Bioprotection and genetic resistance against verticillium wilt of olive and pistachio | Bioprotección y resistencia genética frente a la verticilosis del olivo y pistachero

Verticillium wilt, caused by the soil-borne fungus Verticillium dahliae Kleb., is an important vascular disease affecting a wide range of woody and herbaceous hosts. Olive (Olea europaea L.) and pistachio (Pistacia vera L.) crops are among the most important economically crops seriously affected by the disease worldwide. The persistence of the fungus in the soil, the wide range of hosts and the lack of effective control methods make Verticillium wilt difficult to control. Therefore, searching for new sustainable disease control strategies are needed within an integrated management approach. To date, two of the most environmentally friendly measures available are the use of bioprotection through plant extracts and genetic resistance through resistant cultivars or rootstocks. In this context, since 2011, the Agroforestry Pathology Research Group AGR-216 of the University of Cordoba (Spain) has been developing a broad line of research on the biological control of Verticillium wilt of olive, in which more than 280 compounds have been evaluated under controlled conditions, including microorganisms (fungi, bacteria), organic amendments (animal and food industry waste), natural substances (essential oils, plant extracts), nutrients and resistance inducers. Likewise, the Genetic Resources and Olive Breeding Research Group (UCOLIVO) AGR-157 has developed an olive breeding program with emphasis in resistance against Verticillium wilt and, more recently, a pistachio breeding program, with the evaluation of more than 25,000 olive genotypes and 5,000 pistachio genotypes to date. Thus, this PhD Thesis has been developed in collaboration with both groups, an aspect that will help to lay the bases for future studies on the combined application of control measures based on bioprotection and genetic resistance against Verticillium wilt in olive and pistachio trees. The specific objectives of this PhD Thesis are: i) to evaluate commercial plant extracts against Verticillium wilt of olive trees; ii) to evaluate laboratory-made plant extracts against VWO; iii) to determine the mode of action of the most effective laboratory-made plant extracts; iv) to develop a method to characterize olive cultivars and genotypes based on their genetic resistance to the disease under controlled conditions; and v) to evaluate pistachio trees grafted on different Pistacia rootstocks against Verticillium wilt under field conditions. There is two complementary objectives that will be described later. The first and second objectives of this PhD Thesis were to evaluate in vitro the effect of seven natural-based commercial products; and nine laboratory-made extracts, against the viability of V. dahliae reproductive structures, and the disease progression in olive plants of cv. Picual. Willow and seaweed extracts were the most effective in reducing the disease in plants inoculated with V. dahliae when applied by irrigation compared to the remaining commercial products evaluated. The chemicals aluminium lignosulfonate 1 and copper phosphite, included for comparison, were also effective in reducing the disease. Pomegranate peel, carob pod or carob leaf extracts were obtained using three solvents: ethanol, methanol and ethyl acetate. For this purpose, 120 g of ground dried plant material was shaken for 48 h with 1 l of solvent. The mixture was filtered and evaporated using a rotavapor. An analysis of its secondary metabolite profile was performed using QTRAP ultra-high performance liquid chromatography with mass spectrometry. Pomegranate peel and carob leaf extracts obtained with methanol and ethanol were the most effective in reducing the disease in plants when applied by irrigation. High concentrations of gallic acid were found in all extracts. The aim of this study was to select the best laboratory-made extracts (objective 2) and thus determine their possible modes of action. Objective 3 aimed to determine the mode of action of pomegranate and carob leaf extracts obtained with methanol and whether they act by improving the defence system of olive plants of cv. Picual against V. dahliae through relative gene expression. For this purpose, the relative expression of marker genes related to resistance induction was quantified by RT-qPCR in stems and roots of noninoculated and untreated olive plants (water; negative control), noninoculated and treated plants with pomegranate or carob extracts, inoculated and untreated plants (water; positive control), and inoculated and treated plants with pomegranate and carob extracts. High relative expression values of the salicylic acid (SA)-marker gene were found at 1.5 and 3 months after the third treatment in olive plants inoculated and treated with pomegranate extract relative to inoculated and treated with the same extract after the third treatment. The results suggest that carob and pomegranate extracts do not activate the jasmonic acid (JA) pathway, as no increase in the expression of the marker gene was observed. Both extracts seem to be involved in the expression of abscisic acid (ABA) and polyphenol oxidase (PPO). Objective 4 was carried out to develop an inoculation method under controlled conditions that would allow the characterization of olive cultivars and genotypes for their resistance to Verticillium wilt, highly correlated to the response observed under field conditions. To this end, an artificial substrate based on sterile sand, corn flour and water, colonized by V. dahliae, was used to inoculate olive plants of the cvs. Picual, Arbequina, Koroneiki and Frantoio using two doses of substrate (20 or 80% of final inoculum concentration). The results showed that inoculations carried out with the artificial substrate at a dose of 20% allowed the differentiation of close susceptibility levels (‘Picual’ vs. ‘Arbequina’), while those carried out at a dose of 80% allowed the differentiation of close resistance levels (‘Koroneiki’ vs. ‘Frantoio’). The objective 5 was to evaluate pistachio trees grafted on five commonly used rootstocks: Pistacia terebinthus, P. atlantica, P. integerrima, and the interspecific hybrids ‘Platinum’ and ‘UCBI’, in an experimental field with a naturally infested soil with V. dahliae. The plants grafted on P. atlantica were more resistant than those grafted on P. integerrima, highlighting the intraspecific variability in resistance to the disease. In parallel, the disease was also monitored in an already established commercial field with pistachio trees grafted on P. terebinthus or ‘UCBI’. This monitoring assay served to validate the findings observed in the experimental field. In this sense, P. terebinthus proved to be susceptible to V. dahliae in the presence of inoculum of the pathogen in the soil, regardless of its concentration, while the hybrid ‘UCBI’ can be considered resistant to Verticillium wilt even in the presence of high densities of the pathogen. The objectives 6 and 7 were conducted as complementary tasks within this PhD Thesis. The objective 6 was carried out to determine the effect of the most effective laboratory-made extracts (pomegranate peel and carob leaf extracts obtained with methanol) previously tested in the objective 2, against olive anthracnose caused by Colletotrichum spp., one of the major aerial diseases of this crop. Copper sulphate was included for comparative purposes. Carob leaf extract was the most effective one in reducing the disease incidence progression in olive plants of cv. Arbequina. In order to elucidate its mode of action, H2O2 and phenolic compounds production were quantified by high-performance liquid chromatography with photodiode array detection (DAD-HPLC) in olive leaf samples. Both extracts were able to interfere with the plant defence response, as an increase in the production of H2O2, phenolic compounds and flavonoids was detected in plants treated and inoculated with the pathogen. Finally, the objective 7 was developed as complementary work to determine the existence of cross-pathogenicity by assessing the severity of different V. dahliae isolates of the defoliating ‘D’ and non-defoliating ‘ND’ pathotypes, from olive or pistachio trees, on woody (olive and pistachio) and herbaceous (cotton and tomato) hosts. D pathotype V. dahliae isolate was more aggressive than ND pathotype on olive and cotton plants, while both pathotypes showed similar aggressiveness on P. terebinthus. The ‘UCBI’ hybrid was highly resistant to all isolates regardless the pathotype. The knowledge generated in this PhD Thesis represents a relevant step in the bioprotection of Verticillium wilt in olive trees, opening new avenues to explore in order to better understand the mechanism of action of plant extracts against V. dahliae in this crop. In addition, it contributes to the knowledge of the genetic resistance to be exploited in olive and pistachio against Verticillium wilt. This PhD Thesis helps to lay the groundwork for future work combining both control methods in order to further develop more environmentally sustainable integrated disease management.

La verticilosis, causada por el hongo de suelo Verticillium dahliae Kleb., es una enfermedad vascular de gran importancia que afecta a una amplia gama de huéspedes leñosos y herbáceos. El olivo (Olea europaea L.) y pistachero (Pistacia vera L.) se encuentran entre los cultivos económicamente más importantes gravemente afectados por la enfermedad en todo el mundo. La persistencia del hongo en el suelo, el amplio rango de hospedantes y la ausencia de métodos de control efectivos, hacen que la verticilosis sea difícil de controlar. Por lo tanto, se hace necesaria la búsqueda de nuevas estrategias sostenibles de control de la enfermedad dentro de un manejo integrado. Hasta el momento, dos de las medidas disponibles más respetuosas con el medio ambiente son el uso de la bioprotección mediante extractos de plantas y la resistencia genética mediante cultivares o portainjertos resistentes. En este contexto, desde 2011, el Grupo de Investigación Patología Agroforestal AGR-216 de la Universidad de Córdoba (España) viene desarrollando una amplia línea de investigación sobre el control biológico de la verticilosis del olivo, en la que se han evaluado en condiciones controladas más de 280 compuestos, entre microorganismos (hongos, bacterias), enmiendas orgánicas (residuos animales y de la industria alimentaria), sustancias naturales (aceites esenciales, extractos de plantas), nutrientes e inductores de resistencia. Así mismo, el Grupo de Investigación Recursos Genéticos y Mejora del Olivo (UCOLIVO) AGR-157 viene desarrollando un programa de mejora genética del olivo frente a la verticilosis y, más recientemente, un programa de mejora genética del pistachero, con la evaluación de más de 25.000 genotipos de olivo y 5.000 de pistachero hasta la fecha. Así, esta Tesis Doctoral se ha desarrollado en colaboración de ambos grupos, aspecto que contribuirá a sentar las bases de estudios futuros sobre la aplicación combinada de medidas de control basadas en la bioprotección y la resistencia genética frente a la verticilosis en olivo y pistachero. Los objetivos específicos de esta Tesis Doctoral son: i) evaluar extractos vegetales comerciales frente a la Verticilosis del olivo; ii) evaluar extractos vegetales obtenidos en laboratorio frente a VWO; iii) determinar el modo de acción de los extractos vegetales obtenidos en laboratorio más eficaces; iv) desarrollar un método para caracterizar cultivares y genotipos de olivo en función de su resistencia genética a la enfermedad en condiciones controladas; y v) evaluar pistacheros injertados sobre diferentes portainjertos de Pistacia frente a la verticilosis en condiciones de campo. Hay dos objetivos complementarios que se describirán más adelante. El primer y segundo objetivo de esta Tesis Doctoral fue la evaluación in vitro del efecto de siete productos comerciales de base natural; y nueve extractos elaborados en laboratorio, en la viabilidad de las estructuras reproductivas de V. dahliae, y en el progreso de la enfermedad en plantas de olivo cv. Picual. Los extractos de sauce y algas fueron los más eficaces en la reducción de la enfermedad en plantas inoculadas con V. dahliae cuando se aplicaron por riego, en comparación con el resto de los productos comerciales evaluados. Los productos químicos lignosulfonato de aluminio 1 y fosfito de cobre, incluidos con fines comparativos, también resultaron efectivos en la reducción de la enfermedad. Los extractos de cáscara de granada, vaina de algarrobo u hoja de algarrobo se obtuvieron utilizando tres disolventes: etanol, metanol y acetato de etilo. Para ello, 120 g de material vegetal seco molido se agitaron durante 48 h con 1 l de disolvente. La mezcla se filtró y se evaporó utilizando un rotavapor. Se realizó un análisis de su perfil de metabolitos secundarios mediante cromatografía líquida de ultra alta resolución QTRAP con espectrometría de masas. Los extractos de cáscara de granada y de hoja de algarrobo obtenidos con metanol y etanol fueron los más eficaces en la reducción de la enfermedad en planta cuando se aplicaron por riego. Se encontraron altas concentraciones de ácido gálico en todos los extractos. El objetivo de este estudio fue seleccionar los mejores extractos elaborados en laboratorio (objetivo 2) y determinar sus posibles modos de acción. El objetivo 3 pretendía determinar el modo de acción de los extractos de granada y hojas de algarrobo obtenidos con metanol y si actúan mejorando el sistema de defensa de las plantas de olivo cv. Picual frente a V. dahliae mediante expresión de genes relativa. Para ello, se cuantificó mediante RT-qPCR la expresión relativa de genes marcadores relacionados con la inducción de resistencia en tallos y raíces de plantas de olivo no inoculadas y sin tratar (agua; control negativo), plantas no inoculadas y tratadas con extractos de granada o algarroba, plantas inoculadas y sin tratar (agua; control positivo), y plantas inoculadas y tratadas con extractos de granada y algarroba. Se encontraron altos valores de expresión relativa del gen marcador del ácido salicílico (SA) a 1,5 y 3 meses después del tercer tratamiento en plantas de olivo inoculadas y tratadas con extracto de granada en relación con las inoculadas y tratadas con el mismo extracto después del tercer tratamiento. Los resultados sugieren que los extractos de algarroba y granada no activan la vía del ácido jasmónico (JA), ya que no se observó un aumento en la expresión del gen marcador. Ambos extractos parecen estar implicados en la expresión del ácido abscísico (ABA) y de la polifenol oxidasa (PPO). El objetivo 4 se llevó a cabo para desarrollar un método de inoculación en condiciones controladas que permita caracterizar cultivares y genotipos de olivo en función de su resistencia a la verticilosis de forma similar a la respuesta que se observa en condiciones de campo. Para ello, se preparó un sustrato artificial a base de arena, harina de maíz y agua estériles, colonizado por V. dahliae, para inocular plantas de olivo de los cvs. Picual, Arbequina, Koroneiki y Frantoio utilizando dos dosis de sustrato (20 u 80% de la concentración final de inóculo). Los resultados indicaron que las inoculaciones realizadas con el sustrato artificial a una dosis del 20% permitieron diferenciar niveles de susceptibilidad cercanos (‘Picual’ vs. ‘Arbequina’), mientras que las realizadas a una dosis del 80% permitieron diferenciar niveles de resistencia cercanos (‘Koroneiki’ vs. ‘Frantoio’). El objetivo 5 fue evaluar pistacheros injertados en cinco portainjertos comúnmente utilizados: Pistacia terebinthus, P. atlantica, P. integerrima y los híbridos interespecíficos ‘Platinum’ y ‘UCBI’, en un campo experimental con un suelo naturalmente infestado con V. dahliae. Las plantas injertadas sobre P. atlantica fueron más resistentes que las injertadas sobre P. integerrima, destacando la variabilidad intraespecífica en la resistencia a la enfermedad. Paralelamente, también se realizó un seguimiento de la enfermedad en un campo comercial ya establecido con pistacheros injertados sobre P. terebinthus o ‘UCBI’. Este ensayo de seguimiento sirvió para validar los hallazgos observados en el campo experimental. En este sentido, P. terebinthus se mostró susceptible a V. dahliae en presencia de inóculo del patógeno en el suelo, independientemente de su concentración, mientras que el híbrido ‘UCBI’ puede considerarse resistente a la verticilosis incluso en presencia de altas densidades del patógeno. Los objetivos 6 y 7 se llevaron a cabo como tareas complementarias dentro de esta Tesis Doctoral. El objetivo 6 se llevó a cabo para determinar el efecto de los extractos obtenidos en laboratorio más efectivos (extractos de piel de granada y hoja de algarrobo obtenidos con metanol) previamente testado en el objetivo 2, frente a la antracnosis del olivo causada por Colletotrichum spp., una de las principales enfermedades aéreas de este cultivo. El sulfato de sobre se incluyó con fines comparativos. El de hoja de algarrobo fue el extracto más eficaz para reducir la progresión de la incidencia de la enfermedad en plantas de olivo cv. Arbequina. Para dilucidar su modo de acción, se cuantificó la producción de H2O2 y compuestos fenólicos mediante cromatografía líquida de alta resolución con detección de matriz de fotodiodos (DAD-HPLC) en muestras de hojas de olivo. Ambos extractos fueron capaces de influir en la respuesta de defensa de la planta, ya que se detectó un aumento en la producción de H2O2, compuestos fenólicos y flavonoides en plantas tratadas e inoculadas con el patógeno. Por último, el objetivo 7 se desarrolló como trabajo complementario para determinar la existencia de patogenicidad cruzada mediante la evaluación de la severidad de diferentes aislados de V. dahliae de los patotipos defoliante ‘D’ y no defoliante ‘ND’, procedentes de olivo o pistachero, sobre huéspedes leñosos (olivo y pistachero) y hortícolas (algodón y tomate). El aislado del patotipo D de V. dahliae fue más agresivo que los del patotipo ND en plantas de olivo y algodón, mientras que ambos patotipos mostraron una agresividad similar en P. terebinthus. El híbrido ‘UCBI’ fue altamente resistente a todos los aislados, independientemente del patotipo. El conocimiento generado en esta Tesis Doctoral supone un paso relevante en la bioprotección de la verticilosis en olivo, abriendo nuevas vías a explorar para entender mejor el mecanismo de acción de los extractos vegetales frente a V. dahliae en este cultivo. Además, contribuye al conocimiento de la resistencia genética a explotar en olivo y pistachero frente a la verticilosis. Esta Tesis Doctoral contribuye a sentar las bases para futuros trabajos combinando ambos métodos de control con el fin de seguir desarrollando una gestión integrada de la enfermedad más sostenible medioambientalmente.
