Sequencing of the 16S rRNA gene V3-V4 region to determine the composition and relationships of the microbiota during inflammation of the udder and reproductive tract in cows (Bos taurus) | Секвенирование фрагментов V3-V4 гена 16S rRNA для определения состава и взаимосвязи микробиоты при воспалении молочной железы и репродуктивного тракта у коров (Bos taurus)

Inglés. Inflammatory diseases of the mammary gland and reproductive tract of cattle cause the greatest economic damage to dairy industry. The research goal was to determine the microbiota composition in the reproductive tract and mammary gland of cows with inflammation and compare the obtained bacterial profiles to detect the general etiology of such pathologies. Four experimental groups were formed from each farm in five districts of the Sverdlovsk Province, animals without signs of inflammation of the mammary gland and reproductive tract (NP, group 1); animals with inflammation of the mammary gland, but without inflammation of the reproductive tract (M, group 2); animals with inflammation of the reproductive tract, but without inflammation of the mammary gland (E, group 3); animals with inflammation of the mammary gland and reproductive tract (EM, group 4). Representatives of 19 bacterial phyla were identified in mammary gland secretion samples, including 43 classes, 85 orders, 165 families and 484 genera. Half of the bacterial ОТU was Firmicutes phylum (680 ОТU, 51.7%), the second and third largest phyla were Actinobacteria and Bacteroidetes (14.5% and 11.3%, respectively). Representatives of 22 bacterial phyla were found in cervical flushes, including 50 classes, 93 orders, 172 families and 365 genera. The predominant bacterial phylum was Firmicutes (876 ОТU, 55.3%), the second and third largest phyla were Bacteroidetes and Actinobacteria (13.3% and 11.6%, respectively). Sequencing of variable sections of the 16S rRNA gene allowed establishing the presence of bacteria of the Clostridia class and Facklamia genus, not detectable by cultural methods. This confirms clinical significance of the 16S rRNA gene sequencing method for clarifying etiological agents in the case of unculturable or difficult-to-culture bacteria.

Ruso. Воспалительные заболевания молочной железы и репродуктивного тракта крупного рогатого скота наносят наибольший экономический ущерб молочному животноводству. Нашей целью было определение состава микробиоты репродуктивного тракта и молочной железы коров при воспалении и сравнение полученных бактериальных профилей для обнаружения общей этиологии таких патологий. Для исследования на каждом из с.-х. предприятий, расположенных в 5 районах Свердловской области, сформировали 4 экспериментальных группы: 1-я - животные без признаков воспаления молочной железы и репродуктивного тракта (З); 2-я - животные с признаками воспаления молочной железы, но без признаков воспаления репродуктивного тракта (М); 3-я - животные с признаками воспаления репродуктивного тракта, но без признаков воспаления молочной железы (Э); 4-я - животные с признаками воспаления молочной железы и репродуктивного тракта (ЭМ). В образцах секрета молочной железы идентифицированы представители 19 бактериальных филумов, в том числе 43 класса, 85 порядков, 165 семейств и 484 рода. Половину бактериальных ОТU составлял филум Firmicutes (680 ОТU, 51,7%), второй и третий по численности филумы - Actinobacteria и Bacteroidetes (соответственно 14,5% и 11,3%). В цервикальных смывах обнаружены представители 22 бактериальных филумов, в их числе 50 классов, 93 порядка, 172 семейства и 365 родов. Преобладающим бактериальным филумом оказался Firmicutes (876 ОТU, 55,3%), вторым и третьим по численности - филумы Bacteroidetes и Actinobacteria (соответственно 13,3% и 11,6%). Секвенирование вариабельных участков гена 16S rRNA позволило установить наличие бактерий класса Clostridia и рода Facklamia, которые мы не обнаружили ранее, исследуя образцы с помощью культуральных методов.
