The detection of corynebacterium pseudotuberculosis antibody in goats by the enzymelinked immunosorbent assay (ELISA)

Caseous lymphadenitis, a worldwide disease of goats and sheep, is caused by Corynebacterium pseudotuberculosis (C. pseudotuberculosis). Several laboratory tests are available for the diagnosis of the infection, but none are completely satisfactory in terms of simplicity, accuracy, sensitivity and economy. An enzyme-linked immunosorbent assay (ELISA) is described for the detection of Corynebacterium pseudotuberculosis infection. Cell wall antigen was used to coat a solid phase and alkaline phosphatase was used to conjugate antigoat immunoglobulin. The technique was evaluated by: 1) monitoring the antibody level and abscess development in goats experimentally infected with C. pseudotuberculosis, 2) correlating abscess formation in naturally infected goats and 3) comparing the ELISA with the synergistic hemolysis inhibition (SHI) test. A significant ELISA antibody rise was evident two weeks post infection (pi) and peaked at five weeks pi, then declined slightly, but persisted. Abscesses were seen 8-10 weeks pi. All goats which had abscess were positive to ELISA. The ELISA is slightly more sensitive than SHI. The cell wall antigen is apparently specific, since the ELISA did not cross-react with sera from goats experimentally infected with Corynebacterium pyogenes
