Optimization of cryoprotectants for improved survival rates of animal probiotics Lactobacillus plantarum, Bacillus subtilis, and Saccharomyces cerevisiae

Three probiotics, Lactobacillus plantarum, Bacillus subtilis, and Saccharomyces cerevisiae isolated from chicken cecum were investigated by adding various cryoprotectants to improve viability after both frozen-storage and freeze-drying. During frozen-storage at -20°C, glycerol provided a higher survival rate (15.8–58.3%) than other cryoprotectants for the probiotics. The effect of the cryoprotectants decreased with an increased storage time of 30 days. Bacillus subtilis preserved in a 1:1 mixture of glycerol and mineral oil showed a higher survival rate of 38.1% compared to glycerol only with a survival rate of 30.95%, demonstrating the synergistic effect of the cryoprotectants. The relationship between cryoprotectants and the survival rate at -80°C freeze-drying was also studied. An effective cryoprotectant for L. plantarum was found to be 20% soy peptone/ 1% lactose with a survival rate of 80%. In B. subtilis, 10% soy peptone/5% lactose presented effective synergistic cryoprotectants with a survival rate of 92.9%, whereas in S. cerevisiase, 10% soy peptone 1% mannose showed a survival rate of 52.2%. In conclusion, we demonstrated that an optimization of the ideal proportion of the cryoprotectants glycerol and soy peptone to sugar increased cell survivability for storage according to the respective probiotics.
