Features of formation of productivity of sugar beets hybrids of domestic breeding

Purpose. To reveal features of formation of productivity of modern sugar beet hybrids of domestic selection. Methods. Field, laboratory. Results. Studies show that the lowest yields of sugar beets were obtained in 2016, 2017 and 2019. Moreover, the conditions of the unstable humidification zone of the Right-Bank part of the Forest-Steppe of Ukraine were mainly characterized by unstable precipitation, a decrease in their amount, as well as increased average daily air temperatures. Therefore, the selection of high-yielding hybrids of sugar beets to obtain a high level of productivity is relevant. As evidenced by the data comparing the yield of sugar beet root crops on the test site, they significantly exceeded the average indicators for Kiev region by 10.7–20.0 t/ha, and only in arid 2016 the difference was minimal – 4.6 t/ha. The high stability of the trait “sugar content in root crops” is inherent not only in hybrids with low parameters of its manifestation, but is largely determined by the genetic characteristics of the hybrid. Three clusters were identified based on the reaction of hybrids to growing conditions. The first cluster included triploid hybrids: ‘Zluka’, ‘Oleksandriia, ‘Olzhych’, ‘Kvarta’ and ‘Konstanta’, the second one – the triploid hybrid ‘BTs ChS 57’ and diploid ‘Vesto’, the third cluster - the triploid hybrid ‘Sofiia’ and diploid ‘Ukrainskyi ChS 72’. Conclusions. Proper selection of high-yielding hybrids resistant to adverse growing conditions in the zone of unstable moisture of the Forest-Steppe of Ukraine is extremely important, because the biological characteristics of hybrids affect 45% of the amount of sugar harvest, but the growing season by 33% and the interaction of factors was 17%. It was found that despite the difficult weather conditions, the highest yields were formed by hybrids: ‘Anichka’ – 70.1 t/ha, ‘Ramzes’ – 70.5 t/ha, ‘Romul’ – 73.0 t/ha and ‘BTs ChS 57’ – 73.4 t/ha. But the maximum sugar content in root crops was in hybrids: ‘Ukrainskyi ChS 72’ – 17.3%, ‘Zluka’ – 17.3% and ‘Sofiia’ – 17.7%. Accordingly, the maximum sugar harvest was provided by hybrids: ‘BTs ChS 57’ – 12.1 t/ha and ‘Romul’ – 12.2 t/ha.

Purpose. To reveal the features of the formation of sugar beet hybrids productivity depending on fertilizer options and harvesting time. Methods. Field, laboratory. Results. Biological differences in the growth and development and the mass of root crops formation allowed high yielding hybrids to respond to the use of supplemental mineral nutrition properly, and therefore, high productivity rates were also obtained on fertilized variants. When harvesting root crops in September, the ‘Romul’ hybrid in the control variant formed 54.0 t/ha of root crops, and with fertilizer N150P150K150 and N300P300K300 – 74.2 t/ha. The delay in harvesting did not lead to a significant increase in the productivity of the investigated sugar beet hybrids. Thus, in the third decade of October, sugar beets on average formed 48.8–67.9 t/ha of root crops, and root crops had an average of 18.3–20.7% sugar. The best performance was provided by the hybrid ‘Sofiia’, so the control sugar content was 22.0%, and with the fertilizer N150P150K150 – 20.5% and N300P300K300 – 19.5%. The use of fertilizers N150P150K150 and N300P300K300 contributed to higher yields of root crops with lower sugar content, which corresponded to data obtained by other scientists. Conclusions. When harvesting in the third decade of September and being fertilized with N150P150K150, hybrids formed: ‘BTsChS 57’ – 66.8, ‘Heroi’ – 66.2, ‘Romul’ – 72.0, ‘Quarta’ – 66.0 t/ha of root crops, with fertilizer in a dose of N300P300K300 – 68.8; 68.1; 74.2 and 68.0 t/ha, respectively. It was proved that at the harvesting period in the third decade of October, ‘BTsChS 57’, ‘Heroi’, ‘Sofiia’ and ‘Romul’ were the best. In addition, the introduction of the mineral nutrition in dose N300P300K300 did not provide a yield increase that could recoup additional mineral nutrition compared to the dose N150P150K150. In terms of sugar content of the sugar beet root crops, the hybrids ‘Heroi’ – 21.4%, ‘Sofiia’ – 21.8%, ‘Ukrainskyi ChS72’ – 21.3% and ‘Romul’ – 21.4% were the best at harvesting in the third decade of September; and in the third decade of October the maximum level of sugar content in root crops was in hybrids ‘Sofiia’ – 22.0% and ‘Umanskyi’ – 21.9%. The use of mineral fertilizers led to a decrease in the sugar content in roots by about 1.3–2.5% compared with unfertilized control options. Such a reaction of plants is caused by a sufficient level of soil supply with mineral nutrition compounds.

Purpose. To create a multiplex system for identification glyphosate-tolerant sugar beet by using PCR. Methods. Molecular genetic analysis. Results. The article presents the results of studies to determine the parameters of the polymerase chain reaction (PCR) in order to develop a multiplex system for identification of the structural elements of the design of transgenic gene cp 4 epsps, which provides tolerance to glyphosate. For amplicon target DNA sequences, the following values of temperature conditions of PCR were determined: step 1 (initial denaturation) 95 °C – 3 min; step 2 (specific reaction products accumulation): denaturation 95 °C – 45 s; hybridization of primers 55 °C – 50 s; elongation 72 °C – 1 min; number of cycles – 40; step 3 (final elongation) 72 °C – 6 min. A series of PCR were carried out for the purpose of selecting the optimal amount of DNA matrix for efficient estimate of transgenic sugar beet plants for the presence of specific sequences. Conclusions. To identify transgenic glyphosate-tolerant sugar beet, it is advisable to determine 35S promoter and gene cp 4 epsps in individual genotypes. It was found that during the selection of temperature parameters of multiplex reaction a 5 °C rise in primer hybridization temperature did not affect the identification of gene als that allowed to include specific primers for determination of this sequence as an internal control. Based on the results of test multiplex reactions, concentrations of dNTPs and Mg2+ ions were determined that allowed to exclude the possibility of non-specific fragments and false-negative results. The optimum amount of matrix DNA (100–150 ng) for an efficient estimate of transgenic sugar beet plants for the presence of specific sequences was determined. Obtained results allowed to develop a multiplex test system for identification of transgenic glyphosate-tolerant sugar beet which can be used for simultaneous determination of the 35S promoter, cp 4 epsps gene and als gene as an internal reaction control.

Sugar yield, an important indicator, is resulting from interaction of two components: the yield and sugar content, both they are controlled by polygenes, and influenced significantly by environment. A.V. Kilchevskyi and L.V. Hotyliova pointed to the fact that in breeding, contrary to evolution, stabilizing forms are dominated by driving ones, which determine the growth response to regulated factors and decreased resistance to non-regulated environmental factors [1]. A relevant breeding trend is to create the genotypes adequate with certain technologies, so to say «low input variety high input variety» i.e. varieties with low and high energy contribution to technology. From this perspective, selection of stable in their manifestation lines against the analyzing backgrounds has become an important element of the breeding process [2]. Home scientists, such as A.L. Mazmulov and M.S. Hrytsyk have contributed greatly to the development and application of the agro-backgrounds [3, 4].

Sugar beet hybrids are considered as a factor of intensification of the beet-sugar branch. At the modern stage of development of agriculture under condition of reduction of sugar beet sown area, shortage of material - technical resources which influence the formation of productivity of sugar beet as a crop of intensive type, hybrids must have a high genetically conditioned biological potential of productive ability, resistance to pests and diseases, abiotic stress factors, must have broad adaptive abilities. Analysis of the modern sugar beet hybrids included bately in the state Register of plant varieties of Ukraine is presented. Transfer of the branch to the intensive way of development is possible only under conditions of a complex approach to solving this problem.

Purpose. Finding ways to activate the germination of sugar beet seeds, obtaining even and synchronous sprouts when applying fertilizer compositions with nanoscale elements. Methods. Vegetation and laboratory. The seeds of sugar beet were sown in prepared utensils with soil in accordance with the requirements of the methods for vegetation experiments. Fertilizers were introduced in the form of solutions with different ratios according to six microstages. Results. At 01 microstage on the BBCH scale (130 hours after sowing), an increase in the mass of beet fruits in all variants was observed - in the control variant by 9.78%; in the application of nanofertilizers - 20,4-23,7%. The diameter of the fruit varied similarly to changes in mass: in the control variant, the diameter change was 4.95%; in variants with application of nanofertilizers — 9.56-13.9%. Changes in the rate of sprout organs formation and their linear dimensions were noted in the various fertilization schemes. The length of the embryonic root at 05 microstage with uniform introduction of high norms of zinc and phosphorus, after 40 hours after sowing, was 0.540-2.671 mm. For other fertilizer combinations, the appearance of the germ root was noted only 44 hours after sowing. In 60 hours after sowing (07 microstage on the BBCH scale) there was a complete exit of cotyledons from the socket of the cluster with the introduction of nano chelate microfertilizers and only the beginning of the exit of cotyledons in the control variant. Due to the intensive processes of swelling and germination, the growth of the primary root of the sugar beet was accelerated. Conclusions. Uniform provision of seeds with zinc and especially phosphorus on the background of basic complex fertilizer with nanoscale elements contributed to the activation of seed germination and the intense formation of synchronously developed shoots. On average, the opening of the pericarp lid and the appearance of the root accelerated for 4 hours; 6 hours earlier there was an exit of cotyledons. With the introduction of nano chelate fertilizers, root growth and elongation of the hypocotyl at the first microstages of sugar beet sprouting were accelerated twice, due to which the sugar beet sprouts appeared 4-6 hours earlier. Nano chelate microfertilizers, promoting even and synchronous germination, development of sugar beet seedlings ensured synchronous emergence of seedlings and formation of predetermined sowing density without further reduction of plants.
