Factors affecting hematological profiles in three Ethiopian indigenous goat breeds

Hematological parameters were studied in 163 apparently healthy goats comprised of 3 indigenous breeds at Adami-Tulu Agricultural Research Centre (ATARC) and Sheno, Agricultural Research Centre (SARC), Ethiopia. Data were analyzed for the effect of breed, sex, age, and season. Hemoglobin (Hb) was significantly higher (P < 0.0001) for Arsi-Bale (AB) than Longeared Somali (LES) and Central Highland (CH) goats. Packed cell volume (PCV) was significantly lower (P < 0.0001) for LES than the other two breeds. White blood cells (WBC) (P < 0.05), neutrophils (P < 0.0001) and basophils (P < 0.0001) were higher in CH than in the other two breeds. Conversely, lymphocytes were lowest (P < 0.0001) in CH goats compared to other breeds. Female goats had significantly (at least P < 0.05) higher red blood cells (RBC), PCV, WBC, lymphocytes and eosinophils than male goats. In contrast, males had higher (P < 0.05) mean corpuscular hemoglobin (MCH) and neutrophils than females. There was no significant age difference for the erythrocytic series but the age difference was highly significant (P < 0.0001) for the leukocytic series. WBC and lymphocytes decreased as age increased while the reverse was true for neutrophils and eosinophils. Seasonal variation was also considerable for both erythrocytic and leukocytic series. RBC, Hb, lymphocytes, and neutrophils of the indigenous goat breeds were remarkably different compared to most other exotic goat breeds.

peer reviewed | OBJECTIVE: To evaluate a human assay for quantification of carboxy-terminal cross-linking telopeptide of type I collagen (CTX-I), assess the influence of age on plasma CTX-I concentration, investigate the relationship between plasma CTX-I and serum osteocalcin concentrations, and determine whether concentrations of plasma CTX-I or serum osteocalcin fluctuate in circadian manner in horses. HORSES: 75 clinically normal horses. PROCEDURE: Cross-reactivity between equine serum CTX-I and CTX-I antibodies in an automated electrochemiluminescent sandwich antibody assay (ECLIA) was evaluated via a specificity test (ie, dilution test) and recovery calculation. Serum osteocalcin concentration was measured with an equine-specific osteocalcin radioimmunoassay. To analyze diurnal variations in plasma CTX-I and serum osteocalcin concentrations, blood samples were obtained hourly during a 24-hour period. RESULTS: Results of the dilution test indicated good correlation (r > 0.99) between expected serum CTX-I concentrations and measured serum CTX-I concentrations. The calculated CTX-I recovery was 97.6% to 109.9%. Plasma CTX-I and serum osteocalcin concentrations were correlated. Plasma CTX-I concentration was inversely correlated with age of the horse. No significant circadian variations in plasma CTX-I and serum osteocalcin concentrations were detected. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the fully automated CTX-I ECLIA can be used for evaluation of plasma and serum samples from horses and may be a useful tool to monitor bone metabolism changes. Horses in this study did not have notable diurnal fluctuations in serum osteocalcin and plasma CTX-I concentrations.

Objective--To evaluate the immunologic response of a killed tachyzoite vaccine against Neospora caninum and its effectiveness in preventing fetal loss associated with experimentally induced neosporosis in sheep. Animals--30 Dorset ewes. Procedure--Ewes were randomly allocated to receive vaccination on days 1 and 60 of the study with a killed N caninum tachyzoite preparation in a commercially available adjuvant or a saline-adjuvant mixture. A ram was placed on pasture with the ewes from days 15 to 60. Blood was collected from ewes before primary and booster vaccinations and prior to experimental challenge with N caninum tachyzoite performed on day 90; sera were assessed via Neospora agglutination (NA) and immunofluorescence antibody (IFA) assays. Blood was collected from lambs before they suckled, and sera were tested for antibodies against N caninum. Results--Of the 14 vaccinated ewes that became pregnant, 12 gave birth to live-born lambs; in contrast, 5 of 11 pregnant control ewes gave birth to live-born lambs. Whereas vaccination improved fetal survival in pregnant ewes challenged with N caninum tachyzoites, it did not appear to have any appreciable effect on transmission of N caninum to offspring, as indicated by results of NA and IFA assays. Conclusions and Clinical Relevance--The N caninum tachyzoite vaccine used in this study appeared to provide protection against fetal loss associated with experimentally induced neosporosis in a high proportion of pregnant ewes.

The purpose of this in vitro study is to clarify some of the underlying mechanisms leading to the decreased migratory capacity of polymorphonuclear leukocytes (PMN) during mastitis in dairy cows soon after calving. Surface expression of Mac-1 (CD11b, CR3) on PMN and of CD14 on monocytes was measured in early- (EL), peak- (PL), and midlactation (ML) by flow cytometric analysis. In addition, we evaluated the effect of lipopolysaccharide (LPS) and tumor necrosis factor (TNF)-α on CD11b surface expression in PMN at different stages of lactation in a whole blood model. During EL, while resting monocytes expressed diminished levels of CD14, the basal expression of CD11b on PMN was not significantly altered. The relative increase of CD11b on PMN after incubation with LPS or TNF-α did not significantly differ among EL, PL, or ML at any of the concentrations tested. The current findings do not support an important role for basal CD11b levels nor for a defective mobilization of CD11b by LPS and TNF-α in the reduced migratory capacity of PMN during EL.

The influence of feeding urea-molasses blocks (UMB) on growth and gastrointestinal (GI) nematode parasitism of weaner goats grazing the same pasture was investigated on a farm in Nyandarua District, Kenya. Thirty female Small East African goat kids at an average age of 5 months were initially treated with albendazole orally (5 mg kg-1 body mass) and randomly assigned into one of two groups: group I were fed UMB prepared using a cold process and group II kids (controls) received no block supplementation (NBS). The UMB were given in the evening when the animals returned from grazing and were consumed during the night at a rate of 95.0 g head-1 day-1. Supplementation was undertaken for 3 consecutive months from July to September 2001 and January to March 2002. Body mass of the kids and faecal egg counts were measured monthly and larval cultures were performed on positive faecal samples of kids of each group. Five goats from each group were randomly selected for slaughter and total counts and identification of worms at the end of June 2002. Significant differences (P < 0.05) were found in cumulative mass gains of kids in group I from September compared with those in group II. On termination of the study kids in group I had gained an average of (+ SD) 20.4 ± 1.4 kg while those in group II had gained 11.8 + 1.1 kg. From January 2002, faecal egg counts of the kids in the UMB group differed significantly (P < 0.05) from those of the NBS group and at slaughter, the mean (+ SD) worm counts for the UMB group was 482 + 299 while that of the NBS group was 1 302 + 410. In all the goats, Haemonchus contortus was the predominant nematode recovered. These results indicate that UMB had significant effects in the control of GI nematode parasitism and enhanced growth of the young goats.

During a period of four consecutive years, trypanosomosis surveys were conducted in a tsetse-infested and tsetse-free area of the Amhara Region of north-west Ethiopia. In each study area randomly selected communal cattle were sampled and their blood was investigated using parasitological diagnostic methods. At the same time the population of biting flies was sampled. The monthly average prevalence of trypanosome infections in cattle did not differ significantly between study areas. In both study areas, the prevalence of trypanosome infections was highest during the long rainy season. Trypanosome infections were mainly due to Trypanosoma vivax and they significantly reduced the average packed cell volume and the body condition of the animals. The monthly prevalence of infection was correlated with the density of biting flies, such as Tabanidae and Stomoxys spp., in the preceding month suggesting an important role of mechanical transmission in the epidemiology of trypanosomosis in both areas.

Four trials, three in the United States and one in South Africa, were conducted to evaluate the potential value of a novel self-medicating applicator in the passive control of gastrointestinal nematodes in cattle and deer, and of flies and ticks on cattle using oil-based treatments. The results of the trials demonstrated that this applicator is an effective and practical device for the passive treatment of both deer and cattle for trichostrongyle infections using the endectocide, moxidectin (Cydectin (R) , Fort Dodge Animal Health, USA), of cattle for horn fly (Haemotobia irritans) infestations using the insecticide, cyfluthrin (CyLence (R) , Bayer AG, Germany) and of cattle for tick infestations (in particular Amblyomma hebraeum and Rhipicephalus appendiculatus) using the acaricides deltamethrin and amitraz (Delete All (R) , Intervet, South Africa).
