Broccoli various genotypes regenerated plants (R0) seed set after geitonogamy

Relevance. Broccoli cabbage (Brassica oleracea L. convar. botrytis (L.) Alef. var. cymosa Duch.) is widely spread around the world due to its dietetic attribute, preventive and healing effect, cooking simplicity. F1 hybrids production is the main stream in modern crop breeding as they surpass varieties in evenness and yield quality. However, it takes up to 6 years of inbreeding to obtain pure parent lines capable for hybrid creation. Thus biotechnological methods, especially doubled haploids production technology shell be implemented in selection process. 100% homozygous line can be worked out in one generation by this technology. Nevertheless earlier researches revealed such plants low seed set that hinder their application in mass production. Therefore after pollination seed set level identification is an important step in applying lines gained via in vitro process.Materials and methods. Broccoli cabbage regenerated plants produced by isolated microspore in vitro culture method were used as raw material in our study. Flower and bud pollination was held in climate cells.Results. Study of 11 broccoli cabbage DH-lines disclosed genotype influence the ability to set seeds after bud pollination. 93.3% genotype A and 95.5% A1.3 pods were seedless in our experiments. Most genotypes demonstrated 50.3-85.7% seed set level. Low and middle ability to form seeds prevailed mainly. This peculiarity should be considered while dealing with regenerated plants by raising the number of crossings for successful breeding.

Relevance One of the basic directions of the cabbage crop breeding is the creation of F1 hybrids with a complex of economically valuable traits. This process is difficult and time-consuming as to get pure lines must be within 6-12 years hold inbreeding. Herewith not every line gives the desired heterotic effect that also requires additional verification. Methods Biotechnological method culture of isolated microspores in vitro, which allows in the first generation to receive a line with 100% homozygosity, was used to speed up the breeding process. Combination ability were performed in complete diallel cross on the basic morphological signs. Results Culture medium for cultivation of isolated microspores in vitro was optimized for each genotype of cabbage for the best embryoids regeneration. Maximum amount of embryoids was received on medium with pH 6.2 using ampicillin 100 mg/l and zeatin 1 mg/l: 466.7 ± 153.2 PCs/100 buds. A new source material for breeding – doubled haploid lines of cabbage was received. Lines – the best parents for F1 hybrids with high yield, compact rosette of leaves, with optimum inside and short outside cabbage stump was created. Studies have shown that optimization of breeding process in case of creation of pure lines of cabbage in 3 years with microspore culture requires to reduce the breeding process in 2 times.

The biotechnological methods enable to produce the ‘pure’ lines for the short period of time that intensify the breeding program. Particularly, the anther culture is widely used to obtain double haploid lines (DH lines) in plant breeding. The technology advantage is to fast up essentially the selection of homozygous lines with different morphological traits and high level of uniformity. Contemporary breeding programs are directed at F1 hybrid development; therefore DH lines are in need to serve as the parental form for heterotic hybrid production. Nevertheless, previous data has shown that DH plants have been noticed to produce few seeds on them. To introduce the lines into breeding program for hybrid production and to multiply them, the seed formation of DH lines is worth being studied. The result of assessment of seed generation of DH lines of Broccoli that have been produced from ‘Tonus’ and convariety ‘N1’ is presented in the article There is much difference in morphological traits and both ability to recognize their own pollen and quantity of seeds in the pod among plants produced. Nine DH lines of early maturity were obtained. The differences in seed formation among these lines and between generations were revealed. It was shown the influence of genotype on seed formation in DH lines. It was also noticed the much seed formation in case of geitonogamic pollination in DH lines obtained from cultivar ‘Tonus’. The biological features of flowering in DH lines under conditions of climatic chambers have been studied.

A model of late maturing F1 hybrid of white head cabbage for long-term storing and suitable for national recipe “golubci” have been developed based on requirements for varieties and F1 hybrids grown in the South in the conditions of drought, high temperatures and low air humidity. Combining ability of seven self-incompatible inbred lines of white head cabbage was studied by the system of full diallel crosses in conditions of Pridnestrovian Moldavian Republic. These lines were developed on the base of heat tolerant varieties ‘Biruchekutskaya’, ‘Volna’, ‘Lada’, ‘Moldavanka’ and ‘Kharkovskaya Zimnyaya’ bred in the south. Out of 42 hybrid combinations studied 15 significantly surpassed in productivity the variety ‘Zavadovskaya’ that is traditionally grown by local farmers for pickling and recipe “golubci”. High GCA effects for productivity showed the following breeding lines: Bu1, Ml3 and Kl5. Polygenes, controlling the high yield ability, mainly were dominant and single-directed, however the correlation betweenyield ability and GCA effect was middle, r=0.63±0.35. No correlation was observed inbreeding line between GCA effect and truly heterosis effect in hybrid combinations (r=0.19). Disease resistance analysis revealed lines Bu1 and Kl5 that had shown resistance to Fusarium yellows. Two hybrid combination Kl5xBu1 and Ml3xBu1 were highly tolerant to thrips attacks according to analysis of all promising hybrid combinations, carried out in natural infection background. Weak correlation was observed between the number of damaged leaves in cabbage head and dry matter content, r=0.41±0.21, and also there is no correlation revealed with sugar contents, ascorbic acid and nitrate content. High tolerance of hybrids with participation of line Bu1 explained the partial dominance nature of the trait. Hybrid combinations Kl5xBu1 and Ml3xBu1 called ‘Batal’ and ‘Shedevr’, respectively, included in registry of Moldova and PMR.

The scheme of development of two-line of hybrids F1 in European radish based on self-incompatibility includes five stages, as follows: 1 – selection of self-incompatible lines, common and specified combining ability estimation; 2 – inbreeding and selection to make the lines homozygous for morphological traits, common and specified combining ability estimation; maintenance and reproduction of self-incompatible lines; 4 – production of hybrid seeds. The research work on assessment of hybrid F1 that were obtained from cross of eight self-incompatible lines of European winter radish by the Griffing’s method was carried out in 2016. The assessment of length, diameter and yield of radish root was performed. According to the root shape the heterotic hybrids F1 were divided into three groups: rounded-flat, 48.2%; round, 50.0%; and flatten-round, 1.8%. The level of root marketability of hybrids F1 reached 100%. As a result of the work the promising hybrid combination distinguished by high uniformity, marketability and high yield were selected out.
