The use of cultured ovarian granulosa cells for evaluation of biological activity of hypophysical hormones

The aim of this study was to develop fast and cheap method to evaluate their biological activity of hypophysial hormones. There were assessed the effects of bGH (1-10,000 microgram/ml), LH, FSH and PMSG (all at 0.01-100 microgram/ml) on IGF-1 and progesterone secretion by cultured bovine granulosa cells. It was observed that bGH significantly (P0.05) increased both IGF-1 and progesterone release in a bell-shaped manner with maximal effects at 100 and 1,000 microgram/ml, respectively. All gonadotropins stimulated both IGF-1 and progesterone output, however significant variations between activities of different preparations were found. The observations suggest that secretory activity of cultured granulosa cells can serve as a reliable and fast indicator of biological activity of hypophysial hormones used for stimulation of animal production.
