Extracellular vesicles in the human maternal-embryo crosstalk during the preconception period

ANTECEDENTES: nuestro grupo demostró que el endometrio humano secreta exosomas (EXOs) al líquido endometrial (LE) con un contenido en miRNAs específico que puede ser internalizado por el embrión para favorecer la adhesión embrionaria. OBJETIVOS: definir y caracterizar los diferentes tipos de vesículas extracelulares (VEs) secretados por el endometrio humano al LE y estudiar su contenido en DNA, determinar si es internalizado por el embrión y si afecta a su regulación energética. MATERIALES Y MÉTODOS: El aislamiento de las VEs del LE se realizó mediante centrifugación diferencial. Se estudió su morfología, marcadores moleculares, tamaño y concentración a lo largo del ciclo menstrual. Su contenido en DNA se caracterizó mediante secuenciación de VEs en fase proliferativa (n = 4) y receptiva (n = 10). Las VEs se trataron con DNasa para eliminar el DNA externo. Se investigó la transferencia del DNA al embrión mediante marcaje con 5-etinil-2'-desoxiuridina y microscopía confocal. Finalmente, se estudio la capacidad de las VEs para modular el contenido en ATP de los embriones (n = 250) y su potencial de membrana mitocondrial (JC-1). RESULTADOS: se demostró la existencia de diferentes poblaciones de VEs en el LE humano, i.e: cuerpos apoptóticos (CAPs), microvesículas (MVs) y EXOs, con tamaños, morfología y marcadores moleculares específicos, y concentración invariable a lo largo del ciclo menstrual. Todas las VEs del LE en receptividad se encontraron enriquecidas en GSTT1, involucrado en detoxificación de especies reactivas del oxígeno. Además, las MVs mostraron enriquecimiento en DNA mitocondrial incluyendo sus 13 genes codificantes y sitios de unión de factores de transcripción (SUFT), para factores de transcripción (SRF, GABP, E2F4, TR4, FOXA2, FOXA1, CTCF, GATA2, PAX5) implicados en biogénesis mitocondrial, síntesis de ATP, protección contra el estrés oxidativo y desarrollo de linajes celulares embrionarios. Se observó que las VEs, especialmente CAPs y MVs, eran capaces de transferir DNA al trofoblasto embrionario, mientras que no se observó transferencia en la fracción libre de vesículas del LE. Finalmente, el estímulo de embriones con EXOs redujo sus niveles de ATP, sin afectar al potencial de membrana mitocondrial, pero reduciendo la concentración total de JC-1. CONCLUSIONES: el LE humano contiene diferentes poblaciones de VEs con fenotipo específico y enriquecimiento en secuencias génicas y SUFT en función del momento del ciclo y del tipo de vesícula, destacando las MVs por su contenido en DNA mitocondrial. Dichas secuencias podrían tener funciones en el control de la fisiología mitocondrial y protección frente al estrés oxidativo; y la transmisión de DNA en VEs fue demostrada en el trofectodermo. Los EXOs disminuyen los niveles de ATP embrionario, posiblemente a través de la reducción de la masa mitocondrial. Por ello, nuestros resultados sugieren un papel cooperativo de las VEs secretadas al LE humano como moduladores de la energética embrionaria.

BACKGROUND: our team demonstrated that human endometrium secretes exosomes (EXOs) to the endometrial fluid (EF) with a specific miRNA cargo that can be internalized by the embryo to promote embryo adhesion. OBJECTIVE: to define and characterize the different types of extracellular vesicles (EVs) secreted by the human endometrium to the EF and to study their DNA cargo, determine whether this cargo is internalized by the embryo and whether it modulates embryo energetic regulation. MATERIALS AND METHODS: Isolation of EF EVs was carried out by differential centrifugation. EF EVs morphology, molecular markers, size and concentration throughout the menstrual cycle was studied. Their DNA cargo was characterized by sequencing of EVs in proliferative stage (n = 4) and receptive stage (n = 10) of the cycle. EVs were treated with DNase to remove externally adhered DNA. The transfer of DNA to the embryo was investigated by staining with 5’-ethynyl-2’-deoxyuridine and confocal microscopy. Finally, the ability of EVs to modulate embryo ATP content (n = 250) and mitochondrial membrane potential (JC-1) was studied. RESULTS: the existence of different EVs populations in human EF was demonstrated, i.e: apoptotic bodies (ABs), microvesicles (MVs) and EXOs, all of them associated to specific sizes, morphologies, molecular markers and to an invariant concentration throughout the menstrual cycle. All EVs from receptive EF were found enriched in GSTT1, a gene involved in detoxification of reactive oxygen species. Furthermore, MVs showed enrichment in mitochondrial DNA, including its 13 coding genes and transcription factor binding sites (TFBS), whose associated transcription factors (SRF, GABP, E2F4, TR4, FOXA2, FOXA1, CTCF, GATA2, PAX5) have been involved in mitochondrial biogenesis, ATP synthesis, protection against oxidative stress and development of embryonic cellular linages. EVs, specially ABs and MVs, were observed to transfer DNA to the embryo trophoblast, while no transfer was observed for the EF EVs-free fraction. Finally, the co-incubation of embryos with EXOs reduced their ATP levels without affecting their mitochondrial membrane potential but reducing total JC-1 concentration. CONCLUSIONS: human EF contains different populations of EVs with specific phenotype and enrichment in genic sequences and TFBSs depending on the moment of the menstrual cycle and on the type of vesicle, highlighting MVs due to their content in mitochondrial DNA. Those sequences may have roles in mitochondrial physiology modulation and protection against oxidative stress. Furthermore, transmission of DNA in EVs to the embryo trophectoderm was demonstrated. EXOs reduce embryo ATP levels, likely through the reduction of total mitochondrial mass. In this context, our results suggest a cooperative role of EVs secreted to the human EF as modulators of embryo energetics.
