Cellulolytic potential of Penicillium citrinum FCBP-181 using different carbon sources

Cellulases are industrially important enzymes obtained from various microorganisms like bacteria and fungi. So, the present study was designed for appraisal of the most suitable synthetic substrate for enzyme production from indigenous mycoflora. The indigenous fungus Penicillium citrinum FCBP-181 was selected and analyzed for the evaluation of its cellulolytic enzyme activity by shake flask technique. The biosynthesis of cellulose was induced by addition of different carbon sources such as carboxymethylcellulose (CMC), microcrystalline cellulose (MCC) and soluble cellulase at 30 plus minus 2 degree C up to 96 hours. It was found that CMC induced the enzyme activity in substantial amounts (41.35 U/ml), whereas the enzymatic activity was found to be introverted by using MCC and soluble cellulase i.e., 39.86 U/ml by MCC and 35 U/ml by soluble cellulase. However, 72 h of incubation period was considered to be the best to achieve maximum enzyme ac tivity. The enzymatic activity of P. citrinum FCBP-181 was further evaluated at three pH levels viz., 4, 5 and 6 in submerged culture medium. pH 4 illustrated the best results for optimum enzyme activity by the test species.
