosomes of P. camembertii.
