Identification of meloidogyne species collected from different parts of Turkey and determination of virulence of some root-knot (Meloidogyne spp.) populations | Türkiye'nin farklı bölgelerinden alınan kök-ur nematodu türlerinin (Meloidogyne spp.) tanısı ve bazı kök-ur nematodu populasyonlarının virülentliğinin belirlenmesi

Inglés. Root-knot nematodes (Meloidogyne spp.) cause significant quality and yield losses in crops throughout the world. The objective of this study was to identify the root-knot nematode samples, by molecular and morphological means, that have been collected from different regions of Turkey. The seventy-nine root-knot nematode samples were first homogenized by inoculating susceptible tomato cultivar Simita F1 with single egg masse.Then, second stage larvae obtained from homogenized-egg masses were used for DNA isolation. The molecular identification of the samples were conducted with speciesspecific SCAR (sequence characterized amplified region) and PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) markers.

Turco. Kök-ur nematodu türleri (Meloidogyne spp.) tüm dünyada bitkisel üretimde önemli verim ve kalite kayıplarına sebep olmaktadır. Bu çalışmada Türkiye Kök-ur nematodu türlerinin saptanması amacıyla, farklı alanlardan 79 adet Kök-ur nematodu populasyonu toplanarak moleküler ve morfolojik yöntemlerle teşhis çalışması yapılmıştır. Elde edilen Kök-ur nematodu populasyonları bir yumurta paketi ile hassas domates Simita F1 çeşidi üzerinde saflaştırılmıştır. Saflaştırılan yumurta paketlerinden elde edilen ikinci larva dönemindeki bireylerinden DNA izolasyonu yapılmış, türe spesifik primerlerle PCR kurularak Meloidogyne türleri teşhis edilmiştir. Aynı zamanda her populasyona ait ikinci dönem larvaların ve hassas domates bitkilerinin köklerinden elde edilen dişi bireylerin vulva kesitlerinden daimi preparatlar yapılarak morfolojik yöntemlerle de teşhis çalışmaları yapılmıstır.
