IgG1 and IgG2 participation in anti-hapten, anti-carrier and immunoglobulin-containing cell (Ig-cc) responses was studied in sheep immunised with killed Staphylococcus aureus-Dinitrophenyl (DNP) conjugates. The antigen was given with and without the polyanionic adjuvant dextran sulphate (DXS). DXS markedly increased antibody responses to S. aureus-DNP in the popliteal lymph node, and strongly enhanced immunological memory. Any extrapolation of results from studies of immunomodulation by adjuvants should be undertaken cautiously, since the ratio of IgG2:IgG1 anti-DNP antibody in lymph draining lymph nodes stimulated with S. aureus-DNP alone did not differ significantly from the same ratio in lymph from animals given the antigen with DXS.
