The binary vector system with the cassette for the expression in plants of the antisense RNA of tobacco phospholipase C gene was constructed. The transgenic plants of potato (Solanum tuberosum) of Belarussian variety Vetraz were obtained. The plants created are meant for the investigation of the role of hospholipase C in the mechanisms of adaptation of potato plants to environmental conditions. On the basis of the vector and the agrobacterial strain pBI121deltaGUS GV2260 there was received a vector system for agrobacterial transformation of plants carrying antisense constructs for inhibiting gene phospholipase C. There were selected lines of transgenic potato plants using a selectable marker: for the resistance to kanamycin and by RNA-PCR with primers for the neomycin phosphotransferase gene. Presence of the target gene for the synthesis of antisense RNA gene of phospholipase C tobacco was confirmed by DNA PCR. These lines of potato plants will be used for reception on their basis of transgenic lines with a reduced content of phospholipase C, which can be used as models for studying mechanisms of plant adaptation to environmental conditions and mechanisms of RNA interference.
