PROTEÍNAS VIRALES COMO INDUCTORES DE LA RESPUESTA INMUNE EN PLANTAS Y SU IMPACTO EN LA COMUNIDAD MICROBIANA QUE HABITA EL RIZOBIOMA

"La agricultura desempeña un papel económico fundamental para los países, sin embargo, existen múltiples causas de pérdidas en los cultivos a nivel mundial que reducen significativamente las cosechas. En las últimas décadas, las enfermedades causadas por virus han ocasionado fuertes pérdidas económicas en la producción agrícola en México. Los begomovirus (familia Geminiviridae) son virus que afectan a gran parte de los cultivos de hortalizas. Una de las especies más relevantes es el Pepper Golden Mosaic Virus (PepGMV), que afecta principalmente a las plantas de chile. Adicionalmente, se ha reportado que la interacción con organismos patógenos puede afectar a la comunidad microbiana que habita la rizósfera de las plantas, sin embargo, la información sobre los cambios en el microbioma frente a enfermedades virales es escasa. En este estudio se analizó la expresión de los genes relacionados a la defensa de la planta modelo Nicotiana benthamiana frente a la infección con PepGMV y la inoculación de la proteína viral Rep, así como los cambios en la comunidad microbiana de la planta asociados a la presencia del virus. Para ello se realizó la agroinfiltración, de forma independiente, del virus PepGMV o de Rep en plantas de N. benthamiana y se recolectaron muestras de hoja, raíz y rizósfera a los días 1, 7 y 21 después de la inoculación. A partir de las muestras obtenidas se analizaron los cambios de expresión en genes involucrados en los mecanismos de defensa de la planta. Así mismo, se cuantificó la abundancia de comunidades microbianas de bacteria, archea y fungi, y se realizó el análisis de perfiles taxonómicos por NGS. Los resultados obtenidos demostraron que durante la infección viral aumenta la expresión de los genes DCL2 y DCL3. Por otro lado, Rep aumenta la expresión de los genes DCL1, DCL2 y DCL3, AGO1, y un gen relacionado a la señalización de SA. Durante la infección por PepGMV, las comunidades de bacterias presentes en la rizósfera aumentan durante los primeros 7 días después de la inoculación, mientras que en el compartimento endofítico disminuye la abundancia, destacándose en ambos resultados el aumento de la presencia del filo Firmicutes en el perfil taxonómico de las muestras bacterianas..."

The results suggest that in the presence of PepGMV, the AGO1 and DCL1 genes related to gene silencing in the plant are inhibited by PepGMV, while a viral component, such as Rep, induces the expression of modulated genes during viral infection. On the other hand, it is verified that N. benthamiana plants are capable of modifying the composition of their rhizobiome in the presence of a viral disease."
