Optimization of semen processing and cryopreservation techniques in Philippine native roosters (Gallus gallus domesticus L.)

Five experiments were carried out as an attempt to develop the best technique for processing and cryopreservation of fowl semen. The first experiment was done to evaluate several periods of equilibration and glycerol levels on the quality of rooster semen. The second experiment was an evaluation of the effects of several equilibration periods and freezing rates on the quality of freeze-thawed semen. The third experiment was conducted to evaluate the effects of glycerol level and freezing rate. The fourth experiment evaluated the effects of the different freezing rates and thawing procedure. lastly, experiment 5 was done to test the fertility of the cryopreserved rooster sperm. Results showed that the extender with glycerol concentrations of 3.5, 7.0 and 10.5% and equilibration time from 0.5 to 3 hr in experiment, produced sperm quality that is acceptable for further processing. The equilibration time from 30 min to 3 hr in experiment 2 gave goo d results on the quality of freeze-thawed rooster semen. The level of glycerol (7 are 10.5%) in experiment 3 gave better percentage in terms of quality of sperm compared to 3.5% of glycerol. Thawing at room temperature in experiment 4 gave the highest motility (46.39%) compared to the rest of the treatments. The fresh undiluted rooster semen in experiment 5, however, gave the highest motility and fertility.
