Innovative anthelmintic based on mechanochemical technology and their efficacy against parasitic infection of sheeps Full text

Objective: Solubility and bioavailability are crucial for maximizing the activity of an antiparasitic drug. This study aimed to develop a combined preparation for antiparasitic medicines using iver¬mectin (Iver), fenbendazole (FBZ), and triclabendazole (TBZ), considering their solubility, bioavail¬ability, and activity. Materials and Methods: Innovative preparations in solid dispersions (SD) were obtained using the joint mechanical processing of drug substances with polyvinylpyrrolidone (PVP) in an LE-101 roller mill. The preparations efficacy was studied in 140 sheep spontaneously infected with gastroin¬testinal Strongylata, Dicrococelium dendriticum, Moniezia expansa, and Melophagus ovinus. The preparations were given individually to the sheep in the form of an aqueous suspension orally. Their effectiveness was evaluated using intravital and postmortem parasitological examinations. Results: The results confirmed the increase in solubility of substances by 1329 times. The exper¬iments have shown the high efficacy of SD composition of FBZ/Iver/PVP (1/1/9) containing FBZ (at 3.0 mg/kg b/w) and Iver (at 0.2 mg/kg b/w) when used against gastrointestinal Strongylates and M. expansa (95.8% and 100%, respectively), to a lesser extent against M. ovinus (38.5%). The SD composition of TBZ/Iver/PVP (1/1/9) of TBZ (at 3.0 mg/kg b/w) and Iver (at 0.2 mg/kg b/w) showed a high efficacy against gastrointestinal Strongylata and D. dendriticum (96.8% and 100%, respectively) and less activity against M. ovinus (61.6%). Conclusion: The high parasiticidal activity of SD based on FBZ, TBZ, and Iver in comparison with initial substances is explained by the formation of inclusion complexes of these substances with PVP when SD is dissolved in water and the synergistic effect of the active substances of the prepa¬rations. The resulting complexes have increased solubility in water and bioavailability. The use of such an SD suggests a significant reduction in the dosages of FBZ and TBZ without losing parasit¬icidal activity. [J Adv Vet Anim Res 2020; 7(4.000): 718-725]

Objective: This study aimed to isolate and identify of pathogenic bacteria in tiny freshwater shrimp (Macrobrachium lanchesteri) and in Kung Ten, which is an unusual Thai cuisine that eaten alive shrimp directly. Antimicrobial susceptibility test and identification of antibiotic resistance genes for isolated bacteria were conducted. Materials and Methods: Eighty of fresh shrimp samples and forty of Kung Ten salads were collected from four fresh markets, which were located in Bangkok and Nonthaburi province (N = 120). The isolation, identification, and antimicrobial susceptibility test of pathogenic bacteria were done following the Clinical and Laboratory Standards Institute guidelines. Antibiotic-resistant bacteria were screened for β-lactamase relating genes, such as AmpC (MOX and ACC genes), blaCTX-M, and Int1 genes. Results: The number of bacterial isolates in tiny freshwater shrimp and Kung Ten salad was 136 and 65, respectively. Aeromonas caviae, A. hydrophilla, Proteus penneri, Proteus vulgaris, and Klebsiella pneumoniae were commonly found. Ampicillin, amoxicillin/clavulanic, cefuroxime, tet¬racycline, and trimethoprim/sulfamethoxazole resistance were observed, and common antibiot¬ic-resistant bacteria were A. caviae, P. vulgaris, Enterobacter Aerogenes, and K. pneumoniae. A. caviae, P. penneri, K. Pneumoniae, and A. hydrophilla were positive for MOX gene; blaCTX-M, and Int1 genes; ACC and Int1 genes; and ACC gene, respectively. Conclusion: Raw or uncooked shrimps in Kung Ten salad may a risk in foodborne diseases due to positive for pathogenic bacterial isolates. However, hygienic control on food preparation is diffi¬cult to apply because of the difficulty of changing in local Thai food behavior. [J Adv Vet Anim Res 2020; 7(1.000): 83-91]

Objective: Understanding intraspecific phenotypic plasticity is a prerequisite to fish stock identifi¬cation and sustainable fisheries management. In this study, we assessed intraspecific phenotypic plasticity in terms of meristic and morphometric characters of wild populations of elongate glassy perchlet, Chanda nama from two different rivers, namely Madhumati River Narail (MRN) and Tulshiganga River Jaypurhat (TRJ), and an ox-bow lake, Jhapa Baor Jashore (JBJ) in Bangladesh. Materials and Methods: In this study, six meristics, 15 conventional morphometrics, and 23-truss-based morphometrics were subjected to a one-way analysis of variance (ANOVA), followed by the Tukey-HSD test. The mean values of three meristic counts and nine conventional and 12 truss-based morphometrics demonstrated significant differences in the ANOVA test. Furthermore, principal component analysis (PCA) and discrimination function analysis (DFA) were performed separately using conventional and truss-based morphometric data. Results: In PCA analysis, four principal components were extracted and cumulatively demon¬strated 51.848%. On the contrary, two discriminant functions (DF1, 63.5%; DF2, 36.5%) resulted from DFA analysis. In the bi-plot alignment from the discriminant space, all individuals were exceedingly separated among the three inhabitants. A dendrogram developed using conventional and truss morphometric characters confirmed that two clusters were formed among three pop¬ulations. The TRJ population formed a distinct cluster, and the JBJ population formed a different cluster with a subcluster of MRN. In the discriminant function analysis, precise classification out¬comes displayed 82% of individuals into their unique populations, whereas 66.9% of individuals were categorized as a cross-validated assemblage. Conclusion: The baseline information resulting from the current study would be useful for envi¬ronmental studies and further conservation of glassy perchlet populations in Bangladesh. Objective: The purpose of the experiment was to examine the effect of different food sources on the growth and reproduction performances of an epigeic earthworm Perionyx excavatus. Materials and Methods: The experiment was carried out in 18 cylindrical plastic containers for 10 weeks. The study was designed with six treatments, each having three replicates. In control treat¬ment (T1), cow dung was used as the only food source for the earthworm. In another five treat¬ments, water hyacinth (T2), chopped banana plant trunk (T3), vegetable scrap (T4), paddy straw (T5), and sugarcane bagasse (T6) were used as food sources with cow dung as bedding material. Results: The maximum weight gain of earthworm P. excavatus was 3,294.7 ± 4.5 mg for the food staff of vegetable scrap (T4). Earthworm P. excavatus fed with chopped banana plant trunk (T3) showed a very similar weight gain of 3,243.7 ± 3.8 mg. On the contrary, the minimum weight gain was 1,799.7 ± 3.5 mg for the food staff of paddy straw (T5). The maximum cocoon number of 137.33 ± 6.46 mg was observed in T3, whereas a minimum number of 36.67 ± 4.16 mg in T1. The highest number of hatchlings (12.33 ± 0.88 mg) was recorded in treatment T3, whereas the least number of hatchling (5.00 ± 0.58 mg) was observed in T1. Conclusion: It was concluded that, among the six different food sources, chopped banana plant trunk was preferable food source to the earthworm for growth and reproduction. Objective: This study investigated the prevalence of Listeria monocytogenes in retail poultry shops, characterized the antibiotic resistance profile, and detected the genotypic pattern of vir¬ulence genes. Materials and Methods: Broiler meat (n = 90), intestinal content (n = 40), and environmental samples (n = 95) were collected for this study. Besides, hand swabs (n = 20) were obtained from the poultry shop workers and stool samples (n = 40) were collected from the outpatient clinics of Beni-Suef University Hospital, Egypt. The samples were subjected to isolation and identification of L. monocytogenes by conventional bacteriological examinations and biochemical tests, followed by confirmatory identification by the polymerase chain reaction. Results: Among the collected samples (n = 285), L. monocytogenes could be detected in 14.4% (n = 41/285) of the samples, where 30.0% (n = 12/40) of the intestinal content was positive. Similarly, 10.0% (n = 9/90), 15.0% (n = 3/20), and 12.5% (n = 5/40) of the samples of meat, hand swabs, and stools were found positive for L. monocytogenes, respectively. A total of 12 (12.6%) out of 95 environmental samples were positive for L. monocytogenes. Based on the antimicrobial sensitiv¬ity profile, most of the recovered isolates were multidrug-resistant against most commonly used antibiotics. Conclusion: The findings conclude that poultry shops play a vital role in transmitting L. monocy¬togenes to the consumers. Asymptomatic poultry shop workers should draw attention to their potentials for spreading the infection to the consumers through the contaminated carcasses. Several animals have been in the limelight of basic research associated with metabolic diseases like obesity. Obesity can be considered as a significant public health concern in the world. It raises the chances for a variety of disease conditions that includes diabetes, hypertension, liver disease, and cancers, which, in turn, decreases the overall lifespan of adult men and women. The World Health Organization has considered obesity as a global epidemic. Researchers have made several attempts to classify human obesity, but none have been successful. Animal obesity can be classi¬fied based on their etiology; however, till now, no animal model of obesity can replicate models of the human condition, they have only provided clues into the causes, aftermaths, and preventive remedy to human adiposity. Over the years, there are varieties of animal models used to induce obesity. Some of them include monogenic, polygenic, surgical, seasonal, and other models of obesity. Apart from the advantages of these models, most of them are accompanied by limita¬tions. The primary purpose of this review is, therefore, to highlight the several models with their advantages and limitations. Objective: This study aimed to examine the occurrence of Caryospora (C.) neofalconis in falcons from the central region of the Kingdom of Saudi Arabia (KSA). Materials and methods: Fecal samples (n = 149) from 149 healthy falcons including 56 saker falcons (Falco cherrug), 13 lanner falcons (F. biarmicus), 18 peregrine falcons (F. peregrinus), 40 Barbary falcons (F. pelegrinoides), and 22 gyrfalcons (F. rusticolus) were collected between October 2018 and May 2019. The fecal samples were examined for the presence of C. neofalconis by microscopic examination followed by confirmation by polymerase chain reaction targeting 18S rRNA genes and their phylogenetic analyses. Results: The overall prevalence of C. neofalconis in the falcons was recorded as 10.7% (16/149) by microscopic examination. The highest prevalence was found in F. peregrinus (6/18, 33.3%), followed by F. rusticolus (3/22, 13.6%), F. cherrug (5/56, 8.9 %) and F. pelegrinoides (2/40, 5.0%). There was no C. neofalconis infection observed in F. biamicus. The 18S rRNA gene could be ampli¬fied in eight samples. The phylogenetic analysis of two C. neofalconis isolates exhibited a close relationship with the Mexican isolate (KT03081) with a 99.7% identity. Conclusions: To our knowledge, based on the microscopic and molecular analysis, this is the first report of C. neofalconis in F. cherrug, F. rusticolus, F. pelegrinoides, and F. peregrinus from the central region of the KSA and it emphasize the value of adopting preventive measures to limit the spread of C. neofalconis. Objective: The objective of this study was to assess the veracities of most admired strategy dis¬criminant analysis (DA), in comparison to the artificial neural network (ANN) for the anticipation and classification of milk production level in Holstein Friesian cattle using their performances. Materials and Methods: A total of 3,460 performance records of imported and locally born Holstein Friesian cows were gathered during the period from 2000 to 2016 to compare two alter¬native techniques for predicting the level of production based on performance traits in dairy cattle with the use of statistical software (Statistical Package for the Social Sciences, version 20.0). Results: The findings of the comparison indicated that ANN was more impressive in the expec¬tancy of milk production level than did an imitator statistical method based on DA. The accuracy of the ANN model was high for the winter season (79.5%), whereas it was 47.3% for DA. The cur¬rent findings were assured via the areas under receiver operating characteristic curves (AUROC) for DA and ANN. AUROC curves were smaller in the condition of the DA model across different calving seasons compared with the ANN model. The inaccuracies of variations were significant at a 5% significance level utilizing paired sample t-test. Objectives: The present study estimated the seroprevalence of avian reovirus (ARV) infections in backyard chickens of the Mymensingh district in Bangladesh. Materials and Methods: Considering several risk factors, a total of 460 serum samples were col¬lected from backyard chickens from eight Upazilas of the Mymensingh district in Bangladesh. Blood samples were taken from the wing vein using 3-ml sterile syringes and kept at room tem¬perature for clotting in a slanting position and then transported to the laboratory maintaining the cool chain. Subsequently, the prepared sera were harvested and stored at −20°C until used. Finally, an indirect enzyme-linked immunosorbent assay (ELISA) was performed to detect ARV-specific antibodies using a commercial ARV antibody detection ELISA test kit. Results: The results revealed high prevalence rates of ARV antibodies, with a total seroprevalence of 69.78% (321/460). Area-wise, 74.55% (82/110) seroprevalence was recorded as the highest in Mymensingh Sadar, whereas 64% (32/50) was the lowest in Gauripur Upazila. With regard to sex, female chickens showed a significantly higher (p < 0.05) seroprevalence as 90.33% (271/300) compared to male chickens 31.25% (50/160). With regard to age groups, the seroprevalence of ARV infection was 59.33% (89/150) within 28 weeks, 82% (205/250) within 916 weeks, and 45% (27/60) within 1720 weeks, respectively. Based on hygienic conditions, the highest sero¬prevalence of ARV was noted in backyard chickens housed in poor conditions 80% (120/150) than good conditions 50% (40/80). Backyard chickens reared in free-ranging conditions exhibited a significantly higher seroprevalence 73.33% (220/300) of ARV antibodies compared to rearing in separate houses 63.12% (101/160). The seroprevalence of ARV was higher in crossbreeds 71.67% (43/60), brought from market 76% (38/50), and unhealthy 78.57% (55/70) backyard chickens than non-descriptive indigenous 69.5% (278/400), home-reared 69.02% (283/410), and healthy chick¬ens 68.21% (266/390). Conclusion: The high prevalence of ARV antibodies revealed in the current study indicates an extensive exposure of ARV to backyard chickens in Bangladesh that may be transmitted naturally to other chickens, ultimately leading to ominous economic effects on the poultry sector. Objective: In recent years, lead (Pb) has arisen as a foremost contaminant due to overpopulation, rapid industrialization, and expansion that could contaminate the human food chain. However, the correlation between the environmental contamination of Pb and its spatial transfer to the dairy products is still unmapped. In this paper, we intend to evaluate the concentration of environ¬mental Pb and its spatial distribution in dairy feed and products in Narayangonj, a highly polluted district of Bangladesh. Materials and Methods: A total of 125 samples of soil, water, forage, and milk were collected from five upazilas (Narayangonj Sadar, Bondor, Rupgonj, Araihazar, and Sonargoan) of Narayangonj. The samples were digested by acid digestion, and Pb was detected by flame atomic absorption spectrophotometer and graphite furnace atomic absorption spectrometer. The bioconcentration factor of the samples was also calculated. Results: In this study, 25% of the soil, 20% of the water, 5% of the forage, and 2% of the milk samples contained Pb at a variable level. Among the environmental samples, the highest concen¬tration (2639 μg/kg) of Pb was detected in the soil, followed by in the water (0.0230.059 μg/ kg) and forage (0.0170.035 μg/kg). The contamination (0.0410.068 μg/kg) in the milk, however, was lower than the soil but higher than the water and forage. The concentration of Pb in all the samples was within a safer limit. None of the forage samples was the potential bioaccumulator. Conclusion: Although no linear correlation was established between the environmental samples, forage, and milk, the study identifies the potentials of the spatial distribution of Pb from the environment to the dairy feed and products.
