Callus induction and establishment of suspension culture from seed derived cotyledon of cabbage cv. 864 Green Storm

Callus of cabbage (Brassica oleracea var. capitata L.) were initiated from cotyledon explants derived from seeds. Cotyledon explants cultured on medium containing 8 and 10 mgL-¹ 2,4-D resulted in high intensity of callus formation with 75-100% of the explant surface covered with callus. The callus produced in these treatments was friable, whitish and translucent. On medium without 2,4-D, no callus formation was observed. Upon transfer of the friable callus to liquid MS medium containing 1 mgL-¹ 2,4-D, rounded cells with dense cytoplasm (embryogenic) were obtained.
