Aporte al desarrollo de un hidrogel con extractos de Artemisia absinthium con actividad antimicrobiana | Contribution to the development of a hydrogel with extracts of Artemisia absinthium with antimicrobial activity

Actualmente la industria farmacéutica realiza sus apuestas a productos farmacéuticos de aplicación tópica, que, en conjunto con la utilización de extractos vegetales, permiten obtener productos de gran impacto en el mercado farmacéutico. Por esto la utilización de vehículos como los hidrogeles y la obtención de extractos vegetales, debe ser de gran importancia para el desarrollo de nuevas investigaciones de carácter farmacéutico. Por ende, en este documento se quiere aportar al desarrollo de hidrogeles de colágeno tipo I cargados con extractos acuosos de Artemisia absinthium con actividad antimicrobiana, para ser posibles candidatos en el desarrollo farmacéutico de productos alternativos al tratamiento de heridas abiertas sobre la piel. Para lograr esto, en primer lugar se realizó todo el tratamiento desde la compra de la planta hasta lograr tener extractos acuosos de Artemisia absinthium, esto mediante la evaluación de diferentes condiciones de homogeneización por ultrasonido y el agua como único solvente; donde se determinó que las mejores condiciones de sonicación son un tiempo de exposición de 10 minutos, una amplitud de onda del 50%, 500 watts de potencia y 20 KHz de frecuencia porque fueron las condiciones que obtuvieron una mayor cuantificación de polifenoles en el ensayo realizado. Al mismo tiempo, se realizó la obtención de hidrogeles de colágeno de tipo I, donde el mejor prototipo obtenido fue aquel con una concentración del 1.5% porque posee propiedades reológicas, capacidad de esparcimiento y características físicas ideales para el desarrollo de esta investigación. Posterior a esto, se realizó la evaluación de la actividad antimicrobiana tanto de los hidrogeles, el extracto de Artemisia absinthium, donde se determinó que los hidrogeles estaban contaminados y por ende no ejercieron ningún tipo de inhibición microbiana; pero, el extracto acuoso de Artemisia absinthium a una concentración de 20.0 mg/mL obtuvo valores de inhibición microbiana del 86.54% para Staphylococcus aureus y del 72.5% para Pseudomonas aeruginosa. En conjunto con estos ensayos, se realizó la determinación de la viabilidad celular de los hidrogeles y del extracto de Artemisia absinthium utilizando células L929, donde los hidrogeles demostraron citotoxicidad y el extracto de Artemisia absinthium no generó citotoxicidad en el ensayo y de por sí, puede tener la capacidad de regeneración de tejido dérmico lo que causa valores de viabilidad celular por encima del 100% en el ensayo.

Currently the pharmaceutical industry makes its bets on pharmaceutical products of topical application, which together with the use of plant extracts, allow to obtain products of great impact in the pharmaceutical market. For this reason, the use of vehicles such as hydrogels and the obtaining of plant extracts must be of great importance for the development of new pharmaceutical research. Therefore, this document aims to contribute to the development of type I collagen hydrogels loaded with aqueous extracts of Artemisia absinthium with antimicrobial activity, to be possible candidates in the pharmaceutical development of alternative products to the treatment of open wounds on the skin. To achieve this, first all the treatment was carried out from the purchase of the plant to achieve aqueous extracts of Artemisia absinthium, this by evaluating different conditions of homogenization by ultrasound and water as the only solvent; where it was determined that the best sonication conditions are an exposure time of 10 minutes, a wave amplitude of 50%, 500 watts of power and 20 KHz of frequency because they were the conditions that obtained a greater quantification of polyphenols in the test performed. At the same time, type I collagen hydrogels were obtained, where the best prototype obtained was the one with a concentration of 1.5% because it has rheological properties, spreading capacity and ideal physical characteristics for the development of this research. After this, the evaluation of the antimicrobial activity of both the hydrogels, the extract of Artemisia absinthium, was carried out, where it was determined that the hydrogels were contaminated and therefore did not exert any type of microbial inhibition; but the aqueous extract of Artemisia absinthium at a concentration of 20.0 mg / mL obtained microbial inhibition values of 86.54% for Staphylococcus aureus and 72.5% for Pseudomonas aeruginosa. In conjunction with these tests, the determination of the cell viability of the hydrogels and the extract of Artemisia absinthium was carried out using L929 cells, where the hydrogels showed cytotoxicity and the extract of Artemisia absinthium did not generate cytotoxicity in the test and, may have the ability to regenerate dermal tissue, which causes cell viability values above 100% in the assay.
