Sterilization of diluent in sealed polypropylene plastic bottles by non-rapid cooling steam autoclave

Sterilization of diluent in sealed plastic containers by steam autoclave is dangerous, during their cooling step must use the rapid cooling system by introducing the spray of water mist; coming to contact with the hot containers, the mist absorbed latent heat of vaporization for rapid cooling. The sterilization by steam autoclave without rapid cooling and heat exchanger, an excess vapor pressure inside the hot containers will cause rupturing or deforming of containers. To prevent this effect, the sterilizer should be adjusted with an external air pressure to balance the internal air pressure during cooling step. To protect an excess pressure in containers, the sterilizing temperature should be decrease less than 121 deg C by increasing time of sterilization. The sterilizing temperature at 110 deg C for 40 minutes was suitable for sealed polypropylene plastic bottle containing 200 ml. of 2 percent tryptone. During cooling step, the pressure in chamber should not less than 1.6 bar for 60 minutes by using vacuum pump to prevent deforming of plastic bottles.
